A precursor-product relationship exists between oval cells and hepatocytes in rat liver.

Oval cell proliferation was induced in twelve male Fischer rats by administration of 2-acetylaminofluorene (2-AAF) for 2 weeks and by performing partial hepatectomy one week after the beginning of 2-AAF administration. Albumin expression in liver was studied by using in situ hybridization of 3H-labeled rat albumin riboprobe. Radiolabeled thymidine was administered to a group of animals at day 6 after partial hepatectomy. The animals were killed at 0, 3, 7, 9, 11 and 13 days after partial hepatectomy. Both oval cells and basophilic hepatocytes showed a prominent expression of albumin, whereas albumin expression in acidophilic pre-existing hepatocytes was decreased. Oval cell nuclei were exclusively labeled one day after administration of [3H]thymidine. At day 9, 11 and 13 basophilic hepatocytes became labeled and the area occupied by these cells increased. This is the first demonstration of the transfer of radiolabeled thymidine from oval cells to newly formed hepatocytes in vivo. Thus the precursor-product relationship between oval cells and basophilic hepatocytes has been established.

Experiences with the treatment of clinical hyperprolactinaemia.

67 patients with high serum prolactin values were treated by bromocriptine (Parlodel). Parlodel was administered in 56 cases because of functional infertility, and 27 of them became pregnant. The previously given traditional ovulation inductors were ineffective in many cases. According to the authors' experience Parlodel can be favourably combined with clomiphene citrate. On the other hand, Parlodel treatment could be effectively introduced in cases with only moderately high prolactin values (400-800 mIU/ml) to restore fertility. Parlodel can be effective also in the treatment of polycystic ovarian syndrome.

Antitumor action of N-(2-chloroethyl)-N-nitrosocarbamoyl derivatives of biologically active polypeptide hormone fragments.

The antitumor action of the 2-chloroethylnitrosocarbamoyl derivatives of peptides related to the 9-13 amino acid residues of alpha-MSH/ACTH and of the C-terminal tetrapeptide analogue of gastrin have been investigated. Series of 2-chloroethylnitrosoureas attached to amino acids, di-, tri-, tetra-, or pentapeptides were examined in a primary screening system. Among these compounds the Pro-Val-, Lys-Pro-Val-, and Trp-Gly-Lys-Pro-Val-containing 2-chloroethylnitrosocarbamoyl groups were the most effective in the L1210 system. The human melanoma xenograft line was also affected by these agents, while colorectal xenografts were insensitive. A combination of tripeptide-2-chloroethyl-nitrosourea with BCNU induced more than additive growth inhibition of L1210 leukemia.

Electrostatic lock-and-key model for the study of biological isosterism: role of structural water in the binding of basic pancreatic trypsin inhibitor to beta-trypsin.

We propose the electrostatic lock-and-key model for the analysis of the interaction between beta-trypsin and basic pancreatic trypsin inhibitor (BPTI). Prerequisite for the proper recognition of the ligand by the protein is that, beside a steric complementarity, matching of electrostatic patterns is attained. It is found that the complementarity is imperfect in the vicinity of BPTI backbone carbonyl oxygen atoms and this imperfection is diminished by the presence of structural water molecules bound to the contact surface. Some novel types of biological isosteres are proposed. It is expected that the Gibbs free energy of binding increases upon changing the moieties greater than C = O ... H-OH and greater than C = O to greater than CHCH2 CH2OH and greater than CHOH groups, respectively.

Platelet function studies in myeloproliferative disorders.

Platelet functions were studied in 64 patients with various myeloproliferative diseases. The characteristic alterations were prolonged bleeding time, decreased platelet aggregation (but normal results induced by ristomycin), elevated level of BTG, high production of MDA, increased level of TXB2 with almost normal level of 6-keto-PGF1. However, considering the bleeding time and the amount of BTG in relation to the whole blood platelet count, no differences could be detected.

Chemotherapy of human non-Hodgkin lymphoma (NHL) xenografts.

Three human NHL (B-cell type) were established successfully as serially transplantable xenografts in artificially immunesuppressed CBA mice. All of them preserved the phenotypic characteristics of the original tumour even after several passages. The transplanted tumours were highly sensitive to cyclophosphamide and methotrexate, reflecting the results obtained in clinical practice, and relatively sensitive to dianhydrogalactitol.

A double-quenching method for studying protein dynamics: separation of the fluorescence quenching parameters characteristic of solvent-exposed and solvent-masked fluorophors.

A novel experimental method, suitable for separate analysis of the quenching parameters characteristic of solvent-exposed and solvent-masked fluorophors of macromolecules, is described. The method is based on the modified Stern-Volmer analysis and requires simultaneous application of two kinds of quencher: one that can selectively quench the emission of exposed fluorophors (e.g., ionic quenchers) and another that is nonselective (e.g., oxygen or, in many cases, acrylamide), capable of quenching the fluorescence of both exposed and masked groups. In order to examine the accuracy of the model, a computer simulation was performed. The results showed that the errors are comparable to those arising from the conventional quenching experiments. The method is applicable to phosphorescence quenching as well and is extendable to time-resolved measurements (by replacing fluorescence intensities with lifetimes). The method was applied to resolve the quenching parameters of lysozyme fluorescence by the use of iodide as selective and acrylamide as nonselective quenchers. The determination of the acrylamide quenching constant associated with the internal fluorophor, Trp-108 (Kq = 3.5 M-1), permits specific studies on the dynamics of internal regions of the protein. The quenching constant determined for the more exposed residue Trp-62 (Kq = 1.6 M-1) provides local information about the surface independent of the electrostatic effects observed when an ionic quencher is used.

Problems of differential diagnosis in a case of bone lesions caused by myelosclerosis associated with hyperparathyroidism.

The authors present a report on the case of a female patient with myelofibrosis where X-ray examinations of the bone skeleton predominantly showed such osteolytic changes which are no typical of myelosclerosis. Morphological criteria rather would have corresponded to osteitis fibrosa cystica generalisata. Laboratory examinations partially indicated primary hyperparathyroidism, this assumption could not be confirmed by other examinations. Also histological findings of repeated bone biopsies did not lead to a uniform diagnosis. Autopsy confirmed the presence of both diseases. The authors consider the synergistic effect of both diseases to be the cause for the development of those bone changes mentioned above.

Cytohormonal and cancer screening procedure for endometrial carcinoma.

The results of cancer-screening (by Papanicolaou) and of cytohormonal examinations were evaluated in 208 patients with endometrial adenocarcinoma. The occurrence rate of atypical cells in vaginal smears was 44.2%. It was higher in the more advanced cases and after the menopause, and an inverse relationship was found with the degree of maturation of the vaginal epithelium. -It was concluded that the combination of hormonal and cancer-screening cytodiagnostic procedures, using vaginal smears alone, can hardly facilitate the early diagnosis of endometrial carcinomas. Higher efficiency could only be expected from direct endometrial smears and by frequent screening of the population at highest risk.

Semisurgical treatment of giant condylomata.

A semisurgical method to treat giant condylomata acuminata is described. Several silk ligatures are placed on the tumourous mass. Complete recovery was found to occur even in very extensive lesions. The procedure proved to be safe, neither bleeding, late scarring, nor local recurrence were observed.

Renal cell carcinoma--xenotransplantation into immuno-suppressed mice.

21 human renal cell carcinomas (RCC) were xenotransplanted into artificially immunosuppressed mice. 4 tumors grew successfully retaining some characteristics of the primary tumors (according to morphology and karyotype analysis), but losing metastatic capacity. One of the serially transplantable tumors (HT 40) with hyperdiploid cellular DNA content and estrogen receptor positivity failed to respond to the single maximally tolerated dose of several cytotoxic agents.
