The inhibitory effects of colchicine, vinblastine and cytochalasin D on lymphocyte blastogenic responses.

The inhibitory effects of colchicine, vinblastine and cytochalasin D on blastogenic responses of equine, bovine and canine peripheral blood lymphocytes (PBL) were investigated. These drugs inhibited blastogenic responses of each PBL. The concentrations for 50% recovery in PBL blastogenic response of colchicine and vinblastine were lower in equine and canine PBL than in bovine PBL. This suggested that microtubules may be more concerned in blastogenic response of equine and canine PBL than of bovine PBL. On the other hand, the concentration for 50% recovery in PBL blastogenic response of cytochalasin D was almost the same in the PBL of each animal. This meant that microfilaments made only a small contribution to the lymphocyte blastogenic response.

Morphologic characteristics of a transplantable tumor derived from a spontaneous malignant fibrous histiocytoma in the rat.

Malignant fibrous histiocytoma (MFH) developed spontaneously in subcutaneous tissue of the head of a 15-month-old male Fischer 344 rat. The tumor was serially transplanted into syngeneic rats up to the 45th generation and was designated MFH-MT. Light and electron microscopic examinations revealed that the original and serially transplanted tumors were composed of an admixture of fibroblast-like and histiocyte-like cells arranged in a storiform pattern. Neoplastic cells gave positive reactions for acid phosphatase, alkaline phosphatase, nonspecific esterase, alpha-1 antitrypsin and lysozyme. The tumors transplanted into the lungs and cutaneous tissue of the tail had a mixed histologic appearance of storiform, pleomorphic, myxoid and giant cell types. Moreover sclerosing hemangioma-like and osteosarcoma-like structures were also found. MFH-MT grew well in athymic nude mice showing neoplastic proliferation of pleomorphic cells strongly positive for alpha-1 antitrypsin. Development of MFH-MT was significantly retarded by the two antitumor drugs tested. The retarded tumors consisted predominantly of fibroblast-like cells and abundant collagenic fibers, whereas histiocytic cells decreased in number.

Role of lymphocytes in dogs experimentally re-challenged with canine transmissible sarcoma.

The role of lymphocytes in dogs re-challenged with canine transmissible sarcoma (CTS) was investigated histologically and immunologically. Tumors were detected on the 3rd day, began to regress on the 9th day and disappeared on approximately the 15th day after the secondary transplantation (AST). The CTS cells were surrounded by lymphocytes which had infiltrated from the early stage, degenerated and ultimately disappeared. Most of the lymphocytes expressed thymocyte antigen (Ta) on the cell membrane surface. The numbers of lymphocytes and Ta-positive lymphocytes in peripheral blood increased gradually AST. The blastogenic responses of peripheral blood lymphocytes (PBL) induced by 3 kinds of mitogens were elevated strikingly from the early stage AST. These results suggest that Ta-positive lymphocytes play an important role in vivo in the regression of tumors and that the immunological activity of PBL is stimulated immediately AST.

Characteristics of in vitro passaged cells derived from a rat transplantable malignant fibrous histiocytoma.

Morphologic and functional characteristics were investigated on in vitro passaged cells (MT-P) derived from a rat transplantable malignant fibrous histiocytoma (MFH-MT). There were spindle, polygonal, and giant cell types in MT-P. Ultrastructurally, the polygonal and giant cells had the abundant cytoplasm with many lysosomes and processes, whereas the spindle cells possessed smooth cell surface and a small number of lysosomes in their cytoplasm. Immunorosette formation for Fc- and C3-surface receptors and phagocytic activity were demonstrated in 10-20% of MT-P. MT-P were positive for acid phosphatase, nonspecific esterase and alkaline phosphatase. Chromosomes counted in 100 MT-P ranged from 32 to 100 with two peaks of 64 and 76. Tumors induced in syngeneic rats by inoculating MT-P showed variable histologic patterns. They were composed partly of histiocytic cells arranged in a compact sheet. Fibroblastic cells often arranged in a storiform pattern or were supported by myxoid matrix. Osteosarcoma-like structures were occasionally found in the tumors. These results suggest that MFH-MT is heterogeneous, although some cells constituting the tumors have histiocytic markers.

Prerequisites in distance running performance of female runners.

We investigated which attribute or what combination of attributes would best account for distance running performance of female runners. The subjects were 30 well-trained female distance runners, aged 19 to 23 years. Anthropometric and body composition characteristics, pulmonary function characteristics, blood properties, and cardiorespiratory function characteristics were measured at rest or during submaximal and maximal exercise. Analyses of the data showed that the relationship of oxygen uptake corresponding to lactate threshold (VO2T, ml.kg-1.min-1) with each distance running performance was substantially higher as compared with the relationship of other independent variables including maximal oxygen uptake (VO2max). Furthermore, multiple regression analysis indicated that running performances in 3,000m, 5,000m, and 10,000m are best accounted for by a combination of VO2/LT (X1), fat-free weight (X2), and/or mean corpuscular volume (X3). A multiple regression equation for predicting the 5,000m (Y, s) running performance was formulated as Y = -14.75X1-3. 03X2-5.79X3 + 2282.1. We suggest that VO2max would not stand alone as a decisive factor of distance running success in female runners, and that the distance running performance could be better accounted for by a combination of several attributes relating to lactate threshold, body composition, and/or hematological status. The linear regression of the predicted running performance on the actually measured running performance can be accepted in the range of 986-1197s.

Membrane viscosity of lymphocytes and influence of phytohemagglutinin.

The membrane viscosity of peripheral blood lymphocytes (PBLs) of equine, bovine and canine was measured by the use of time-resolved fluorescence depolarization technique with 1, 6-diphenyl-1,3,5-hexatriene (DPH). The viscosity values were 0.55, 0.59 and 0.50 poise for equine, bovine and canine PBLs, respectively. These values were compared with steady-state anisotropies and order parameters measured from electron spin resonance (ESR) of 5-doxyl stearic acid. Both values were increased with increase of viscosity. The fluid property of the membranes stimulated with phytohemagglutinin-P (PHA) was measured with steady-state fluorescence anisotropy and ESR. Little change of membrane fluidity was recognized with both methods during the stimulation with PHA. It appears that PHA activation process for these lymphocytes does not included large increase of the membrane fluidity which significantly accelerate the diffusion velocity of receptors in the plasma membrane.

Transformation of Golgi membrane into the envelope of herpes simplex virus in rat anterior pituitary cells.

Envelopment of herpes simplex virus type-1 (HSV-1) was investigated in relation to membrane differentiation in dissociated anterior pituitary cells. The number of cells stained positively with anti-HSV-1 serum was increased from 16 h to 31 h post infection. During this period, electron microscopy revealed that a number of nucleocapsids (unenveloped particles) were accumulated in the Golgi area, where they frequently became surrounded by a double membrane of short Golgi cisternae or by one with a Golgi associated endoplasmic reticulum lysosome (GERL)-like structure. The inner membrane of the cisterna surrounding the nucleocapsids showed regional specialization which was characterized by increased thickness and electron opacity. Acid phosphatase activity, a marker for GERL or trans Golgi cisternae, appeared in the cytoplasmic short cisternae surrounding the nucleocapsids, whereas glucose-6-phosphatase activity, a marker for the nuclear envelope or for endoplasmic reticulum, was not demonstrated in such cisternae. Monoclonal antibody against glycoprotein gD revealed that gD was localized in the trans Golgi membrane as well as in the envelope of the virion. The antibody-binding sites were highly concentrated in the area where Golgi membranes showed increased opacity. Furthermore, nucleocapsids were surrounded exclusively by gD-positive cisternal (Golgi or Golgi-derived) membranes. Thus, our results indicate that the envelope of HSV is derived from trans Golgi cisterna (GERL), and that some viral components, including gD, destined for the envelope may be assembled initially in the Golgi membrane, which is thereby transformed into the envelope of the virus.

[A case of retroperitoneal abscess accompanying pregnancy].

A 40-year-old pregnant woman, who suffered a continuous episode of fever and anemia since the 26th week of pregnancy, was referred to our clinic for evaluation of her left flank pain and tumor. As retroperitoneal tumor was suspected by retrograde pyelography, sonography and CT, translumbar tumorectomy was performed under general anesthesia. The histological diagnosis was retroperitoneal abscess.

[Detection of Chlamydia trachomatis in urethra of patients with urogenital infection].

Between January, 1984 and August 1986, we examined 151 male patients with urogenital infections including 114 cases of urethritis, 18 cases of epididymitis, 19 cases of prostatitis at the outpatient clinic of the department of urology, Toho University's Ohmori Hospital. The positive rate for Chlamydia trachomatis from the urethra based on cell cultures was 29.0%, (30.5% for urethritis, 27.8% for epididymitis, and 21.1% for prostatitis). The positive rate for non-gonorrheal urethritis was 33.0% and chlamydia infections were complicated by gonorrheal urethritis in 23.3% of the cases. The mean age of the patients with positive Chlamydia trachomatis was 31.9 years. The route of infection in 77.8% of the cases was sexual intercourse the most common cause of which was infection from a prostitute in 62.1% of the cases. The latent period was in most cases from 2-3 weeks.

Comparative studies between counterimmunoelectrophoresis and microprecipitation method of identification of human minute bloodstains.

A minute bloodstain on a thread 2 mm in length was tested to identify human origin by counterimmunoelectrophoresis (CIE), modified CIE, and microprecipitation method (MPM), using anti-human HbA serum. The detection limits expressed as the highest dilution of human blood on the thread in positive reaction were 1:160, 1:320, and 1:320 in CIE, modified CIE, and MPM, respectively. About 1 h was required to obtain the results. In CIE and modified CIE, the detection limits diminished according to the reduction of the samples from one half to one-eighth of the thread, but not in MPM.

Primary carcinoma of seminal vesicle. Diagnosis assisted by sonography.

A case of primary carcinoma of the seminal vesicle in a nineteen-year-old man is described. This patient represents the twelfth case reported in the literature in Japan. The experience gained in this case indicated the potential usefulness of ultrasonography in the diagnosis of primary carcinoma of the seminal vesicle.

Primary lipogranuloma of male genitalia.

Lipogranulomas developing secondarily in the genitourinary system have been reported rather frequently, but primary lipogranulomas without any past history of etiologically related conditions are rare. We report on 2 cases recently encountered in which tumors were diagnosed as sclerosing lipogranuloma on histopathologic examination.
