Binding of [3H]-4,5-secodihydrotestosterone by androgen receptor.

Specific binding of [5,6-3H2]-17 beta-hydroxy-4,5-secoandrostan-3-one (or [3H]-4,5-secodihydrotestosterone, IX) to rat prostate cytosol receptors was observed and confirmed by two independent methods: by adsorption to dextran-charcoal and by polyacrylamide gel electrophoresis. The electrophoresis of [3H]-4,5-secodihydrotestosterone--cytosol mixtures showed a single radio-activity peak at different radioligand concentrations, displaceable by nonradioactive 4,5-secodihydrotestosterone; the mobility of the peak corresponds to a protein with a relative molecular weight of 90 000-130 000. The apparent association constant of the compound with respect to adsorption to dextran-charcoal is 1.48 X 10(9) l X mol-1, similar to the value observed with dihydrotestosterone (4.8 X 10(8) l X mol-1).

Changes induced in rabbit plasma and aqueous humour by ACTH4-10 and ACTH1-24.

In the rabbit the effect of a single i.m. dose of ACTH4-10 (130 micrograms kg-1) was compared with that of ACTH1-24 (20 micrograms kg-1). In contrast to ACTH1-24 the heptapeptide ACTH4-10 did not stimulate the adrenal secretion of corticosterone. Both ACTH fragments induced a slight hyperglycemia and in the aqueous humour they increased the concentration of proteins.

Estrogen receptors in human goitrous and neoplastic thyroid.

An attempt was made to quantify the number of specific estrogen binding sites in specimens of human goitrous or neoplastic thyroid tissue obtained by surgery. Using synthetic estrogen [3H]moxestrol as a radioligand, the concentrations of estrogen receptors found were very low (average 0.57 +/- 0.47 fmol mg-1 of protein) approaching the detection limit. Accordingly, no significant sex dependent differences in estrogen receptors could be revealed.

Effect of glucocorticoids on the composition of the aqueous humour of the rabbit eye.

Exogenous corticosteroids (dexamethasone, cortisol) inhibit the adrenal secretion of corticosterone in the rabbit and decrease the concentration of corticosterone in aqueous humour, increase glycaemia and cause a rise in glucose concentration in the aqueous. Except for a slight rise 16 h after the corticosteroid administration no effect on plasma or aqueous content of total protein was observed. This is in striking contrast with the increase of proteins in aqueous humour after ACTH injection. Thus it seems likely that corticosteroids do not mediate the effect of ACTH on the disruption of the blood-aqueous barrier.

Content and binding of aldosterone in human ocular lens.

The concentration of aldosterone was determined by radioimmunoassay in homogenates from a total of 227 human cataractous lenses divided into six groups according to age and accompanying diseases (i.e. glaucoma, essential hypertension, diabetes and combination of the two latter diseases). Aldosterone concentration in cataracts ranged from 0.020 to 0.036 fmol mg-1 protein. Significantly higher values were found in lenses of patients with hypertension (0.047 +/- 0.022 fmol mg-1 protein). Using two independent methods an evidence was brought that specific and saturable aldosterone binding occurs in cytosol of human lens epithelium obtained from autopsy specimens, which is not identical with glucocorticoid binding. Electrophoretic properties of this aldosterone binding entity correspond to a protein with a relative molecular mass of 90 000 daltons.
