[Hepatic artery embolization therapy for advanced hepatoma].

A total of 244 cases of hepatoma was subjected to hepatic artery embolization, and the 1-year, 2-year and 3-year cumulative survival rates were 45%, 27% and 12% respectively. The one year survival rate was far higher than the corresponding rate (7%) of intrahepatic arterial chemotherapy. The human livers which were autopsied or resected after the embolization were examined histologically. In 3 cases with main portal vein occlusion due to tumor thrombi, not only tumor tissue but also surrounding liver tissue were affected by extensive necrosis. But in 4 cases with patency of portal vein and severe liver cirrhosis, no necrotic region was detected in the liver parenchyma. Four of the seven patients who had been alive for more than 3 years after the therapy were accompanied with severe liver cirrhosis. It is suggested that liver cirrhosis is not so harmful factor in hepatic artery embolization therapy for advanced hepatoma.

Selenium efficiently depressed toxic side effect of cis-diamminedichloroplatinum.

Effect of selenium on toxic side effects of cis-diamminedichloroplatinum (Cisplatin; CDDP) which is a potent anticancer drug containing platinum was examined. Remarkable protective effects of selenium against lethal and renal toxicity of CDDP were observed when selenite was administered to mice once a day after single injection of lethal dose of CDDP. On the other hand antitumor activity of CDDP was hardly depressed by selenite.

Temporary spinalization reverses the inhibitory effect of cyclazocine on the nociceptive response of rabbit spinal dorsal horn lamina V-type neurons.

Cyclazocine (0.5 mg/kg i.v.) inhibited the activity of lamina V-type neurons of the rabbit spinal dorsal horn induced by intra-arterial injection of bradykinin. This inhibition was reversed during temporary spinalization by cold block. Thus, cyclazocine probably depresses nociceptive transmission in the spinal dorsal horn by facilitating descending inhibitory systems which originate from supraspinal structures.

A newly devised reliable method for evaluating analgesic potencies of drugs on trigeminal pain.

A pain-producing substance, bradykinin (0.63--1.25 ng in 0.5--1.0 microliter of distilled water), was applied onto the tooth pulp of the lower incisor of unrestrained rats through a cannula fixed on the surfaces of the incisors using a microsyringe. Such a microapplication of bradykinin induced biting response and other aversive behaviors. When the microapplications were repeated at intervals of 60 min, the biting response seemed most characteristic and reproducible. Therefore, the duration of biting response was used as a measure in studying the effects of drugs on trigeminal pain. If biting duration was inhibited 90% or more after drug administration the effect was considered analgesic. Carbamazepine and phenytoin, which are clinically employed for treating trigeminal neuralgia, and morphine, a narcotic analgesic, produced dose-dependent analgesic effects in this method and the corresponding ED50 values were 13.1 intraperitoneally (ip), 75.0 ip, and 3.00 subcutaneously (sc) mg/kg, respectively. On the other hand, pentobarbital (15 mg/kg, ip), diazepam (1.0 mg/kg, ip), and aspirin (150 mg/kg, ip) were not effective in suppressing the biting response. These results indicate that this newly devised method in the rat is reliable and feasible in evaluating pain related the trigeminal system.

Effect of L18-MDP(Ala), a synthetic derivative of muramyl dipeptide, on nonspecific resistance of mice to microbial infections.

By subcutaneous treatment with an aqueous solution of 6-O-stearoyl-N-acetylmuramyl-L-alanyl-D-isoglutamine [6-O-CH3-(CH2)16-CO-MurNAc-L-Ala-D-isoGln] [referred to here as L18-MDP(Ala)], an augmentation of the resistance of mice to Escherichia coli, Pseudomonas aeruginosa. Staphylococcus aureus, and Candida albicans infections was observed, but not to infections with Klebsiella pneumoniae and Listeria monocytogenes. Against E. coli infections, L18-MDP(Ala) was highly protective, irrespective of the administration route. Bacteremia occurring at an early phase of such infections was almost completely prevented by subcutaneous treatment 1 day before infection. Single or multiple doses were also effective against C. albicans infection. The phagocytosis of E. coli by mouse peritoneal polymorphonuclear cells was enhanced by treatment with the adjuvant, and the phagocytosis of K. pneumoniae was also enhanced, but only when the mice were treated either with rabbit normal serum or with a specific immune serum. The growth of the fungus in the kidneys was significantly inhibited, and growth was eliminated from the kidneys by treatment with the adjuvant once a day for 4 consecutive days, starting 1 day before infection. However, no growth suppression of L. monocytogenes in the livers or spleens of infected mice was observed when they were treated with a single dose of the adjuvant. This difference may be ascribed to the differences in the effector mechanisms of defense and to the different degree of augmentation of each defense mechanism by L18-MDP(Ala).

Elevation of colony stimulating factors in mouse serum after injection of PSK, an antitumor polysaccharide.

The level of colony stimulating factors (CSF) in mouse serum was elevated by a single intraperitoneal injection of PSK, a host-mediated antitumor protein-bound polysaccharide obtained from Basidiomycetes. The assay for CSF activity was performed by employing a semisolid methylcellulose culture method using mouse bone marrow cells, and the activity was estimated by an equation well matched with the dose response curve obtained in the CSF assay. A temporary increase of CSF activity within 10 h after the injection of an antitumor polysaccharide, Krestin (PSK) (250-1000 mg/kg) was followed by a fast decline in the activity, but no significant increases were detected in the cases of 62.5 and 125 mg/kg PSK injections. The CSF activity in PSK (500 mg/kg)-treated mouse serum was separated into two active fractions by DEAE ion-exchange chromatography, and both fractions were found to induce colonies comprised of cells with the properties of macrophage in regard to morphology, cytochemistry of non-specific esterase, phagocytic function and expression of Fc receptors on the cell surface. The elevation of the serum CSF level due to administration of so-called host-mediated antitumor agents might be one of the tumor-defense mechanisms in vivo.

Synthesis and secretion of prolactin in the decidualized human endometrium unassociated with pregnancy in vitro.

The effect of the decidualization induced by the administration of sex steroid hormones on the human endometrial PRL contents and the mechanism of synthesis and secretion of PRL in this decidualized endometrium of non-pregnant women were investigated. The PRL contents increased in proportion to the degree of decidualization. In vitro secretion of PRL in decidualized human endometrium was inhibited by the protein synthesis inhibitors, while TRH, dopamine, and bromocriptine did not have any effects on PRL secretion in incubation experiments. These data suggest that the production and secretion of PRL in the human endometrium have no relation to the presence of conceptus, and furthermore, they are regulated by mechanism different from the pituitary PRL.
