Mycoplasma hyopneumoniae infection in pigs immunosuppressed by thymectomy and treatment with antithymocyte serum.

The effect of immunosuppression on Mycoplasma hyopneumoniae infection was evaluated by comparing data from infected, thymectomized, and antithymocyte serum-treated pigs (group 1) with data from infected (group 2) and noninfected (group 3) healthy pigs. After groups 1 and 2 pigs were inoculated intranasally with M hyopneumoniae, mycoplasmas tended to multiply slightly more in the lungs and bronchial lymph nodes of group 1 pigs than that of group 2 pigs. Organisms were also isolated from the spleen of 1 of 3 group 1 pigs. Pneumonia developed in group 2 pigs and was characterized by massive peribronchial, peribronchiolar, and perivascular lymphoid hyperplasia and exudate consisting mainly of polymorphonuclear leukocytes in the alveoli and lumina of the bronchioles and bronchi. In group 1 pigs, perivascular and peribronchiolar cuffings by lymphocytes were less prominent, and the extent of intraluminal exudate was severe and widespread. Bronchial lymph nodes from group 2 pigs had marked hyperplasia of germinal centers and paracortical areas. In group 1 pigs, germinal centers were hyperplastic, whereas in the paracortical areas, depletion of lymphocytes was evident. Seemingly, cell-mediated immune mechanisms are important in the development of pneumonic lesions in enzootic pneumonia of pigs.

Effect of levamisole on the immune response of chickens to infection with Eimeria tenella.

The effect of levamisole on the immunization to coccidiosis in chickens was investigated. The protection to the challenge infection with Eimeria tenella was higher in those chickens immunized by inoculating a small number of oocysts of E. tenella and administered orally with levamisole at dosage levels of 1.25 to 5 mg/kg body weight, once daily for 3 days, one week after immunization than in immunized and levamisole-untreated or unimmunized and levamisole-treated chickens. The activities of peritoneal macrophages of chickens from the immunized and levamisole-treated group were higher than those of chickens from the other groups. Furthermore, serum antibody titers and the leucocyte migration inhibitory factor-inducing activities measured in immunized and levamisole-treated chickens reflected the immune state of chickens.

[A case of primary lipogranuloma of the spermatic cord].

A 54-year-old male who noticed a growing mass in his right inguinal region one month earlier visited our clinic with a pigeon-egg sized, hard elastic, and movable mass with an unsmoothed surface. The removed mass was 4 X 2 X 2.5 cm, weighed 11.5 g and its cut surface was greyish white. The histopathological diagnosis was primary lipogranuloma. Postoperative course was uneventful with no sign of recurrence one year later.

[Chronic cardiotoxicity of anthracycline derivatives and possible prevention by coenzyme Q10].

Adriamycin (ADR), one of the anthracycline derivatives, has the most strong cardiotoxicity. We studied the cardiotoxicity caused by ADR in New Zealand white rabbits and its protection by the medication of CoQ10. The findings of ECG and the myocardial tissue examined by the electron microscope showed the effectiveness of the injection of CoQ10 to prevent the cardiotoxicity caused by ADR. The concomitant injection of CoQ10 dissolved in saline was tried in patients with various kinds of neoplasm who were given more than 200 mg of ADR or DM. Only one patient showed the ST-T change. On the contrary, 3 of patients given more than 200 mg ADR or DM alone showed abnormal change of ECG.

[Serum tissue polypeptide antigen (TPA) in patients with urogenital cancer].

We measured the serum TPA level in 260 patients with benign urogenital diseases, 189 patients with urogenital cancer and 72 healthy blood donors by using a radio-immunoassay kit in order to evaluate its usefulness as an indicator for the presence of cancer. TPA value (mean +/- 2SD) in healthy blood donors was in the range of 36.8-110.8 units per liter, so that values higher than 110 U/L were considered to be abnormal. Significantly higher serum TPA levels were observed in patients with non-treated cancers, compared to healthy blood donors. But elevated serum TPA levels were also observed in the patients with benign diseases, especially in cases of benign prostatic hyperplasia with urinary tract infection. Consequently, it is considered that the serum TPA test is very useful for detecting urogenital diseases, but not for screening urogenital cancer. In cases of urogenital cancer, serum TPA levels elevated significantly by recurrence or recrudescence of the disease. Therefore, a good correlation was established between serum TPA and the efficiency of a given therapy in patients with prostatic cancer and bladder cancer.

Myoclonus in a case of suspected progressive rubella panencephalitis.

Predominantly unilateral myoclonus induced by movement of the affected parts in a 21-year-old Japanese man with suspected progressive rubella panencephalitis appeared in the course of treatment for epilepsy with anticonvulsant drugs after five years. Antirubella antibody levels in the serum and CSF were elevated and IgM antibody against rubella was found in the serum. An EEG recorded while the patient was awake showed diffuse 3- to 4-Hz theta activity with high voltage. The neurological symptoms are progressing slowly.

Isolation of new serotypes of bovine rotavirus. Brief report.

Two cytopathogenic bovine rotavirus strains were isolated. The infectivity of both isolates was relatively stable at pH 3.0 and resistant to ether. Their replication was not affected by 5-iodo-2'-deoxyuridine. The new isolates shared immunofluorescent antigen with the Lincoln strain of bovine rotavirus. By neutralization test, however, these strains were clearly distinguished from one another.

Effect of age on plasma bile acids and lipid components in the rat.

With increasing age, total plasma bile acid contents increased in rats over a period of 11 months, and also total plasma cholesterol and carcass fat contents increased in the same manner. Plasma showing high bile acid levels at 11 months was found by means of high performance liquid chromatography to contain cholic acid as one of the major components, chenodeoxycholic acid and trace deoxycholic acid. These results suggest that there are close relationships between the plasma bile acids and age-dependent changes of lipid components in the rat.

Activation of macrophages by culture fluid of antigen-stimulated spleen cells collected from chickens immunized with Eimeria tenella.

The effect of spleen cell factors on the activation of macrophages was investigated in chickens immunized with Eimeria tenella. The abilities of peritoneal macrophages obtained from normal chickens to kill sporozoites of E. tenella and to inhibit intracellular development of Toxoplasma gondii were enhanced by exposing them to 33 and 50% culture fluid of antigen-stimulated spleen cells of chickens immunized with E. tenella. The parasiticidal activity of normal macrophages was also distinctly enhanced by the treatment with culture fluid of phytohemagglutinin-stimulated normal spleen cells. On the other hand, the parasiticidal activity of normal macrophages could not be enhanced by the treatment with culture fluid of antigen-stimulated normal spleen cells under conditions similar to those of culture fluid of antigen-stimulated immune spleen cells. It thus appears that the macrophage activator was induced from immune spleen cells in response to the stimulation by the antigen.

A new mutant of Japanese quail (Coturnix coturnix japonica) characterized by generalized glycogenosis.

A flock of Japanese quail with generalized glycogenosis has been established. Affected quail showed difficulty in raising their wings. Excessive accumulation of glycogen was seen in the liver, heart, skeletal muscle and brain, apparently due to decreased acid maltase activity. The condition appeared between 2 and 12 weeks of age and tissue deposition of glycogen increased with age. The growth of affected quail was normal and there were no deaths from the condition. Although genetic analysis has not yet been completed, an autosomal recessive inheritance is suspected.

A hereditary nervous disorder in Fayoumi chickens.

A hereditary nervous disorder in Fayoumi chickens is described. Affected chickens developed seizure symptoms characterized by sudden crouching to the ground with neck rigidly flexed. The duration of the seizure varied from case to case, usually lasting for 15 to 30 s. The seizure was triggered by unexpected loud noise, sudden movement, bright light and other audiovisual stimuli. The main histopathological lesions consisted of degeneration of small arteries, with cellular infiltration in the cerebrum of all chickens examined and demyelination and/or loss of nerve fibres with oedema and Schwann cell proliferation in the cervical spinal nerves of most birds. Genetic analysis revealed that the condition was heritable and apparently controlled by an autosomal recessive gene.

Release of lipoteichoic acid from Staphylococcus aureus by treatment with cefmetazole and other beta-lactam antibiotics.

The effect of cefmetazole on the growth together with the release of cellular lipoteichoic acid from cefazolin-resistant strains of Staphylococcus aureus was compared with that of cefazolin, cefotiam, cefoxitin and cefuroxime. Bacteriolytic actions were measured by turbidity and bactericidal actions were followed by viable cell count. Release of cellular lipoteichoic acid was measured by the radioactivity in the supernatant of the cultures. Cefmetazole exerted more potent effects on the bacterial growth and induced more marked release of cellular lipoteichoic acid from resistant strains as compared with other beta-lactams.

The beta-lactamases of genus Bacteroides.

Two hundred anaerobic isolates from clinical specimens were examined for beta-lactamase production by Nitrocefin methodology. Altogether, 77 strains were beta-lactamase producers. Organisms belonging to Bacteroides melaninogenicus group, i.e., B. intermedius and B. bivius, were found to have a significant frequency of beta-lactamase production. Substrate profile and sensitivity to the beta-lactamase inhibitors, sulbactam, clavulanic acid, cloxacillin, and cefmetazole, a cephamycin derivative, were determined for these enzymes. All enzymes hydrolyzed cephalosporins more rapidly than penicillins. Cefmetazole was not hydrolyzed at all. Strains of B. fragilis, B. thetaiotaomicron, B. uniformis, B. ovatus and B. oralis produced beta-lactamases sensitive to all four inhibitors. Strains of B. intermedius, B. bivius and B. disiens produced enzymes of different nature which were inhibited only by cefmetazole. B. vulgatus enzyme was inhibited by three of the four inhibitors. These results suggest that the beta-lactamases of the genus Bacteroides may be classified by substrate profile and inhibitor pattern.
