In order to elucidate mechanisms of protein plug formation, histochemical studies were performed on aggregates and protein plugs present in pancreatic juice. Pancreatic juice was obtained from three control subjects and five patients with chronic pancreatitis through endoscopic retrograde catheterization of the papilla. Specimens for staining were prepared in two ways: (1) fixed with 10 per cent formaldehyde, embedded in paraffin and sectioned, and (2) placed on slide glass and fixed with isopropylalcohol. Staining included hematoxylin-eosin, periodic-acid Schiff, von Kossa, alcian blue, toluidine blue and double staining with PAS and AB. The process of protein plug formation can be as follows: (1) a prerequisite for aggregate formation, consisting of clusters of desquamated epithelial cells, highly concentrated sulfated acidic mucopolysaccharide and neutral mucopolysaccharide, (2) formation of aggregates in which epithelial cells and amorphous substance are interlaced with developing fine reticular substance, (3) enlargement of aggregates by fusion with adjacent aggregates through bridging action of the reticular substance sprouting, like prickles, from their surface, and (4) "maturity" of aggregates, taking a three-dimensional form which result in a spherical, spheroidal or cylindrical protein plug.
Mouse myeloid leukemia cells can be induced to differentiate into macrophages in vitro by 1 alpha,25-dihydroxyvitamin D3, the active form of vitamin D3. The minimal concentration of 1 alpha,25-dihydroxyvitamin D3 to induce the cell differentiation was 0.12 nM. The degree of cell differentiation in various markers induced by 12 nM 1 alpha,25-dihydroxyvitamin D3 was nearly equivalent to that induced by 1 microM dexamethasone, the most potent known stimulator. Among several markers of the differentiation by 1 alpha,25-dihydroxyvitamin D3, phagocytic activity was induced within 24 hr, and this was followed by induction of lysozyme and locomotive activities. Similar changes were also induced by 0.01-1 microM 1 alpha-hydroxyvitamin D3. 25-Hydroxyvitamin D3 and 24R,25-dihydroxyvitamin D3 showed only weak inducing activity. These results suggest the possibility that, in addition to its wellknown biological activities in enhancing intestinal calcium transport and bone mineral mobilization, 1 alpha, 25-dihydroxyvitamin D3 is involved in the differentiation of bone marrow cells.
Pig heart phosphoprotein phosphatase [phosphoprotein phosphophydrolase, EC 3.1.3.16] of Mr 224,000 was dissociated by gel-filtration on Sephacryl S-300, into an active subunit (alpha subunit) of Mr 31,000 and inactive subunits of higher molecular weight in the presence of 6 M urea. After the removal of urea, these subunits reassociated, forming two enzyme forms of Mr 237,000 (Form 1) and Mr 123,000 (Form 2). Form 2 was produced by association of the alpha subunit with an inactive subunit (beta subunit) of Mr 80,000, while Form 1 was formed by combination of the alpha subunit with a complex of inactive subunits which was eluted from a Sephadex G-150 column in fractions of molecular weight range greater than 80,000. The dissociation and reassociation of the subunits of Form 1 by the same urea method produced not only Form 1, but also significant amounts of Form 2, indicating that the inactive subunits of Form 1 were a complex of the beta subunit with another inactive subunit(s). The molecular parameters and other properties of Form 1 were very close to those of the original enzyme. By the conversion of Form 1 to Form 2, the activities of Form 1 towards phosphorylase a and glycogen synthetase b were enhanced 2-3 fold with no significant change in activity towards P-H1 histone or in response to the stimulatory effect of Mg(CH3COO)2 on the dephosphorylation of P-H2B histone. However, removal of the beta subunit from From 2 resulted in strong suppression of activity towards P-H1 histone and response to the salt effect with lesser effects on the activities of Form 2 towards phosphorylase a and glycogen synthase b.
Beck's dual system theory (DST) is examined theoretically and experimentally by investigating the oviposition rhythm of Ostrinia nubilalis and its entrainment by light cycles. Several well-known circadian phenomena are not accounted for by the DST. 1) It does not generate transient cycles when light pulses fall during the advance portion of the circadian cycle. This is also reflected in DST-predicted phase-response curves (PRC's) for both Drosophila pseudoobscura and O. nubilalis. Steady-state phase advances are predicted to occur on day 1 after the light pulses by the DST, not several cycles later as has been observed in many cases. 2) It does not account for the observation that the magnitude of a phase shift (delta phi) is often a function of pulse duration of both delays and advances. The DST predicts the same + delta phi, for example, for a 0.5-h and a 6.0-h light pulse beginning 5.0 h after dusk. 3) The DST does not accurately predict steady-state phase relationships between the light cycle and the gating oscillation (P-system) in non-24-h light cycles. 4) The driver (S-system) is given the property of being temperature sensitive whereas the driven rhythm (P-system) is temperature compensated. This is contrary to accumulated data suggesting that the circadian pacemaker is temperature compensated.

After administration of monoamine precursors, developing taste buds of newborn and young mice were observed by means of electron microscopy and fluorescence histochemistry. Gustatory (type III) cells occurred in the primitive taste buds during stage 1 (0-1 day after birth). These cells had an immature type of afferent synaptic contacts with nerve terminals; however, no specific fluorescence was found in the taste buds after administration of 5-HTP or L-DOPA. During stage 2 (2-7 days), mature types of afferent synapses, taste pores, type I cells and type II cells appeared in the taste buds, and fluorescent cells also appeared following treatment of 5-HTP or L-DOPA. During stage 3 (14-21 days), the gustatory cells underwent ultrastructural changes following injection of 5-HTP; i.e. small dense-cored vesicles (30-60 nm) appeared scattered throughout the cytoplasm and were found to intermingle with small clear vesicles accumulated at the presynaptic membranes of afferent synapses, and the electron densities of large dense-cored vesicles (80-100 nm) were elevated as compared with those of untreated mice. Consequently the ability of gustatory cells to take up amine-precursors started simultaneously with the formation of taste pores and mature afferent synapses between the gustatory cells and the sensory nerves.
