Primary structure of the Saccharomyces cerevisiae GAL7 gene.

We present the nucleotide sequence of a 1599-base pair (bp) DNA fragment containing the entire GAL7 gene that encodes galactose-1-phosphate uridyltransferase of Saccharomyces cerevisiae. The deduced peptide was composed of 364 amino acid residues. The expected molecular weight was 42,005 daltons, which agreed with the observed value for the purified enzyme. The 3'-end of the GAL7 transcript mapped at a position 82 bp downstream from the UAA termination codon by the S1 nuclease protection experiment. We constructed a GAL7'-lac'Z fusion on various types of yeast plasmid vectors. The fused gene on any type of vector was induced by galactose and repressed by glucose as for the GAL7 gene on the chromosome. The response of GAL7'-lac'Z fusion to gal4 delta and gal80 delta regulatory mutations was also similar to the response of the chromosomal GAL7 gene. By using various deletions in the 5'-flanking region of the gene fusion, we delimited the sequence essential for galactose controlled expression with a 180 bp-fragment of DNA lying 92 bp upstream of the transcription initiation site.

A new mutant strain of Syrian hamster with myelin deficiency.

A new disorder of central myelination was found in black mutant hamsters showing trembling. The condition was inherited in an autosomal recessive mode. Histologically, a marked myelin deficiency was distributed throughout the central nervous system (CNS), and it was closely related to clinical manifestations observed in the mutants Ultrastructurally, abnormally thin myelin sheaths were observed in all axons with different diameters, and most axons below 1 micron in diameter were unmyelinated in the white matter of the spinal cord. There was no overt evidence of demyelination, but some myelin sheaths showed an aberrant configuration. The numbers of glial cells were not reduced, and there were no striking morphological abnormalities in them. It was suggested that there may be a dysfunction of oligodendrocytes resulting in retarded or defective myelination. This trembling hamsters may be a suitable animal model for the study of normal and abnormal myelinogenesis.

[An outbreak of staphylococcal dermatitis in laboratory mice].

An epizootic of dermatitis with erosion, ulcer and crust broke out in an experimental colony of JCL-ICR mouse over a period from December 1975 to June 1976. The disease was detected in 592 of a total of 1831 mice of 3-24 months old, especially in males of 7-24 months old (517/821). At the beginning of December 1975, only a few males of 12 months old were found to have the lesion on the back skin, and thereafter the dermatitis prevailed gradually among the mice. Histopathologic examinations showed the loss of the epidermis, necrosis and/or collapse of the corium, accumulation of serous exudate with neutrophilic cell infiltration and a few cocci scattered on the surface. In chronic cases, fibrous granulation tissues with neutrophilic cell infiltration were formed in the corium. Staphylococcus aureus was isolated in pure culture from the skin lesions in all of the mice examined. Skin disease similar to that of the field case was reproduced in mice inoculated subcutaneously with 10(7) viable organisms of the fresh isolate. By giving chlortetracycline in drinking water for 7 days, treatment of the affected mice was efficacious in mild cases, but not in severe cases.

A new method of preparing specimens for cytodiagnosis of lung cancer.

A simple and effective method for concentrating sputum in the preparation of smears for cytodiagnosis of lung cancer was developed. The procedure, which utilizes a fixative consisting of 2,3-dihydroxy-1,4-dithiol-butane, 3,5-ditert-butyl-4-hydroxy-toluene, polyoxyethylene-cetyl-ether, polyethylene glycol 1540, and ethyl alcohol in distilled water permitted preparation of an amucoid, thin smear. Sputum was collected in a screw-cap plastic tube containing the fixative. After 12 hours mucus in the sputum was liquefied and the cellular elements were obtained from the bottom of the tube. Cellular morphology was well preserved for cytodiagnosis. Microscopic observation was facilitated by lysis of the mucus. Estimation of cellular origin was rather easy because of the distinct fixation. Cells from the sputum sediment following liquefaction were more representative of the entire specimen than with a random selection method. The new method should be useful not only in hospitals but also in mass surveys.

Cefmetazole: a broad spectrum cephem antibiotic effective on methicillin- and cephem-resistant Staphylococcus aureus.

Antibacterial activity of cefmetazole (CMZ) was investigated by the plate dilution method. Since CMZ is stable to any type of bacterial beta-lactamases, it inhibited growth of Escherichia coli carrying R plasmids, Klebsiella spp., Proteus vulgaris, and Bacteroides fragilis at the concentration of less than 0.78 to 25.0 micrograms/ml. The distinct characteristic of CMZ is its anti-MRSA (methicillin- and cephem-resistant Staphylococcus aureus) activity. Exclusively in MRSA, a new fraction of penicillin binding proteins (PBP) with a relative molecular mass of 78 kd apears, and the 78 kd/PBP possesses low binding affinity to beta-lactam antibiotics. By the competitive binding experiment of beta-lactam drugs to the PBPs of MRSA, it was revealed that CMZ and cephaloridine retain binding affinities to the new 78kd/PBP fraction of MRSA.

[A case of metastatic carcinoma of the spermatic cord due to cholangioma].

Tumor in the spermatic cord is infrequent. Further, among such cases, it is extremely rare to encounter a carcinoma other than a nonepithelial malignant tumor; almost all are metastatic tumors. We recently experienced an interesting case of spermatic cord tumor in a 78-year-old male. He entered our hospital with the chief complaint of a right inguinal tumor. He underwent 2 operations to excise the tumor. The extirpated tumor mass from the right spermatic cord was composed of adenocarcinoma with abundant fibrous stroma, which is considered to be metastasis from the other viscerae. We were unable to find the primary focus in spite of extensive clinical and pathological examinations. In an autopsy about four months after the 2nd operation, the tumor was confirmed to be primary intrahepatic bile duct carcinoma, which is called cholangioma. There have been only 20 cases of metastatic spermatic cord tumor reported in Japan. However, we considered that this is the 1st case caused by cholangioma in Japan and even in other countries.
