Loss of genes on chromosome 22 in medullary thyroid carcinoma and pheochromocytoma.

Using polymorphic DNA markers, we compared the constitutional and tumor genotypes of patients with multiple endocrine neoplasia type 2A (MEN2 A). We found loss of constitutional heterozygosity at the D22S9 locus in one out of 9 medullary thyroid carcinomas (MTCs). No loss of heterozygosity was detected at 12 other loci in any of the MTCs tested. Loss of heterozygosity at D22S9 and/or D22S1 was also demonstrated in 2 out of 5 pheochromocytomas tested. These results suggest that loss or mutation of a gene on chromosome 22 may play an important role in tumorigenesis in MEN2A.

Mg2+-F-interaction during hydroxyapatite formation.

Two series of Mg-containing fluoridated hydroxyapatites with various fluoride contents were synthesized at 80 degrees C and pH 7.4 and compared with a corresponding Mg-free series. The Mg content of the apatites increased slightly with the degree of fluoridation despite the constant magnesium concentration in the feed solution. A small amount of magnesium significantly affected the crystallinity of synthetically fluoridated apatites. The patterns of crystallinity with the increase of fluoride content were unique in each series with different magnesium contents. These phenomena might have been caused by the Mg2+-F-interaction during the apatite formation.

Drift of an erythrocyte flow line due to the magnetic field.

Drifts of erythrocyte-flow lines due to inhomogeneous magnetic field in a laminar flow in a buffer solution are shown for the first time, and are interpreted as being due to the paramagnetism of hemoglobins included in the erythrocytes. The drifts were dependent on the hematocrit of the flowing erythrocyte suspension.

Conjugal transfer of hydrogen-oxidizing ability of Alcaligenes hydrogenophilus to Pseudomonas oxalaticus.

Conjugal transfer of hydrogen-oxidizing ability (Hox) of the hydrogen bacterium Alcaligenes hydrogenophilus was examined. Intraspecific cross of plasmid pHG21-a that encodes hydrogenases that mediate hydrogen oxidation was most frequent at 25 C; the optimal temperature for growth was 30 C. The plasmid could be transferred from A. hydrogenophilus to Pseudomonas oxalaticus OX1 and OX4, and the resulting strains gained the capacity for autotrophic growth with H2 and CO2. Plasmid pHG21-a was maintained in P. oxalaticus OX1 and OX4 as stably as in A. hydrogenophilus.

[A case of adult Still's disease with cardial symptoms of remittent high fever and arthralgia for more than six months].

A 46-year-old woman was admitted to our hospital because of fever of unknown origin. Her fever continued with a daily rise to around 38 degrees C to 40 degrees C for more than six months, occasionally accompanied by polyarthralgia, erythematous rash and cervical lymphadenopathy. The erythrocyte sedimentation rate was 75 mm per hour and CRP was over 6+. The white-cell count was 15,100 with 21 percent band forms and the alpha 2-globulin was 18.8 percent of total protein. The tests for autoantibodies were negative. Clinical course and laboratory findings in this case were most compatible with adult Still's disease. A radionucleotide bone scan with 99mTc showed a greatly increased uptake of the radionucleotide at the large joints of the whole body, and a gallium scan also revealed accumulation of the radionucleotide at the bone marrow of the sternum. These findings were recognized during high fever and disappeared when the body temperature returned to normal. These examinations might be useful for investigating the active site of inflammation and for studying the pathogenesis of adult Still's disease.

Crystallinity and solubility behavior of iron-containing fluoridated hydroxyapatites.

Iron-containing fluoridated hydroxyapatites with various fluoride contents were synthesized at 80 degrees C and pH 7.4 using FeCl2 X nH2O as a source of iron. The Fe2+ uptake of fluoridated apatites was independent of fluoride concentration in the solution. a-Axis dimensions of Fe-containing apatites decreased with the degree of fluoridation in addition to the decrease related to the substitution of Fe2+ ions. All Fe-containing fluoridated apatites were less well crystallized than Fe-free fluoridated apatites previously reported, although with increasing degree of fluoridation, the crystallinity behavior of the former apatites appeared analogous to that of the latter apatites. In contrast to this inhibited crystallinity behavior, the apparent solubility of Fe-containing fluoridated apatites decreased more than that of Fe-free fluoridated apatites at low fluoride content.

Chlorpromazine-sensitized photooxidation of squalene.

In connection with chlorpromazine-induced skin photosensitivity, chlorpromazine-sensitized photooxidation of squalene was studied in vitro. When squalene (in ethanolic solution or in an aqueous suspension) was irradiated with UV-A light in the presence of chlorpromazine, the yield of peroxidation products was greatly increased. Lines of evidence for the peroxidation were obtained by the iodometric determination and the thiobarbituric-acid method. Gas chromatography indicated that squalene was decomposed by the photosensitized reaction and the remaining amount was decreased exponentially with increasing fluence. The quenching effects of NaN3 and 2,5-dimethylfuran and the enhancing effect of D2O showed participation of singlet oxygen in the photosensitized reaction.

Studies on alterations in blood coagulative and fibrinolytic activities after single and multiple administrations of carbon tetrachloride in mice.

The time course of alterations in coagulative and fiblinolytic activities was studied in CCl4-induced liver disease in mice. Liver disease was induced by administration of 20% CCl4 in olive oil (p.o.). After single administration of CCl4, significant prolongation of r and k values and decrease in the ma value of thromboelastogram and apparent prolongation of PT and PTT were seen at 24 hr. Fibrinogen content decreased from 12 to 72 hr after single administration, while a mild but significant decrease in fibrinogen was observed after multiple administrations. The activity of factor XIII increased from 5 to 12 hr and then decreased from 24 to 168 hr after single administration. The activity of the hepaplastintest and Antithrombin III decreased apparently after single and multiple administrations. The plasminogen content and the activity of alpha 2-plasmin inhibitor decreased severely after single and multiple administrations. These results indicate that the coagulative and fibrinolytic activities were decreased to the most lowest value at 24 or 48 hr after single administration of CCl4, and the severe suppression of fibrinolysis and the mild decrease in coagulative activity were observed after multiple administrations of CCl4. The reason for the different effects between single and multiple administrations on coagulative and fibrinolytic systems was discussed.
