Effect of ACTH on the composition of the aqueous humour of the rabbit eye.

Tetracosactide (Synacthen, Ciba) increases the concentration of corticosterone in the aqueous humour of the rabbit eye. The maximal increase, about 5-fold of the basal level, occurs with a slight delay of approximately 30 min after the maximally stimulated corticosterone level in blood plasma. Glucocorticoid elevation is paralleled by an increase of protein content and a slight rise of glucose in aqueous. A disruption of the blood-aqueous barrier by ACTH similar to the action of alpha-melanocyte-stimulating hormone is suggested.

A selective radioimmunoassay of androstenedione in plasma and saliva.

A specific radioimmunoassay for 4-androstene-3,17-dione (androstenedione) based on rabbit antisera to 6 beta-hydroxy-4-androstene-3,17-dione-6-hemisuccinate-bovine serum albumin conjugate was developed. Cross reaction by testosterone was eliminated by addition of experimentally established amounts of testosterone antiserum, which cross reacted by only 0.004 with androstenedione. The method has been used for simultaneous determination of androstenedione in both plasma and saliva of normal women under basal conditions and following dynamic tests. The plasma levels in normal women averaged 3.6 +/- 0.9 nmol/l and correlated (r = 0.93) with those found in saliva (0.353 +/- 0.089 nmol/l).

Androgen receptor binding and antiandrogenic activity of some 4,5-secoandrostanes and ring B cyclopropanoandrostanes.

Eight androstane derivatives with modified ring A or B (4,5-secoandrostanes and ring B cyclopropanoandrostanes) were assayed in vivo on mice for their antiandrogenic activity and the effect was compared with that of cyproterone acetate. The inhibition of dihydrotestosterone binding to rat prostate cytosol and to human plasmatic sex hormone binding protein was correlated with the in vivo effect. The antiandrogenic activity of 6 alpha,7 alpha-cyclopropano-5 alpha-androstane-3 beta,17 beta-diol was nearly as high as that of cyproterone acetate. The opening of ring A of androgens, such as testosterone or 17 alpha-methyltestosterone, moderately reduced the binding and changed the biological activity to weakly antiandrogenic.

Aqueous humour and plasma cortisol levels in glaucoma and cataract patients.

By means of radioimmunoassay, the content of endogenous cortisol in the aqueous humour and plasma of 35 patients suffering from various types of glaucomas and 35 cataract patients was determined, and the ratio of the plasma to the aqueous humour cortisol levels was calculated. The highest cortisol level in both plasma and aqueous humour was found to occur in patients with open-angle glaucoma suffering at the same time from systemic hypertension. The lowest plasma to aqueous humour cortisol ratio was found in patients with secondary glaucoma treated with steroids. In woman suffering from open-angle glaucoma, the rising cortisol level in plasma with age contrasted to the decreasing cortisol level in aqueous humour. The authors suggest that there is an active interference with homeostatic mechanisms responsible for ensuring the stability of the eye inner milieu and a certain protection of the trabecular meshwork of the angle of the anterior eye chamber against noxious effects of the endogenous cortisol.

Free dehydroepiandrosterone and testosterone in human amniotic fluid and prediction of gonadal sex.

In order to correlate the gonadal production of male sex hormones with overall steroidogenic activity of the fetus, unconjugated dehydroepiandrosterone (DHEA) and testosterone were measured in amniotic fluids from mid-pregnancies with male and female fetuses by highly specific radioimmunoassay. The antiserum was raised in rabbits with 3 beta-hydroxy-17-oxo-5-androsten- 19-al 19-carboxymethyloxime BSA as immunogen and its cross-reactions with several steroids were checked. Significantly higher testosterone level was found in amniotic fluid from male fetuses (average concentration 249 +/- 109 pg ml-1 from males as compared with 70 +/- pg ml-1 from females), whereas the difference in DHEA concentration was significant (184 +/- 96 pg mol-1 for males vs. 177 +/- 92 pg ml-1 for females). The positive correlation between testosterone and DHEA concentrations (r = 0.53) was found only in amniotic fluids from pregnancies bearing male fetuses indicating that in males more DHEA may originate from fetal gonads.
