A method of computing the distribution of a ligand among multiple binding sites on different proteins in plasma: thyroxine as an illustrative example.

This paper describes a computer program for estimating the amount of free or unbound ligand in the plasma as well as the distribution of the ligand among different sets of binding sites on different proteins. The input data consist of: the number and concentrations of proteins involved, the number of binding sites on each protein, the association constant and binding capacity of each set of sites for the ligand, and the total concentrations of ligand. The output provides tables of estimates of the concentration of free drug, the amount of ligand bound by each set of sites on each protein, the concentration of free protein, and the input values for reference. The distribution of thyroxine between thyroid binding globulin, prealbumin, human serum albumin and free drug is used as an illustrative example.

The distribution of halofenate in plasma: a comparative analysis using Scatchard vs. stepwise association constants.

The distribution of Halofenate between the free and albumin bound forms was calculated by the use of two computer programs using both Scatchard association constants for a 3,3,7 model, and six stepwise equilbrium constants over a range of drug concentrations reported in man. The calculations, using either set of constants, showed that only 0.3 to 0.5% of the drug would be free. Using Scatchard association constants, it was estimated that 92 to 95% of the drug would be bound by the high affinity set of sites, and lesser amounts by sites of lesser affinity. A more complex pattern of distribution was obtained with the stepwise equilibrium constants. At low concentrations of Halofenate the complex with one mole of drug/mole of protein was most abundant and at the highest concentration studied the complex with two moles of drug/mole of protein was most abundant.

Changes in the alkaline phosphatase activity of granulocytes from the first to the sixth day of life in newborns.

GAP activity was studied in 40 newborns, from the first to the sixth day of life after normal pregnancy and labor. Evaluation of GAP activity was performed in stained blood smears by the method of Kaplow. The results of this study indicate, that GAP activity on the first day of life was higher (121.0 +/- 16.7 GAP score) than on the sixth day (68.6 +/- 12.0 GAP score). The authors, discussing the possible cause of the increased alkaline phosphatase activity in granulocytes during the first day of life, cannot exclude the possibility that the GAP activity is influenced by estrogens of maternal origin.
