A study of 162 females, actively employed as medical practitioners, was conducted to examine perceived characteristics of their husbands and to determine whether such attributes might relate to the women's commitment to and satisfaction with medical practice. A questionnaire assessing personality characteristics of such husbands was produced by a factor analytic technique, and the final measure is tabled. Eighteen items loaded clearly on one of three factors measuring perceptions of general support by husbands, levels of type A characteristics within the husbands, and the degree to which husbands were against their wives' medical career. Factor-specific scores on those dimensions were examined against other variables, and associations were found linking scale scores with variables in the female practitioners including age, type of medical practice, hours of employment, and satisfaction with medical practice. Husbands who were medical graduates were scored higher on type A characteristics and fathered more children than the other husbands.

Mitotic activity of extra- and intra-pulmonary airway epithelium has been studied in male rats exposed to tobacco smoke for 1, 2, 3, 7 or 14 days, with and without addition of the anti-inflammatory agent phenylmethyloxadiazole (PMO) to the tobacco. In both control and exposed animals the mitotic index is greater in extrapulmonary regions than intrapulmonary ones. A single exposure to tobacco smoke significantly increases mitotic activity in both airway regions. This initial level of mitotic response is not maintained but is rapidly restored by 1 day free from tobacco exposure: the second peak is as high as the first. Exposure to tobacco + PMO modifies timing and amplitude of the mitotic response. The effect of PMO is somewhat paradoxical since the first peak occurs later, i.e. after 2 days of exposure, but the increase is almost twice that seen after tobacco alone. The timing of the second peak is the same as after tobacco alone, but its amplitude is only half. In each experimental group the mitogenic effect is exerted on an intact epithelium. In animals exposed to tobacco alone, or tobacco + PMO, in extrapulmonary airways mitoses are located mainly in the basal region of the epithelium, whereas in intrapulmonary airways they are located mainly at the mid or superficial level.

1. The distribution and properties of superoxide dismutase were examined in mammalian semen, and the enzyme was used to investigate the role of superoxides in metal-ion-catalysed lipid-peroxidation reactions in spermatozoa. 2. Superoxide dismutase activity was detected in seminal plasma and spermatozoa from all species studied, exceptionally high activity being found in donkey semen. The enzyme is easily solubilized from spermatozoa, as 85-90% of the total activity is released by cold shock, a relatively mild form of cellular damage. 3. Purification and characterization of the enzyme from supernatant fractions prepared from cold-shocked boar spermatozoa showed it to be cyanide-sensitive, to have a mol.wt. of 31 000, a pI of 5.9 and to contain 1.85 g-atoms of copper and 1.91 g-atoms of zinc per mol of protein. However, extensive sonication of spermatozoa released a small amount of a cyanide-insensitive enzyme, presumably a mangano superoxide dismutase, from the mitochondrial matrix. 4. The presence of superoxide dismutase in spermatozoa, either intracellularly or extracellularly, did not inhibit ascorbate/Fe2+-catalysed lipid-peroxidation reactions, suggesting that superoxides are not essential intermediates in this system.
EMD 28422 (N6-[2-(4-chlorophenyl)-bicyclo-2.2.2.octyl-(3)]-adenosine) was demonstrated to increase the number of binding sites for [3H]diazepam (Bmax) in vitro without an accompanying increase in receptor affinity (KD). The increase in receptor number was observed in both crude synaptosomal preparations (P2) and thrice-washed membrane preparations with and without the addition of 50 microM GABA. Furthermore, this effect appeared to be independent of the concentration of chloride ion, since the increases in Bmax were observed in both Tris-HCl and Tris-maleate buffer. The effects of EMD 28422 were stereospecifically antagonized by the GABA antagonist bicuculline, despite the lack of effect of EMD 28422 on [3H]muscimol binding at concentrations which markedly increased benzodiazepine receptor number. Neither EMD 39011 nor adenosine, the two parent moieties of EMD 28422, increased [3H]diazepam binding at concentrations of up to 1 mM. The increases in benzodiazepine receptor number observed with EMD 28422 in vitro suggests that this compound induces a conformational change in the benzodiazepine receptor which may cause the dissociation of an endogenous noncompetitive inhibitor of [3H]diazepam binding from the membrane, thus 'unmasking' binding sites. The stereospecific antagonism of this effect by bicuculline and the apparent inability of GABA to alter the action of EMD 28422 suggests the presence of a novel type or different functional state of GABA receptor which may play a permissive role in the rapid modulation of benzodiazepine receptor number in vitro.
This article describes three cases of lead poisoning and 20 cases of excessive lead absorption in a factory producing inorganic lead compounds. Evidence is presented of a general system failure in that environmental surveys, application of legislation and acceptance of advice by management were ineffective.
1. The regulation by testosterone of mRNA complexity and mRNA activity was investigated in rat caput and cauda epididymidis. 2. The sequence complexity of cytoplasmic poly(A)-containing RNA from normal rats was determined by homologous hybridization with radiolabelled complementary DNA probes by using RNA in excess. Computer analysis of results suggested that hybridization could best be described by curves composed of two components distinguished by their relative abundance. Thus caput-epididymidal RNA consists of approx. 260 moderately abundant and 16400 scarce sequences, whereas cauda-epididymidal RNA consists of approx. 124 moderately abundant and 13400 scarce sequences. Judging by heterologous-hybridization reactions, castration did not result in appreciable alterations in either sequence complexity or the relative abundance of the two classes of poly(A)-containing RNA. 3. To investigate if individual mRNA sequences were regulated by androgens, mRNA was translated in a cell-free system derived from reticulocyte lysate. Since most of the translation products had a different mobility on sodium dodecyl sulphate/polyacrylamide gels from the authentic proteins synthesized in tissue minces, antibodies were used to identify specific translation products. Antibodies to the two related major proteins (mol.wt. 18500 and 19000) secreted by the caput epididymidis and whose synthesis is stimulated by testosterone both precipitated a single translation product of mol.wt. 21000. That this polypeptide was a precursor to the secreted proteins was suggested by the fact that the addition of microsomal membranes isolated from dog pancreas resulted in the appearance of a polypeptide of mol. wt. 19000. 4. Translation of RNA from the caput epididymidis of rats of different hormonal status showed that mRNA activity for the 21000-dalton polypeptide declined after castration, but could be restored by treating rats with testosterone. 5. It is concluded that testosterone stimulates the synthesis of a major protein secreted by the caput epididymidis by regulating its mRNA activity.
The activity of phosphoenolpyruvate carboxylase (orthophosphate:oxaloacetate carboxy-lyase (phosporylating) EC 4.1.1.31) purified from Bryophyllum fedtschenkoi has been measured in the presence of various concentrations of phosphoenolpyruvate, L-malate and glucose 6-phosphate. At high pH, the enzyme is competitively inhibited by L-malate and activated by glucose 6-phosphate. A reaction scheme describing the interaction of enzyme, substrate and effectors is proposed. Values for the appropriate equilibrium constants have been calculated for the enzyme acting at pH 7.8, which is one of its two pH optima. The kinetics are more complicated at low pH, partly because of non-linear reaction rates and partly because inhibition by L-malate is not competitive. Activation by glucose 6-phosphate is similar at high and low pH values. The behaviour of a wide range of other possible effectors is described briefly.
1. Protein synthesis has been investigated in different regions of the rat epididymis by measuring incorporation of [35S]methionine in tissue minces incubated in vitro followed by analysis of labelled proteins on polyacrylamide gels containing sodium dodecyl sulphate. Rates of synthesis were highest in the proximal cauda > distal cauda > initial segment > ductuli efferentes > corpus > distal caput > proximal caput. One protein (mol.wt. 23 000) characterized the initial segment, three proteins (mol.wts. 18 500, 19 000 and 32 000) the caput and one protein (mol.wt. 47 000) the cauda. 2. After castration, [35S]methionine incorporation in all regions of the epididymis was reduced to < 10% of that in normal animals but could be restored to control levels within 5 days by testosterone treatment. Other steroids (corticosterone, oestrogen or progesterone) were ineffective. 3. The synthesis of the 18 500, 19 000, and 32 000 mol.wt. proteins in the caput and the 47 000 mol.wt. protein in the cauda were preferentially regulated by androgens, whilst the synthesis of 23 000 and approx. 80 000 mol.wt. proteins in the initial segment was dependent upon factors present in testicular fluid. 4. The androgen-dependent and testicular fluid-dependent proteins were major components of epididymal secretion. Purification and characterization of the 18 500, 19 000, 23 000 and 32 000 mol.wt. proteins showed them to be acidic glycoproteins with a carbohydrate content of 7.6-13.2%. The 47 000 mol.wt. protein, on the other hand, is highly basic. 5. A possible role for these proteins in the acquisition of motility, fertilizing capacity and storage of spermatozoa in the epididymis is discussed.
