A case of multiple infantile fibromatosis of unclassified type.

We report an unusual case of fibromatous disease in infancy, diagnosed as a multiple infantile fibromatosis of unclassified type. Histologically, the tumors are composed of a mixture of fibrous component and mature adipose tissue. The clinical manifestations resemble those of infantile myofibromatosis. The symptoms, including size and number of tumors, have not been progressive for one year.

Methylsuccinate and mesaconate in urine of patients treated with sodium benzoate.

Two unusual peaks were found on gas chromatography of urine from four hyperammonemic patients treated with sodium benzoate. These peaks were identified by gas chromatography/mass spectrometric analyses as methylsuccinate and mesaconate. Of the two unusual substances, methylsuccinate was found to be a contaminant of sodium benzoate administered for the treatment of hyperammonemia. However, mesaconate was not a contaminant of sodium benzoate, though it could be detected in all urine samples from hyperammonemic patients treated with sodium benzoate. Mesaconate can be produced from methylsuccinate in vivo. Considering that mesaconate is an inhibitor of fumarase, the toxic effects of sodium benzoate may be attributable to the mesaconate. It is recommended that methylsuccinate-free sodium benzoate should be used for the treatment of hyperammonemia.

[Observation with SEM on the displacement of mucosal surface in process of denture fabrication].

The process of denture fabrication entails a series of steps that begin with impression-taking. Dimensional accuracy and displacement of surface repeatability affect results in these individual steps. Although changes in individual materials have already been measured in the past, there has been almost no measurement up to now of denture fabrication up to the denture base, using the mucous membrane as the standard and then measuring changes in the displacement of surface repeatability for each step. Noticing this lack, the authors used a Scanning Electron Microscope (SEM), presently the only non-contact measuring instrument that can be used, and measured changes at the different stages including the mucous membrane, the impression materials, the plaster and the resin. A drying process is required for the mucous membrane, and such a process can cause changes in the mucous membrane which is the basis of measurement. For this reason, the authors used Araldite, a product from Ciba-Geigy AG, as a replica material and made replicas for all the test materials, using the replicas for measurement. Results were as follows: 1. Araldite was used as the replica material, thus making it possible with the SEM to observe changes in the displacement of surface repeatability at the individual stages. 2. Major changes in the accuracy of surface were observed during impression-taking and in the period of moving from the impression material to the plaster. 3. During high-power observation with the SEM, the fine bumps on the surface of the plaster and resin disappeared and the surfaces became smooth. This is believed to be the result of the separating agent and heat.

[Clinical effects of cefodizime against infectious diseases in obstetrics and gynecology].

Clinical studies on cefodizime (CDZM) were performed in patients of obstetrics and gynecology. CDZM was given to patients via drip infusion at a daily dose of 2-4 g, and the results obtained are summarized as follows: 1. A total of 10 cases included 5 cases of intrauterine infections (3 cases of puerperal endometritis, 1 endometritis and 1 puerperal fever), 2 intrapelvic infections (1 pelvic dead space infection and 1 parametritis), and 3 Bartholin's abscess. Clinical efficacies were excellent in 3 cases, good in 6 and poor in 1. The efficacy rate was 90% (9/10). 2. Bacteriological efficacies were as follows: eradicated in 3 cases, replaced in 2, decreased in 1, persisted in 1 and unknown in 3. 3. Neither subjective and objective adverse reactions nor abnormal changes in laboratory test values were observed.

Binding of the new calcium entry blocker nilvadipine to rat aortic and guinea pig left ventricular membranes.

The binding of 3H-nilvadipine to the vascular (rat aortic) and cardiac (guinea pig left ventricular) microsomes was reversible and saturable. The results of the competitive binding experiments showed that nilvadipine had a higher affinity (Kd: 1.5 +/- 0.2 nmol/l) for the vascular binding sites than did nifedipine (Kd: 10.7 +/- 0.8 nmol/l) and nicardipine (Kd: 6.9 +/- 2.0 nmol/l), while the affinity of nilvadipine for the cardiac binding sites (Kd: 3.8 +/- 0.4 nmol/l) was similar to that of nifedipine (Kd: 3.3 +/- 0.1 nmol/l) and nicardipine (Kd: 4.2 +/- 0.3 nmol/l). The time courses of dissociation of the unlabeled dihydropyridines from the vascular binding sites were investigated using 3H-nitrendipine. Nilvadipine and nicardipine dissociated more slowly from the binding sites than did nifedipine. These results suggest that the high potency, selectivity for the vascular smooth muscle and long duration of action of nilvadipine may be related to its binding properties.

Protective effect of glutathione against oxygen-induced growth inhibition of human diploid fibroblasts.

The reduced glutathione level in human diploid fibroblasts was increased by the addition of N-acetylcysteine or reduced glutathione ethylester into the culture medium, while it was decreased by the addition of L-buthionine-(R,S)-sulfoximine or diethyl maleate. The hyperbaric oxygen-induced reduction in colony-forming ability was prevented in the N-acetylcysteine- or reduced glutathione ethylester-treated cells, and enhanced in the L-buthionine-(R,S)-sulfoximine- or diethyl maleate-treated cells. The extent of the growth inhibition is well correlated with the cellular glutathione level. It is deduced that glutathione is an important safeguard against the oxygen-induced growth inhibition of human diploid cells.

Separation of the stereoisomers of an allenic E-type prostaglandin.

Enprostil (I) is a synthetic dehydro-prostaglandin E2 containing a chiral allene moiety which is unresolved relative to the four remaining chiral centers. The relative configuration of the four remaining chiral centers is consistent with that of the naturally occurring E series of prostaglandins. Thus, enprostil exists as enantiomeric pairs of two allenic epimers. An analytical procedure has been developed that separates the four optical isomers present in enprostil. This procedure involves, first, the acid-catalyzed dehydration of enprostil to its corresponding prostaglandin A analogue followed by derivatization with beta-naphthylsulfonyl-L-prolyl chloride. The resulting diastereomeric sulfonate esters are separated on an achiral silica gel high-performance liquid chromatographic column. This procedure has been applied to the analysis of both enprostil drug substance and enprostil formulated in a propylene carbonate solution from soft elastic gelatin capsules. An efficient procedure for the recovery of enprostil from the solution formulation is also described.

A comment on the evaluation of equilibrium constants for alpha-tocopherol interactions with fatty acids by absorbance in the ultraviolet region.

This paper comments on the evaluation of Erin and co-workers (Biochim. Biophys. Acta 774 (1984) 96-102) of equilibrium constants for alpha-tocopherol interactions with fatty acids on the basis of the changes of absorbance in a 200 nm ultraviolet region. It is concluded that the ultraviolet method is inadequate because it is affected by absorption in that region of the solvent, ethanol and fatty acids which they used.

Membrane-stabilizing effect of vitamin E: effect of alpha-tocopherol and its model compounds on fluidity of lecithin liposomes.

The effects of vitamin E (alpha-tocopherol) and its model compounds on the fluidity of liposomes composed of dipalmitoylphosphatidylcholin (DPPC) and fatty acids were investigated by the measurement of the fluorescent polarization (P) using 1,6-diphenyl-1,3,5-hexatriene (DPH) as a plobe. Although all tocopherols decreased the fluidity of liposomes which was perturbed by the inclusion of an unsaturated fatty acid having more than one double bond, alpha-tocopherol was more effective than the others. The fluidity in arachidonic acid-containing liposomes was decreased most in the presence of alpha-tocopherol and was decreased considerably by the inclusion of model compounds having a side chain at least one isoprene unit or a long straight chain instead of isoprenoid side chain. However, the chromanol with methyl group instead of the above side chain, and phytol, having no chromanol moiety, had no effect. These results show that a structural requirement for a membrane stabilization is to be either the chromanol moiety with methyl groups born on its aromatic ring or a side chain of appropriate length; an isoprenoid side chain of full length or one containing 4'a- and 8'a-methyl groups is not necessarily needed.

Cytotoxicities of a linoleic acid hydroperoxide and its related aliphatic aldehydes toward cultured human umbilical vein endothelial cells.

An assay method for the quantification of the cytotoxicities of various agents toward cultured human endothelial cells was developed using Earle's solution as an incubation medium. By this method, the cytotoxicities of a linoleic acid hydroperoxide (LOOH) and its related aliphatic aldehydes toward human umbilical vein endothelial cells were investigated. Saturated aldehydes, pentanal, hexanal and 9-oxononanoic acid, are nontoxic; alpha, beta-unsaturated aldehydes, 2-hexenal, 2-heptenal, 2-octenal and 2-nonenal, are toxic only at high concentrations; LOOH and alpha, beta-unsaturated aldehydes with a hydroxy group or an additional double bond, 4-hydroxy-2-nonenal, 2,4-nonadienal and 2,4-decadienal, are highly toxic. In particular, 2,4-decadienal, whose 50% lethal concentration is 9 microM, is the most injurious. The cytotoxicities of LOOH and its related aldehydes were found to be much reduced in growth medium containing serum, growth factors, heparin, amino acids and vitamins.

Effects of serum from human subjects of various ages on proliferation of human lung and skin fibroblasts.

We carried out a study to determine whether serum from old human subjects inhibited cell proliferation. The results showed that serum from old subjects of either sex did not greatly inhibit the proliferation of human fetal lung fibroblast TIG-1 cells, even when serum from subjects in their 80s was used. The same results were obtained when the effects of serum on cell proliferation were examined up to a serum concentration of 50%. It was also found that serum from old subjects did not inhibit proliferation of human skin fibroblasts from a young adult to any greater degree than serum from young adult subjects, and that serum from young adult subjects did not stimulate proliferation of skin fibroblasts from an elderly donor to any greater degree than serum from old subjects.

Relationships between the cellular glutathione level and in vitro life span of human diploid fibroblasts.

In order to examine the role of cellular glutathione (GSH) in the in vitro aging of human diploid fibroblasts, we studied the effects of manipulated cellular GSH levels on their in vitro life span. An increase in cellular GSH level was produced by the addition of N-acetylcysteine (NAC), a carrier of cysteine across cell membranes, into the culture medium, while a decrease in GSH level was produced by the addition of L-buthionine-(R,S)-sulfoximine (BSO), a specific inhibitor of GSH synthetase. When the cells were serially subcultivated in a medium containing NAC or BSO, their life spans were markedly extended or shortened, respectively, in comparison to the life span of cells grown in a control medium. These results suggest that the cellular GSH level is a determinant of the in vitro life span of human diploid cells.

Pancreatic secretory trypsin inhibitor as a marker for early detection of rejection in canine pancreas allotransplantation.

Serum pancreatic secretory trypsin inhibitor (PSTI)* was measured in the course of canine segmental pancreas allotransplantation without immunosuppression. Serum PSTI concentrations showed two distinct elevations: the first elevation was on the first day and the onset of the second elevation was the sixth day after operation. The first postoperative elevation of the serum PSTI level is thought to be related to the operative procedures, because the first elevation was observed after both autotransplantation and allotransplantation, and biopsies of the autograft and allograft at the first day after the operations showed nonspecific neutrophilic infiltration and no perivascular lymphoid infiltrates. The second postoperative elevation of the serum PSTI level is thought to reflect a rejection process because this elevation was not seen after autotransplantation, and biopsies of allografts at the sixth day after the operation showed typical perivascular lymphoid infiltrates and cellular rejection of the exocrine tissue. In addition, the onset of the second elevation of serum PSTI level preceded by about three days the onset of the elevation of blood sugar at the ninth day after the operation. The results suggest that serum PSTI can be used as a marker for diagnosis of early pancreatic allograft rejection.

Interaction of vitamin E and its model compounds with unsaturated fatty acids in homogeneous solution.

Either alpha-tocopherol (vitamin E) or one of its model compounds having side chains of different length at the 2-position of alpha-tocopherol, forms complexes with an unsaturated fatty acid in methanol. For complex formation, the isoprenoid side chain and hydroxy group of alpha-tocopherol are unessential and, rather, the methyl groups attached to the aromatic ring of the chromanol moiety seems to be responsible. For better interaction, more than three methylene-interrupted Z double bonds of a fatty acid are necessary. These findings are incompatible with the hypothesis of Diplock and Lucy on the interaction of vitamin E with each polyunsaturated fatty acid.
