Effects of cyclazocine on the evoked-potentials at the trigeminal subnucleus caudalis, nucleus ventralis posteromedialis of the thalamus, somatosensory S1 area of the cerebral cortex and dorsal hippocampus following electrical stimulation of the tooth-pulp were studied using the rabbit. Pentazocine and morphine were used as reference drugs. Cyclazocine in doses of 0.5-1.0 mg/kg, i.v., significantly suppressed the evoked-potential recorded at the dorsal hippocampus but not those at the other sites. The hippocampal evoked-potential was significantly inhibited also by pentazocine (5.0-10 mg/kg, i.v.) and morphine (2.0-4.0 mg/kg, i.v.). The latter but not the former significantly depressed the evoked-potential at the S1 area of the cerebral cortex. The evoked-potential at the trigeminal subnucleus caudalis was not affected by these three drugs in the doses indicated. These results suggest that cyclazocine as well as pentazocine and morphine inhibit the electrically induced painful impulses at the hippocampus and/or the afferent pathways to the hippocampus from the trigeminal subnucleus caudalis such as the tooth-pulp-hypothalamo-hippocampal pathway.

Changes of the components of the myelinated fibre of lumbar spinal nerves and their relationships to the corresponding intervertebral foramina were studied on fifteen autopsied subjects over 65 years of age with a histo-morphometrical technique and compared with those of 14 younger aged cases as controls. Mixed spinal nerves were obtained at the outlet of each foraminal canal, fixed in 10% formalin, and embedded in paraffin. Sections for light microscopy were cut at 2.5 micra thick and stained with luxol-fast blue. A histo-morphometrical study was performed by actual measuring on enlarged photomicrographs (x 2,000). The size-frequency histograms of the myelinated fibre showed a bimodal distribution with two peaks at 1-3 micra (small fibre) and 7-10 micra (large fibre) in control group. In senile group, it was noteworthy that the peak of the large fibre was diminished or sometimes absent with marked intraneural fibrosis and an increase in small fibre density was observed. And these changes were more prominent in cases with severe disc degeneration.

Helix contents of sodium poly(L-glutamate) in aqueous NaCl solutions were estimated as functions of the degree of ionizalion, alpha. and the salt concentration by CD measurement. The helix content increases with increasing salt concentration but this helix stabilization decreases with decreasing alpha and at alpha<0.25 the helix conformation is destabilized by salt addition. The alpha dependence of the helix stabilization was qualitatively interpreted by Manning's theory in which electrostatic interactions between charges on alternatively arrayed coil and helix segments in a simplified polymer model were incorporated.
In this investigation Hürthle cell adenoma of the thyroid was observed by electron microscopy. The cytoplasm of the cells consisted chiefly of an accumulation of numerous irregularly shaped mitochondria. Very few other organellae were observed. In addition electron microscopy revealed various degrees of electron density in the matrix of the mitochondria. When 3 dimensional models of the mitochondria were constructed, they revealed the mitochondria to have 3 dimentional forms which sometimes looked as if they might be dividing into two or three parts. The authors felt that this indicated a process of vigorous division of mitochondria and an extension of their life span. However, it was not possible to identify the cause of this division of mitochondria.
With the aim of finding a prospective therapeutic compound with a promising potential for the treatment of urolithiasis, we evaluated the effectiveness of a new potent inhibitor of urease, N-(pivaloyl)glycinohydroxamic acid. The present study revealed that N-(pivaloyl)glycinohydroxamic acid effectively inhibited the alkalinization of urine and the stone formation in vitro and in vivo, due to its strong inhibitory potency against the ureolytic activity of intact Proteus mirabilis. The possibility of the clinical application of this compound in the prevention of struvite stone formation caused by infection of urea-splitting bacteria awaits evaluation of the safety of this compound.
Anti-inflammatory properties of a newly synthetized compound, 6-chloro-4-oximino-1-phenyl-1,2,3,4-tetrahydroquinoline (M-7074), have been investigated. Anti-edema activities of M-7074 were more potent than those of phenylbutazone in carrageenin, bradykinin and mustard edema tests in rats. M-7074 showed an inhibitory effect on adjuvant arthritis, especially on the secondary inflammatory lesions in rats. Inhibitory effect of M-7074 on cotton-pellet granuloma formation was all but equal to that of phenylbutazone in rats. M-7074 also showed inhibitory effects on ultraviolet erythema in guinea-pigs and increased vascular permeability in mice, moderate analgesic activity in rats and mice, and antipyretic activity in rats. Furthermore, inhibitory effects of M-7074 on prostaglandin biosynthesis in guinea-pig lung homogenate and arachidonic acid-induced aggregation of rabbit platelets were fairly equal to those of indomethacin. However, M-7074 showed no effect on humoral nor cellular immunity in mice. M-7074 possessed no ulcerogenic activity in rats and mice, and LD50 value of M-7074 was 8.01 g/kg, p.o. in mice. These data indicate that M-7074 is a novel anti-inflammatory agent with large margin of safety.

Significantly increased levels of plasma beta-thromboglobulin (beta-TG) (76.8 +/- 25.5 ng/ml, p less than 0.01) were observed in 24 patients with chronic renal failure (blood urea nitrogen (BUN) greater than 20 mg/100 ml), as compared with normal subjects (13.2 +/- 5.6 ng/ml). The increase in beta-TG was highly correlated with BUN (r = 0.651, p less than 0.01), creatinine (r = 0.778, p less than 0.01) and creatinine clearance ( t = -0.723, p less than 0.01). Plasma platelet factor 4 (PF4) and normal 5.0 +/- 2.0 ng/ml) also increased significantly to 8.5 +/- 3.4 ng/ml (p less than 0.01). However, statistical correlation between beta-TG and PF4 was not found in these patients. The reason is thought to be due to differences in molecular weight (PF4 8,000 MW; beta-TG 36,000 MW) and half-life time (PF4 30 min; beta-TG 100 min), and due to the difficulty in calculating statistically the correlation because of the narrow distribution of PF4 levels. The high levels of beta-TG (89.4 +/- 3.4 ng/ml) showed a further increase (109.4 +/- 5.8 ng/100 ml, p less than 0.01) after dialysis. This is thought to be due to hemoconcentration, because other blood factors such as RBC, WBC, platelets, fibrinogen, etc were elevated by about 20% during hemodialysis and because no adhesion of platelets to the cellulose membrane did occur. The increase in PF4 levels at 15 min (55.2 +/- 19.6 ng/ml, p less than 0.01) and 1 hr (23.7 +/- 8.4 ng/ml, p less than 0.01) of hemodialysis from the level before it (7.7 +/- 1.3 ng/ml) is thought to be caused the effect of heparin infusion. The change in PF4 was not accompanied by the change in beta-TG. During hemodialysis the decrease of other platelet functions such as adhesiveness, aggregation induced by ADP, collagen and PF3 remained unchanged.

Pharmacological properties of 2-[(5-chloro-2-methoxyphenyl)azo]-1H-imidazole (M6434) were investigated in vivo and in vitro experiments. M6434 showed a dose-dependent pressor effect in conscious rats at oral doses ranging from 1 to 5 mg/kg. The pressor effect of M6434 was 10 times as potent as those of phenylephrine and methoxamine. M6434 also showed a dose-dependent pressor effect in anesthetized dogs at i.v. doses ranging from 3 to 30 micrograms/kg. The effect of M6434 returned to control level 30 to 45 min after i.v. injection. M6434 caused a contraction of isolated guinea-pig aortic spiral strip at concentrations ranging from 10(-8) to 10(-4) mol/l. pD2 value of M6434 was larger than those of the other sympathomimetics, yet the maximal response of M6434 was 83.4% of the maximal response of noradrenaline. Pretreatment with phentolamine caused a parallel shift of the concentration-response curve of M6434 to the right. However, this effect of M6434 on an isolated aorta was not diminished even in reserpinized guinea-pigs. Therefore, the pressor effect of M6434 may be due to the direct adrenergic alpha-stimulation. M6434 is expected to be effective for the oral treatment of hypotensive disorders.
