Background pathology in BDF1 mice allowed to live out their life-span.

Fifty male and 50 female BDF1 mice were observed allowing them to live out their life-span. Mortality up to 104 weeks of age was higher in males (42%) than in females (34%), and the 50% survival age was 112 weeks for males and 118 weeks for females. Body weight reached the peak at 82 weeks of age in males and 92 weeks of age in females, showing the mean body weight of 54.3 g for males and 48.0 g for females. The incidence of calculi and proteinaceous casts in the kidneys, that were not associated with exposure to chloroform, cell-alteration in the adrenal cortex, and islet cell hyperplasia in the pancreas was significantly higher in males than in females. On the other hand, hyaline droplet degeneration of the renal tubular epithelium, spindle cell proliferation in the adrenal cortex and milk-retention in the mammary glands occurred at a significantly higher incidence in females than in males. Cerebral mineralization in both sexes, atrophy and calcification of the testes and enlargement of the seminal vesicles of males, as well as cyst-formations in the ovary and endometrium of females developed at a very high incidence. Frequent neoplasms in males were hepatocellular adenomas and carcinomas, blood vessel tumours, pulmonary adenomas and carcinomas, and malignant lymphomas. In females, malignant lymphomas were the most common neoplasm, followed by blood vessel tumours, chromophobe pituitary adenomas and hepatocellular adenomas. Hepatocellular carcinomas developed only in males, whereas the histiocytic and lymphocytic types of malignant lymphomas and chromophobe cell adenomas arose solely or at a significantly higher incidence in females than in males.

The Wolff-Parkinson-White syndrome in a Holstein-Friesian cow.

A case of Wolff-Parkinson-White (WPW) syndrome in a Holstein-Freisian cow aged 10-year-old was examined in detail. In electrocardiogram (ECG), the P-wave was the same configuration in both the normal and abnormal ECG. The PR-interval shortened from 0.2 to 0.1 second and the duration of the QRS-complex prolonged from 0.1 to 0.12 second compared with normal ECG. The delta wave, characterized in WPW syndrome, could not be recognized. In echocardiogram, notches were recognized at the early stage of ventricular contraction in the interventricular septum. This cow was, therefore, diagnosed as type B WPW syndrome. The abnormal ECG disappeared by the administration of procainamide. It was strongly indicated that the ventricular contraction showing abnormal ECG was generated only by the stimulation through an accessory pathway in this cow.

Occurrence of a canine distemper-like disease in aquarium seals.

Outbreaks of a canine distemper-like acute disease brought high mortalities to seal populations in north-west Europe and Lake Baikal from late 1987 to 1988. During these outbreaks three seals which were introduced from Lake Baikal to an aquarium in Japan developed a distemper-like disease and other seals raised in the same room were similarly affected. Clinical signs of dead seals were anorexia, diphasic fever, dyspnea, and neuromuscular tics. Characteristic microscopic lesions of acute interstitial pneumonia seen in the lung were accompanied with hyperplasia and syncytial giant cell formation of type II pneumocytes. Eosinophilic intranuclear and cytoplasmic inclusion bodies were present in bronchial epithelial cells, type II pneumocytes, epithelial cells of bile ducts and interlobular ducts of pancreas, transitional epithelium of renal pelvis, and reticular cells of lymph nodes. Ultrastructure of inclusion bodies was similar to that seen in cells infected with morbilliviruses. Serum samples from recovered seals had virus-neutralization antibodies against canine distemper virus. The present cases were the first report of morbillivirus infection of aquarium seals in Japan.

Hematological changes in the mice transplanted with methylcholanthrene-induced fibrosarcoma.

Hematological values of the peripheral blood in the mice transplanted with methylcholanthrene-induced fibrosarcoma were examined. Values for red blood cells and platelets reduced after tumor transplantation. Values for white blood cells increased with the percentage of neutrophils being increased significantly. Phagocytic activity of neutrophils from the tumor-bearing mice was not reduced. The cultured fluid of the tumor used in the experiments possessed strong colony stimulating activity to bone marrow cells.

[The effect of silicon on the growth of Staphylococcus aureus].

Various studies on the effect of silicon on the development of chronic inflammatory diseases of the mucous membrane such as chronic sinusitis have been carried on for the past twenty years. Staphylococcus aureus is usually detected frequently in chronic sinusitis. In this study, proliferation of Staphylococcus aureus in various culture fluids with Si concentrations ranging from 10 to 410 ppm was observed through 16 hours by means of the measurement of optical density, protein concentration and viability count. As the result of this observation, higher concentration of Si in the culture fluid showed a remarkable growth accelerating effect on Staphylococcus aureus compared with normal culture fluid containing about 10 ppm of Si. High Si concentration in mucous membrane tissue was considered to help the growth of bacteria in the sinus.

Characteristics of cis-diamminedichloroplatinum-selected in vitro passaged cells derived from a transplantable rat malignant fibrous histiocytoma.

A cis-diamminedichloroplatinum (CDDP)-selected cell line (MT-R10) was induced by continuous exposure of an in vitro passaged cell line (MT-P) established from a transplantable rat malignant fibrous histiocytoma (MFH-MT) to CDDP. MT-R10, capable of proliferating in the presence of 1.0 microgram CDDP/ml, was passaged in CDDP-free medium. The doubling time of MT-R10 at passage 10 (MT-R10/10) was almost the same as that of MT-P, being 22.3 and 25.5 h, respectively. The concentration of CDDP required for 50% inhibition of MT-R10/10 proliferation was two-fold higher than that of MT-P. MT-R10 consisted of round, epithelial-type cells arranged in compact sheets. Ultrastructurally, MT-R10 had numerous free ribosomes, some mitochondria, and other poorly developed cytoplasmic organelles suggesting its undifferentiated nature. MT-R10 showed no reaction for acid phosphatase or non-specific esterase. Tumors induced in syngeneic rats by inoculation with MT-R10 consisted of small, round, undifferentiated cells with scanty cytoplasm. They showed organoid and trabecular patterns, and were often arranged in compact sheets. The neoplastic cells showed no reaction for any of the histiocytic lysosomal and antigenic markers tested, but exhibited a strong reaction for alkaline phosphatase. Bone formation was often observed in the tumors. These observations suggest that CDDP-selected, undifferentiated cells may have osteogenic potential and may be one of the progenitor cells of MFH-MT.

[A case of signet-ring cell carcinoma of the urinary bladder].

Signet-ring cell carcinoma of the urinary bladder is a rare disease. A 78-year-old man was admitted to our hospital on March 18, 1988 with the complaints of microhematuria and pollakisuria. Cystoscopic examination revealed non-papillary tumor at the dome of the bladder. partial cystectomy was done under epidural anesthesia. Pathological findings revealed signet-ring cell carcinoma. The gastrointestinal, respiratory and genitourinary tracts were examined but no other tumor lesions could be found. The postoperative course was uneventful, and chemotherapy such as biological response modifier was administered. However, the patient died of heart failure on July 14, 1988. This is the sixtieth case reported in the literature.

[Treatment using CDDP and radiation therapy in treating bladder tumor].

Combination therapy consisting of cis-diamine-dichloroplatinum (CDDP) and radiotherapy was used to treat bladder cancer. The subjects were 20 patients with bladder cancer we experienced since September, 1986. They were between 33 and 83 years old and consisted of 13 males and 7 females. Among the histotypes and grades, there were 3 cases of G2, 7 of G2-G3 and 10 of G3 metastatic epithelial carcinoma. The classification of primary tumors was T2 in six cases, T3a in 12 cases and T4 in two cases. In 12 cases where the bladder was preserved, this therapy was performed after TUR biopsy or TUR, and the effects were evaluated after observation periods of 5-17 months. Side effects caused by CDDP were anorexia in 15 cases, nausea and vomiting in 10 cases, leukopenia in 10 cases and thrombocytopenia in four cases. Those caused by the radiotherapy were diarrhea in seven cases and skin disorders in six cases. All of these side effects were mild and the patients recovered spontaneously.

Relationship between mitogen receptors in peripheral blood lymphocytes and blastogenic responses to mitogen.

The relationship between the optimum concentration of mitogen which induces lymphocyte blastogenic response and the receptor occupancy by mitogen was investigated. The receptor occupancies which induced maximal blastogenic activity in equine, bovine and canine peripheral blood lymphocytes (PBL) were 31.1 per cent, 26.5 per cent and 38.4 per cent with phytohaemagglutinin-P, and 48.2 per cent, 17.9 per cent and 24.5 per cent with concanavalin A, respectively. The data clearly show that each animal species had its own optimum concentration of mitogen for stimulation of PBL. Optimum concentration for blastogenesis and number of binding sites of each mitogen had a good correlation with each other for all three species.

Chromosome analysis of canine transmissible sarcoma cells.

The chromosomal banding patterns of canine transmissible sarcoma (CTS) cells were analyzed and compared to those of normal canine cells with four banding techniques. In addition, N-myc and N-ras oncogenes on the chromosomes of the CTS cells were investigated by the in situ hybridization method. The modal chromosome number of the CTS cells was 58:17 metacentrics and 41 acrocentrics. Based on their G- and Q-banding patterns most of the chromosomes of the CTS cells were present in normal cells except for the second largest metacentric chromosomes. It was considered that the metacentric chromosomes of the CTS cells results from Robertsonian translocation of normal chromosomes. The long arm of the second largest metacentric element of the CTS cells was stained negatively by the Q-banding technique. The C-band was found on the long arm of the second largest metacentric element and three pairs of N-bands were recognized in the same region. It was suggested that this amplification of N-bands enhanced tumorigenic properties in the CTS. No N-myc or N-ras oncogenes were detected on chromosomes of the CTS cells by the in situ hybridization method.

Influence of cholesterol administration and aging on the development of pulmonary foam cells in F344 rats.

The influence of experimentally induced hyperlipidemia and aging on the development of pulmonary foam cells (PFCs) was examined in Fischer 344 rats. The male and female rats were administered orally with cholesterol at a dosage level of 1000 mg/kg/day for 30 days from 6 to 10 weeks or from 33 to 37 weeks of age. The control rats received the vehicle only in the same manner. Plasma levels of total cholesterol, phospholipid, triglyceride (TG), beta-lipoprotein (beta-LP) and calcium in both control and cholesterol-administered groups were higher at 37 weeks than at 10 weeks of age. Plasma beta-LP and TG levels in the treated groups were significantly higher or tended to be higher than those of the controls at 10 and 37 weeks of age. Males of the treated group showed the highest level of beta-LP at 37 weeks of age, positively correlated with the highest incidence of PFCs. PFCs developed singly or in a small cluster in peribronchial and subpleural regions. PFCs had an abundant cytoplasm filled with many fine vacuoles containing neutral lipid and cholesterol. PFCs stained with PAS and reacted immunohistochemically with both anti-rat monocytes/macrophages monoclonal antibody and anti-lysozyme antibody. Moderately swollen macrophages with a foamy appearance were detected in perivascular connective tissues of the lungs and they were considered to represent an initial stage of the development of PFCs. These observations suggest that hyperlipidemic conditions, particularly hyper beta-lipoproteinemia, resulting from cholesterol administration or aging may be involved in the development of PFCs in rats.

The banding patterns of normal canine chromosomes.

The chromosomal banding patterns of canine normal chromosome were analyzed by G-, Q-, C- and N-banding techniques. The chromosomes of canine normal lymphocytes consisted of 38 acrocentric autosome pairs and X-, Y-metacentric sex chromosomes. Their G- and Q-banding patterns corresponded to the those reported by Manolache et al. in 1976. C-bands of chromosomes were found on the centromeric regions of the 7th, 26th, 34th, 35th, 37th and 38th pairs of chromosomes. In addition, N-bands whose location had not been confirmed until now were recognized on the telomitic regions of the 7th, 9th and 22nd pairs of chromosomes.
