Antithrombin III levels in term and preterm infants measured by rate nephelometry.

The plasma antithrombin III level was measured in term and premature newborns within the first 24 h of life; in addition to the authors', own immunological method utilizing rate nephelometry the assays of Laurell and Kabi Coatest were used. Antithrombin III concentration showed a linear correlation with the degree of maturity, in accordance with previous data. The lowest values were found in premature babies affected by idiopathic respiratory distress syndrome who later developed pulmonary haemorrhage. At all gestational ages the antithrombin III functional index was lower than 1.0, indicating antithrombin III consumption. Since both concentration and activity of antithrombin III are low in prematures, they are at an increased risk for coagulopathies, hence substitution therapy is indicated. Measurement of antithrombin III by rate nephelometry can also be used for following changes in inhibitor concentration.

Segmental mobility in glycogen phosphorylase b.

The dynamics and structuredness of the pyridoxal 5'-phosphate-binding region in glycogen phosphorylase b (EC 2.4.1.1) has been investigated with different techniques of fluorescence spectroscopy. Fluorescence polarization data of the thermal Perrin plot indicate some mobility in the cofactor binding site, while the isothermic measurements (at 20 degrees C, in high-viscosity solvents) demonstrate that the mobile unit carrying the emission oscillator is practically insensitive to the external viscosity. Characteristics of the thermal Perrin plots obtained for both native and reduced phosphorylase b can be interpreted either as a freely moving cofactor in a medium of high viscosity (0.3 P) or as the motion of a unit larger than a lysine-bonded pyridoxal 5'-phosphate in a medium with the viscosity of water. Data for acrylamide quenching and time-resolved fluorescence measurements suggest that the latter interpretation should valid. These data also suggest a tightly packed microenvironment around the pyridoxal moiety.

Contribution to the methodology of optimization and in-process control of some physical properties of pharmaceutical bulk substances and granulates.

Many experts consider drug analysis exclusively as a chemical measuring technique. In the pharmaceutical industry, physical and physico-chemical parameters form an integrated part of the quality specification, because they play an essential role both in quality assurance and in the technical and economic aspects of production on the commercial scale. Such physical and physico-chemical properties are analysed by special, non-chemical methods. In their programme of systematic research in this field, the authors have elaborated new methods to test some essential properties of powders and granulates used in the production of compressed tablets. The following test and production methods have been developed: (i) the determination of flowing-sliding characteristics of granulates based on the measurement of the mass-flow (g s(-1)) and mass-flow density (g s(-1) cm(-2)); (ii) the determination of optimum granulometric parameters with regard to tablet diameter; (iii) the direct determination of the temperature-dependent equilibrium vapour pressure; and (iv) the compression of tablets under controlled temperature.

[Action of oestrogens on in vitro metabolism of trophoblast from human early pregnancy (author's transl)].

Warburg's manometric method was used to check the action of oestrone, oestradiol, and oestriol on aerobic and anaerobic glycolysis of placental respiration. Oestrogen concentrations of 10(-4) M were found to reduce oxygen consumption and to increase aerobic glycolysis. Such reduction of oxygen consumption was most strongly pronounced in connection with oestradiol, while the strongest rise in aerobic glycolysis took place in the wake of oestradiol and oestrone. Oestrogen action upon anaerobic glycolysis was variable, with the latter remaining unchanged by oestradiol, reduced in response to oestriol, and slightly increased by oestrone.

[The effect of prednisolone and ACTH on the in vitro metabolism of human placenta from early pregnancy (author's transl)].

The effects of prednisolone and ACTH on in vitro oxygen consumption and aerobic and anaerobic glycolysis of human placentas of early pregnancy were studied. Oxygen consumption and anaerobic glycolysis were significantly reduced by prednisolone, while ACTH resulted in a significant increase in aerobic glycolysis during the 2 hr experimental period.

[The effects of progesterone and androgens on in vitro metabolism of human placenta from early pregnancy (author's transl)].

The effects of progesterone, testosterone and dehidroisoandrosterone on oxygen consumption, aerobic CO2-production and anaerobic glycolysis of human placentas from early pregnancy were studied. Oxygen consumption and anaerobic glycolysis were reduced by progesterone and dehidroisoandrosterone. Testosterone inhibits oxygen consumption first off all, while moderately decreases CO2 production in aerobic environment.

Intrapleural streptokinase in the treatment of haemothorax and empyema.

Thirty-eight patients with haemothorax or empyema, in whom conventional drainage treatment had proved ineffective were treated with streptokinase instillations. An increased yield of fluid was noted in all cases, up to 1300 ml after one instillation. Re-expansion of the lung was observed radiologically in 30 cases. No serious complications occurred. The method seems very useful and is recommended.

A method of computing drug distribution in plasma using stepwise association constants: clofibrate acid as an illustrative example.

A program is described to compute the distribution of ligand among various complexes of ligand and protein using stepwise association constants. The key input data consists of stepwise association constants and the concentration of macromolecule and various concentrations of ligand. The key output data consists of the concentration of free drug and protein and the concentration of the various complexes.
