Identification of lactate dehydrogenase-X translated in vitro from mouse testicular poly A-containing mRNA.

1. The LDH-X polypeptide was specifically immunoprecipitated from the cell-free translation products of poly A-containing mRNA from mouse testes, and it represents 1-2% of the total proteins synethesized in vitro. 2. The in-vitro synthesized LDH-X polypeptide appears to have the same mol. wt of 36,000 as mouse authentic LDH-X and, thus, any presequence of LDH-X must be very short, if present at all. 3. The LDH-X was not found in the mouse liver mRNA translation products immunoprecipitated by anti-LDH-X antibodies.

Chemo-immunotherapy of methylchoranthrene-induced fibrosarcoma by concanavalin A-bound tumor vaccine, levamisole and mitomycin C.

Concanavalin A (ConA)-bound-tumor cell vaccine of methylchoranthrene-induced fibrosarcoma (Meth 1) induced tumor-specific immunoprophylactic and immunotherapeutic response against an inoculum of live Meth 1 cells in histocompatible animals. ConA-free Meth 1 vaccine induced much less response under the identical experimental conditions. Immunotherapeutic potency of ConA-bound Meth 1 vaccine was enhanced by levamisole, and 37% of the animals inoculated with 10(4) live Meth 1 cells at day 0 were cured when they were administered 10(6) cells of ConA-bound Meth 1 vaccine at days 1 and 8 and 0.63 mg/kg levamisole at days 1,2 and 3. Delayed administration of levamisole at day 8,9 and 10 was less effective than the earlier administration, but still produced a 17% cure of Meth 1-bearing animals when combined with ConA-bound Meth 1 vaccine. Immunotherapeutic response under these regimens was further enhanced by mitomycin C, and approximately 60% of the animals inoculated with 10(5) Meth 1 cells were cured when three agents were administered at the defined intervals. These results suggest the feasibility of the regimen in which the therapeutic response induced by immunotherapeutic agents is further enhanced by the selected chemotherapeutic agents.

Capacitation inducing activity of serum albumin in fertilization of mouse ova in vitro.

The role of serum albumin in in vitro fertilization of mouse ova was studied relating to the fertilization inhibitory activity of Zn2+. The time required for mouse sperm capacitation was dependent on the concentration of serum albumin which was added to the medium. The majority of Zn2+ added to the medium was shown to be bound to serum albumin (95% out of 200 microM) by means of equilibrium dialysis. Furthermore, it was suggested that the serum albumin was necessary for the initiation of the sperm capacitation process by the periodic observation of sperm penetration through zona pellucida.

The action of 7-N-(p-hydroxyphenyl)mitomycin C [M-83] in suppressing murine immune response.

The immuno-suppressive activities of 7-N-(p-hydroxyphenyl)mitomycin C [M-83], a derivative of mitomycin C (MMC), which has better antitumor activity against murine tumors, was investigated in mice. A single injection of M-83 induced an apparent but relatively transient reduction in both plaque-forming cell production and delayed type hypersensitivity reaction to sheep red blood cells. The suppressive effect of M-83 was significantly milder than that of MMC at the same effective and molar equivalent doses. Moreover, the recovery from immunosuppression induced by M-83 was also relatively rapid as compared with MMC. The inhibiting activity of M-83 against the blastogenic response of spleen cells was milder in vivo, but was more significantly cytotoxic in vitro than that of MMC.

Studies on the OA-6129 group of antibiotics, new carbapenem compounds. II. In vitro evaluation.

The OA-6129 group of antibiotics, new carbapenem compounds, had relatively potent antimicrobial activities against Gram-positive and Gram-negative bacteria. Synergism of compound OA-6129A in combination with conventional beta-lactam antibiotics was observed in antimicrobial activity against beta-lactamase producers such as Proteus vulgaris GN 76 and Citrobacter freundii GN 346. The new carbapenem compounds were slightly superior to PS-5 in stability to kidney homogenates of various animal species.

Zinc effects on mouse spermatozoa and in-vitro fertilization.

The fertilizing ability of mouse epididymal and capacitated spermatozoa was tested by mixing with normal or zona-free ova. In the presence of Zn2+, the epididymal spermatozoa failed not only to penetrate the zona pellucida but also to fuse with zone-free ova, while no effect was observed on capacitated spermatozoa. Zn2+ also inhibited fertilization with spermatozoa preincubated in Ca2+-free medium. However, shortly after the addition of Ca2+ to the preincubated spermatozoa, Zn2+ lost its fertilization inhibitory activity.

PS-6 and PS-7, new beta-lactam antibiotics. Isolation, physicochemical properties and structures.

Antibiotics PS-6 and PS-7 which are shown to be 5R,6R-3-(2-acetamido)ethylthio-6-isopropyl-7-oxo-1-azabicyclo[3.2.0]hept-2-ene- 2-carboxylic acid and 5R,6R-3-(E)-(2-acetamido) vinylthio-6-ethyl-7-oxo-1-azabicyclo[3.2.0]hept-2-ene-2-carboxylic acid respectively, are new beta-lactam compounds isolated from fermentation broths of Streptomyces cremeus subsp. auratilis A271, S. fulvoviridis A933, S. olivaceus ATCC 31126 and S. flavogriseus NRRL 8139. Fermentation, isolation, physicochemical properties and structures of antibiotics PS-6 and PS-7 are described.
