[Effects of antipyretics and anticancer drug on human cells at high temperature condition].

The effects of antifebriles and anticancer drug on human vascular endothelial cells (HVE) and several cultured human cells were investigated. The HVE were isolated from umbilical cord veins by enzyme treatment and cultured successively in aerated synthetic medium, RPMI-1640, with 20% preclostrum new born calf serum. The presence of factor VIII antigen in the HVE was determined by enzyme-labeled antibody method. Cell count and protein amount were examined at regular intervals. At 3 hour-expose, sulpyrine was more toxic to the cultured cells than aspirin at 37 degrees C. The cytotoxicity of sulpyrine was markedly enhanced at 40 degrees C than at 37 degrees C. However, there was no enhancement in the cytotoxicity of aspirin at 40 degrees C. Cultured HVE and normal human fetal lung (HAIN-55) cells at 37 degrees C were sensitive to sulpyrine, and their sensitivity of the cells to the drug were markedly enhanced when they were incubated at 41 degrees C. In contrast, sensitivity of malignant human cells (HeLa cells) to sulpyrine was not found at 37 degrees C, however sensitivity of the cells to the drug was manifested at 41 degrees C of incubation. There was no effect of 5-fluorouracil (FU) on the growth of HVE and HAIN-55 cells at 41 degrees C, while HeLa cells showed high susceptibility to FU at the same temperature. The results showed the possibility that normal human cells may be sensitive to antifebrile drugs but not to anticancer drug at ordinary and high temperature, whereas malignant human cell may be susceptible to both antifebrile drugs and anticancer drug at high temperature.

Magnesium action on the stability of fluorapatite.

Fluorapatites with various magnesium contents (Mg-FAp) were synthesized at pH 7.4 and 80 degrees C. The contraction of their c-axis dimensions observed in X-ray diffraction analysis, together with the results of chemical analyses, suggested that Mg2+ ions may substitute into the apatite crystals to a limited extent. The apatites became less well crystallized with increase in magnesium content. At any given magnesium-calcium feed-in molar ratio (Mg/Ca)f, the magnesium uptake by Mg-FAp was higher than that by the corresponding F- -free Mg-HAp. Therefore, the former became amorphous at lower (Mg/Ca)f than the latter. However, in terms of crystallographic considerations, the Mg-FAp synthesized at high (Mg/Ca)f revealed its apatitic features after pyrolysis at 1,000 degrees C, while Mg-HAp displayed the tricalcium phosphate X-ray pattern.

[Local treatment of locally recurrent breast cancers].

For the treatment of recurrent breast cancer, surgical treatment of the local lesion is only of limited significance, leaving a major role for systemic treatment such as chemotherapy and endocrine therapy. Therefore, a new method of local treatment other than surgical removal alone, or systemic treatment of these locally recurrent breast cancers is required. For the treatment of locally recurrent breast cancer, intra-arterial infusion chemotherapy was performed through both the internal mammary artery and subclavian artery or individually. Fourteen cases of locally recurrent breast cancer in our hospital were treated with this therapy. Continuous infusion of 5-fluorouracil and intermittent injections of mitomycin, adriamycin or 4'-epi-adriamycin were jointly or individually done into each artery. Regression of the locally recurrent mass was periodically measured from 4 to 6 weeks after infusion. The percentage of patients with PR was 35.7%. Intra-arterial infusion chemotherapy is a useful preliminary procedure for systemic treatment of local lymph node recurrence from breast cancer. A treatment involving local infusion of the non-specific immuno-activators, OK-432 and PSK, has also been performed for local recurrent lesions of breast cancers. We injected OK-432 and 5% PSK solution into patients with locally recurrent breast cancers and studied their suppressive effect on tumor proliferation. Macroscopically, PR was noted in 3 of 12 patients given OK-432. CR+PR were obtained in 9 of 12 patients given PSK. These results suggest that local injection of OK-432 and PSK solution is beneficial in the management of nodular or scattered-type locally recurrent tumors except for infiltrating type.

Distribution of erythrocyte in a model vessel exposed to inhomogeneous magnetic fields.

In order to investigate magnetic field effects on blood flow, changes in the flow of erythrocytes in a model branched vessel were observed in an inhomogeneous magnetic field. The magnetic field was applied perpendicular to the straight vessel before branching. When the suspension containing paramagnetic erythrocytes with high spin methemoglobin or deoxygenated hemoglobin flowed in the model vessel, the erythrocytes were attracted towards the stronger magnetic field (i.e. to the side branch) and an excess flow of erythrocytes to the side branch was detected. This excess flow of erythrocytes to the side branch was the highest at a hematocrit of about 5% for the suspension containing erythrocytes with high spin methemoglobin. In the case of mixed suspensions containing erythrocytes with high spin methemoglobin and oxygenated erythrocytes, the excess flow of erythrocytes to the side branch reached its maximum at the "partial hematocrit" for the paramagnetic erythrocyte of around 5% and remained nearly constant with a further increase of the "partial hematocrit." The effect of magnetic field decreased as the flow velocity increased. These results are explained with the paramagnetism of erythrocytes and with the assumption of a hydrodynamic interaction among erythrocytes which are pulled in the direction of the magnetic field. It is suggested that a strong inhomogeneous magnetic field is not totally negligible to the blood circulation.

[Intraarterial chemo-embolization with adriamycin containing albumin microspheres].

For local treatment of inflammatory breast cancer and recurrent breast cancers with liver metastasis, we applied intraarterial infusion chemoembolization therapy utilizing albumin microspheres containing adriamycin (ADM-ms), which was developed by Morimoto. In this study, there were 2 cases of inflammatory breast cancer and 3 cases of recurrent breast cancers with liver metastasis. Arterial canulation of primary breast cancer were made through both arteries, the internal mammary artery and lateral thoracic artery, which were feeding arteries of the breast. ADM-ms was injected into both arteries and its tumor effect was evaluated clinically and histologically. Remarkable reduction in tumor size was recognized. Histologic examination of resected specimens revealed extensive fibrosis in the center of the primary tumors. A few cancer cells, degenerated in various degrees, were found remaining in the periphery of the necrotic mass. Local side effects of the microspheres were skin disturbances such as pain, redness and blistering. ADM-ms was injected into the hepatic artery for the liver metastatic cases, and its antitumor effect was evaluated by CT scan or echography. Effective reduction in tumor size was only recognized in one case. No serious systemic side effects, not recognized in any case. Our report reviews the effectiveness of this therapy as a preliminary procedure for surgical treatment of inoperable inflammatory breast cancer.

Effects of an inhomogeneous magnetic field on flowing erythrocytes.

Effects of an inhomogeneous magnetic field on narrow erythrocyte streams in a wide and transparent laminar buffer flow were studied. The stream line of erythrocytes containing paramagnetic hemoglobin showed distinct displacement toward the stronger magnetic field. The displacement increased in the order, oxygenated erythrocytes (no displacement), erythrocytes containing cyanomethemoglobin, deoxygenated erythrocytes, erythrocytes containing methemoglobin in the high spin state; more precisely the displacement was proportional to the square of the paramagnetic moment of hemoglobin contained in the erythrocytes. In addition, the displacement was proportional to the product of the magnetic flux density and its gradient, and approximately proportional to the hematocrit of the flowing-erythrocyte suspension, and was much larger than that calculated for a single erythrocyte. These phenomena could be successfully interpreted by the interaction of paramagnetic erythrocytes with the inhomogeneous magnetic field, the resistance force (Stokes Law) from the bulk water, and the hydrodynamic interaction between erythrocytes.

Function of reticuloendothelial system on ethionine induced liver injury in mice.

Phagocytic activity as a function of the reticuloendothelial system (RES) has been studied in ethionine induced liver injury by using the carbon clearance test. Liver damage in male and female mice was induced by DL- and L-ethionine injections (1000 mg/kg/day, i.p.). In both female and male mice, a single dose or three injections of DL- or L-ethionine caused increases in liver/body weight ratio, A/G ratio, GOT and GPT levels, and BSP retention. There was the decrease of the total protein levels in the serum. The degree of liver injury was more severe after three injections of DL- and L-ethionine than after a single injection of them. After a single injection of ethionine, the L-isomer induced a slightly greater response than the racemic mixture, except for BSP retention. On the other hand, phagocytic activities by the carbon clearance test were increased after a single injection or three injections of DL- and L-ethionine. That is, the K value was increased in all ethionine treated mice except for females with three injections of DL-ethionine. The alpha value was increased after three injections in DL- and L-ethionine treated males and DL-ethionine treated females. In addition, the increase in carbon uptake by Kupffer cells can be seen by light microscopy after a single injection or three injections of DL- or L-ethionine. These findings indicate that ethionine injections induce the enhancement of RES phagocytosis, although the biochemical parameters indicating liver injury are changed severely. These results support the data indicating no correlation between the alteration of RES activity and the degree of liver injury.

A novel method for measuring the erythrocyte deformability, under oscillatory shear stress.

A novel apparatus for detecting a subtle decrease in erythrocyte deformability was developed. The oscillatory shape change of erythrocytes was monitored under an oscillatory shear stress (modulating the oscillation frequency in the range of 9-90 dyn/cm2). The ellipsoidal deformation of intact erythrocytes perfectly followed the oscillatory force of up to about 2 Hz, but the diamide-treated cells showed a phase difference of the oscillatory deformation at 1.7 Hz.
