Antibiotic PS-5 is a new beta-lactam antibiotic isolated from fermentation broths of Streptomyces sp. strain A271. The strain was considered to be a new subspecies of Streptomyces cremeus and the name, Streptomyces cremeus subsp. auratilis, was proposed. Fermentative production, isolation and physico-chemical properties of PS-5 are described.

Intraperitoneal inoculation of either levamisole or 2 x 10(6) cells of concanavalin-A (Con-A)-bound L1210 leukemia vaccine produced no cured mice after subsequent ip inoculation of 10(3) live L1210 cells. Combined inocluation of levamisole and Con-A-bound vaccine produced about 20% cure incidence but no prolongation of life span of tumor-bearing mice after inoculation of live L1210 cells. Combined inoculation of levamisole and Con A-free vaccine did not induce detectable immune resistance in mice. These results suggest that levamisole enhanced host response to cell-bound Con-A associated with immunogenic potency of the vaccine. Levamisole given intravenously and orally was as effective as that inoculated intraperitoneally in enhancing the induction of resistance in mice by Con-A-bound vaccine. Dose of levamisole was very critical for this enhancement, being effective at 0.38 mg/kg but not either at 0.75 or 0.19 mg/kg. Furthermore, levamisole restored immune resistance induced by a larger inoculum of Con-A-bound vaccine cells (10(7)) and impaired by cyclophosphamide.

More than half of the patients with glomerulonephritis, nephrotic syndrome and renovascular hypertension showed obviously abnormal profiles of serum lipoproteins (Lps). The abnormal profiles returned to normal or near-normal when the disease was ameliorated or corrected surgically. A unique Lp profile (broad midband pattern, BMP) was observed in approximately 89% of uremic patients on hemodialysis therapy. The results of the current investigations indicated that the BMP could probably be formed by the accumulation of catabolic remnants of VLDL.

The fertilization rate of mouse gametes in a Krebs-Ringer bicarbonate medium was 60%. The rate fell to 0% when 250 micron-ZnCl2 was added, but MgCl2, CaCl2, MnCl2, FeCl2, and CuCl2 (each at 250 micron) had no effect. The inhibitory effect of Zn2+ was not evident if the spermatozoa or eggs were preincubated with Zn2+ or if previously capacitated spermatozoa were used. The Zn2+ is thought to act by interference with capacitation.

Antitumor activities of the cell-wall skeleton of Nocardia rubra and related bacterial fractions in autochthonous tumor-host system were tested on autografts of spontaneous mammary adenocarcinoma in SHN mice and of 3-methylcholanthrene-induced fibrosarcoma in ICR/JCL mice. The oil-attached cell-wall skeleton of N. rubra was the most effective in suppressing the autografts of the mammary adenocarcinoma but less of the fibrosarcoma, when the autografts were mixed with oil-attached preparation and implanted subcutaneously in the original host, while peptidoglycolipid of Mycobacterium tuberculosis Aoyama B was the most suppressive on the autografts of the fibrosarcoma but not on the mammary tumor autografts. The cell-wall skeleton of Mycobacterium bovis BCG slightly suppressed the autografts of the fibrosarcoma. Presensitization of tumor-bearing mice with the cell-wall skeleton of N. rubra resulted in a more marked suppression on autografts of both tumors than without the presensitization, but local destruction of these tumor autografts did not induce recognizable systemic immunity to the respective tumors. Intralesional injection of cell-wall skeleton of N. rubra showed prolongation of survival days of mice with fibrosarcoma.

Percutaneous transhepatic bile drainage was performed in 13 patients with obstructive jaundice, using a combination of the PTC technique and a Seldinger angiography catheter. In 11 cases, the outflow of bile through the catheter was satisfactory and complications were few. Since the risk of the procedure is low and it can be done without laparotomy, it is an ideal technic for biliary decompression before attempting to do a resection. Also, repeated cholangiography through a catheter which is left in place is helpful as a diagnostic aid before and after surgery.

Adjuvant and antitumor activities of CWS prepared from cells of mycobacteria, nocardia, and corynebacteria were examined. Oil-attached CWS of M. bovis BCG (BCG-CWS) stimulated the generation of cell-mediated cytotoxic effector cells in mice. Tumor growth was suppressed in mice inoculated intradermally with a mixture of oil-attached CWS and living tumor cells. Systemic and specific tumor immunity was demonstrated in mice in which tumor growth was suppressed. Tumor growth was also suppressed by oil-attached CWS of BCG or N. rubra in autochthonous autografts of spontaneous mammary adenocarcinoma and methylcholanthrene-induced fibrosarcoma in mice. The intravenous injection of oil-attached BCG-CWS prevents the appearance of lung cancer in rabbits by the instillation of chemical carcinogens. It was also shown that treatment with oil-attached BCG-CWS was able to elevate the immunologically depressed state of tumor-bearing mice to a normal level, as determined by a cell-mediated cytotoxicity assay that empolyed chromium release as the standard. Preliminary results suggest that oil-attached BCG-CWS is useful as an immunotherapeutic agent for both lung cancer and for malignant melanoma, leukemia, Hodgkin's disease, and other neoplastic diseases and that this agent operates without any significant complications.
