A new human male diploid cell strain, TIG-7: its age-related changes and comparison with a matched female TIG-1 cell strain.

A new human diploid cell strain, TIG-7, which has the male karyotype, was established and characterized. Isozyme and histocompatibility typing of the cell strain was performed. The average in vitro life span of the cells is 73 population doublings. Changes in cell volume, doubling time, saturation density, the efficiency of cell attachment, plating efficiency, and relative DNA content were examined during in vitro cellular aging. Hydrocortisone slightly prolongs the life span of the cell strain when the hormone is administered to the cultures during middle passages. The age-related changes in the parameters of TIG-7 are not appreciably different from those of the previously established TIG-1 cell strain. These results show that this cell strain is useful for research on cellular aging; further profit is anticipated from research using a combination of these two sexually different cell strains.

Characteristics of the vasorelaxing action of (3E)-4-ethyl-2-hydroximino-5-nitro-3-hexamide FK409, a new vasodilator isolated from microbial sources, in isolated rabbit arteries.

We examined the vasoinhibitory effect of (3E)-4-ethyl-2-hydroximino-5-nitro-3-hexamide FK409, a new vasodilator, on contractile responses in isolated rabbit arteries. FK409 (10(-8)-10(-5) M) inhibited contractile responses to norepinephrine (NE), histamine (His), and 5-hydroxytryptamine (5-HT) in rabbit aorta. The pattern of inhibition by FK409 was not competitive. The inhibitory effect of FK409 on the 5-HT response was much greater than that of nitroglycerin (NG). A high concentration of FK409 (10(-5) M) was necessary to inhibit the response to KCl (10-70 mM). The effect of combined treatment with FK409 (10(-5) M) and a subthreshold concentration of nifedipine (10(-9) M) on the KCl response was much greater than a single treatment with either agent. In addition, 3 x 10(-6) M D600, but not FK409 (10(-6) or 10(-5) M), inhibited the increase in the rate of 45Ca influx stimulated by a 40-mM KCl substituted solution. In a Ca2(+)-free medium containing EGTA and nifedipine, FK409 (10(-9)-10(-5) M) inhibited phasic responses to NE, His, and 5-HT, and subsequent sustained responses owing to addition of Ca2+. The response to caffeine in rabbit iliac arteries incubated in Ca2(+)-free medium was also inhibited by FK409 (10(-6) and 10(-5) M). In rabbit aorta precontracted with NE (10(-5) M) and partially inhibited by prior exposure to NG (10(-5) M), the relaxing effect of FK409 was slightly attenuated. Pretreatment of tissues with FK409 (10(-6) M) inhibited the relaxing action of NG much more than prior NG inhibited the relaxing action of FK409. Methylene blue (10(-5) M), but not hemoglobin (10(-6) M), inhibited the relaxing action of FK409, whereas M&B 22,948 (3 x 10(-4) M) potentiated it. FK409 caused a relaxation of precontracted aorta without endothelium that was inhibited by methylene blue. In rabbit aorta precontracted with NE, FK409 (10(-6) M) increased cyclic GMP but not cyclic AMP content. FK409 (10(-5) M) had no effect on the NE-mediated increase in tissue inositol monophosphate (IP). These results suggest that FK409 inhibits the responses attributed to both intracellular Ca2+ release and Ca2+ influx through receptor-operated channels. The inhibitory effect of FK409 on both the KCl contractile response and KCl-stimulated 45Ca influx appears to be different from that of nifedipine or D600. Furthermore, the inhibitory action of FK409 may be partially mediated by cyclic GMP.

Higher serum retinol-binding protein concentration in Indonesian neonates in Surabaya than Japanese neonates in Kobe.

The concentrations of serum retinol-binding protein (RBP), prealbumin (PA) and vitamin A of Indonesian neonates were compared with those of Japanese neonates. The mean serum concentrations of both PA and vitamin A did not differ significantly in Indonesian and Japanese neonates. Indonesian neonates had significantly higher serum RBP (2.75 +/- 0.87 mg/dl, mean +/- SD) than Japanese neonates (1.65 +/- 0.55 mg/dl, p less than 0.01). The molar ratio of vitamin A:RBP in Indonesian infants was significantly lower than in Japanese infants (p less than 0.02), and this in turn is indicative of increased concentrations of retinol-free RBP in the plasma of Indonesian neonates. The results suggest that RBP metabolism in Indonesian neonates differs from that in Japanese neonates.

Exon skipping during splicing of dystrophin mRNA precursor due to an intraexon deletion in the dystrophin gene of Duchenne muscular dystrophy kobe.

Recent molecular studies have shown that in a patient with Duchenne muscular dystrophy (DMD) Kobe, the size of exon 19 of the dystrophin gene was reduced to 36 bp due to the deletion of 52 bp out of 88 bp of the exon. The consensus sequences at the 5' and 3' splice sites of exon 19 were unaltered (Matsuo, M., et al. 1990. Biochem. Biophys. Res. Commun. 170:963-967). To further elucidate the molecular nature of the defect, we examined the primary structure of cytoplasmic dystrophin mRNA of the DMD Kobe patient across the junctions of exons 18, 19, and 20 by gel electrophoresis and sequencing of polymerase chain reaction-amplified cDNA. The mRNA coding for dystrophin was reverse transcribed using random primers, and the cDNA was then enzymatically amplified in vitro. The targeted fragment was smaller than expected from the genomic DNA analysis. By sequencing of the amplified product, we found that exon 18 was joined directly to exon 20, so that exon 19 was completely absent, suggesting that this exon was skipped during processing of the dystrophin mRNA precursor. All other bases in the amplified product were unaltered. Therefore, the data strongly suggest that the internal exon deletion generates an abnormally spliced mRNA in which the sequence of exon 18 is joined to the sequence of exon 20. We propose that the deletion is responsible for abnormal processing of the DMD Kobe allele. This finding has important implications regarding the determinants of a functional splice site.

Effect of dexamethasone on pulmonary surfactant metabolism in hyperoxia-treated rat lungs.

We have examined the effect of dexamethasone on the metabolism of pulmonary surfactant in normal and hyperoxia-treated rats. The relative abundance of the surfactant-specific apoprotein A (SP-A) mRNA in lung tissues and the contents of disaturated phosphatidylcholine (DSPC) and SP-A were measured in bronchoalveolar lavage fluids and in lung tissues in 4-wk-old rats exposed to room air or greater than 90% oxygen for 7 d with or without simultaneous treatment with dexamethasone (0.5 mg/kg body wt for 7 d). The relative abundance of the SP-A mRNA was marginally increased by hyperoxia (1.3-fold over controls). Dexamethasone increased the relative abundance of the SP-A mRNA to a level comparable to that with hyperoxia treatment (1.5-fold over controls). In lavage fluids, the contents of DSPC and SP-A were increased by 4- and 6-fold over controls by hyperoxia, respectively, but they were increased only by 2-fold by dexamethasone. In lung tissues, the contents of DSPC and SP-A were increased by 3- and 2-fold over controls by hyperoxia, respectively. These values in lung tissues in the air-exposed rats were not significantly increased by dexamethasone. In hyperoxia-treated rats, dexamethasone did not significantly affect the relative abundance of the SP-A mRNA level and the contents of DSPC and SP-A in lavage fluids and lung tissues. These results indicate that mechanisms other than increased synthesis of SP-A are involved in hyperoxia-induced SP-A accumulation and that dexamethasone does not affect the abnormal accumulation of pulmonary surfactant induced by hyperoxia.

Anticoagulant therapy in a congenital antithrombin III (ATIII)-deficient patient with chronic renal failure undergoing regular hemodialysis.

We describe a 43-year-old male patient with congenital antithrombin III deficiency requiring hemodialysis due to extension of venous thrombus from recurrent deep vein thrombosis. During dialysis with adequate heparinization, the patient often revealed clot formation in the extracorporeal circuit resulting in unexpected discontinuation of dialysis. When either a combination of antithrombin III concentrate + heparin or the newly developed synthetic antithrombin preparation, MD805, was administered during dialysis, he could be uneventfully dialyzed with either of the two regimens.

Increased MR signal intensity due to cervical myelopathy. Analysis of 29 surgical cases.

The cases of 29 patients with cervical myelopathy, who had been treated by anterior spine fusion, were reviewed. The relationship between pre- and postoperative magnetic resonance (MR) images was investigated with special reference to increased signal intensity in the spinal cord on the T2-weighted images and the relevance of this finding to clinical conditions. Preoperatively, there were areas of increased signal intensity in 12 patients whereas there were no areas of increased signal intensity in the other 17. The lesions were not clearly demonstrated on T1-weighted images. The pre- and postoperative clinical condition of the patients whose preoperative MR images showed areas of increased signal intensity in the spinal cord on T2-weighted images was worse than that of the patients who did not have areas of increased signal intensity. Of the 12 patients with regions of increased signal intensity preoperatively, five showed decreased signal intensity postoperatively compared to the preoperative levels and seven had no change. The postoperative recovery of the five patients who showed decreased signal intensity postoperatively was better than that of the seven patients who exhibited no change. The areas of increased MR signal in the spinal cord might be due to edema, cord gliosis, demyelination, or microcavities.

[Clinical study of intramuscular imipenem/cilastatin sodium in the field of obstetrics and gynecology].

We evaluated the clinical efficacy and safety of intramuscular (as a new route of administration) imipenem/cilastatin sodium (IPM/CS) in patients with intrauterine infections which are typical in the field of obstetrics and gynecology. The obtained results are summarized as follows. 1. Twenty-seven patients were treated with IPM/CS, 250 mg/250 mg b.i.d. (3 patients), 500 mg/500 mg b.i.d. (22) and other dosages (a change in dosing regimen, 2). The duration of treatment ranged from 3 to 11 days and the total dosage during an entire course of treatment varied from 1.5 to 9.0 g. The drug was suspended in a lidocaine solution and administered in the gluteal muscle of the patients. 2. Clinical efficacies were excellent in 7 patients (26%), good in 19 (70%) and poor in 1 (4%) and the overall efficacy rate was 96.3%. All of the 8 patients who had not previously showed improvements with treatment by other antibiotics responded well to this drug. 3. Bacteriologically, the clinical efficacy rate was 95.8% (23/24) and the eradication rate was 76.2% (16/21). 4. No adverse effects due to the drug were observed. As abnormal laboratory test results, transient elevations of GOT and GPT were noted in one patient.

[Fibrin monomer assay].

Quantitative assay for fibrin monomer was done by use of a chromogenic substrate (S-2390, Coa set fibrin monomer). Samples from DIC prone patients with the underlying disease were assayed and classified into four groups. The pre DIC group showed higher FM values than the control with no laboratory coagulation abnormality, although the FDP . D-dimer showed no significant rise. FM assay is a useful marker for the detection of early coagulopathy in DIC. Administration of the AT III concentrate in the case of low level of plasma ATIII, thrombin . antithrombin complex I (TAT) caused a significant transient rise. The clinical course of DIC by TAT is often affected by the fluctuation of ATIII level in plasma, the usefulness of FM is that it reflects the real thrombin generation in DIC.

Biomarkers↗

[Thrombin.antithrombin III complex].

Increase of TAT is reflected by the generation of thrombin in hypercoagulable state. TAT might increase in DIC characterized by the formation of disseminated micro-thrombosis. DIC was classified into three groups according to the results of screening tests (FDP, platelet count, fibrinogen, prothrombin time). TAT values significantly increased in the stage of pre-DIC compared with the control group consisting of DIC prone underlying disease. Pre-DIC was easily detected by an increase of TAT during the clinical course. Management of high TAT began with the use of an anticoagulant such as heparin under the condition of sufficient ATIII level. The lowering effect of TAT was easily obtained by the anticoagulant. In ATIII-deficient DIC, the high TAT reduced with the substitution of ATIII concentrate, though a transient increase of TAT was found during the administration of ATIII. To reduce the high TAT under the deficient state of ATIII, MD805, a synthetic thrombin inhibitor, was introduced to avoid further consumption of ATIII. The TAT was decreased by the use of MD805 without administration of ATIII. MD805 could be used as an effective anticoagulant in high TAT due to DIC under an ATIII-deficient state. Although the TAT improved with an adequate anticoagulation in DIC, spontaneous bleeding sometimes appeared as a complication associated with the high level of alpha 2 plasmin inhibitor plasmin complex. In this case, the combined use of tranexamic acid relieved the bleeding.

Southern blot and PCR analyses of dystrophin gene deletions in Japanese patients with Duchenne muscular dystrophy.

We have analyzed 34 Japanese patients of 31 families with Duchenne muscular dystrophy (DMD) by Southern blot and PCR (polymerase chain reaction) to detect large deletions in the genomic dystrophin gene on the X chromosome. Fifteen families (48%) had various deletions in the dystrophin gene by Southern blot analysis using the dystrophin cDNA probes 1-2a, 2b-3 and 8 which effectively covers the high-risk regions of deletion. Lengths and positions of the gene deletions detected were variable. Although there were no common deletions of exons in the gene of the DMD patients examined with the above cDNA probes, the deletion frequency of each exon was almost the same as the result previously reported in Caucasian DMD patients. The PCR method was also applied to confirm gene deletions in 9 cases from the above 15 families by amplifying 6 different regions of the dystrophin gene. Eight out of 9 DMD cases had gene deletions at the same genomic regions as found by Southern blot analysis. In a single DMD case, the Southern blot analysis indicated the exon 12 deletion, but PCR successfully amplified a 331 bp DNA fragment containing exon 12 and flanking introns. This difference may arise by a large deletion except for the above small 331 bp sequence in the HindIII digests of dystrophin gene.

Chiral separation of diltiazem, trimetoquinol and related compounds by micellar electrokinetic chromatography with bile salts.

The separation of optically isomeric diltiazem hydrochloride, trimetoquinol hydrochloride and related compounds by micellar electrokinetic chromatography was investigated employing four bile salts as chiral surfactants. The chiral separation of diltiazem hydrochloride and timetoquinol hydrochloride was successfully achieved by use of sodium taurodeoxycholate under neutral conditions, although enantiomers of carboline derivatives A and B and 2,2'-dihydroxy-1,1'-dinaphthyl were resolved with all the bile salts under conditions from neutral to alkaline. The chiral separation of diltiazem-related compounds was affected by the structure of the samples in addition to the effects of bile salt structures and pH of the buffer solutions. Application to the optical purity testing of trimetoquinol hydrochloride by the area percentage method is described. A possible chiral separation mechanism is briefly mentioned.

Separation and determination of aspoxicillin in human plasma by micellar electrokinetic chromatography with direct sample injection.

Both the separation and determination of aspoxicillin in human plasma by micellar electrokinetic chromatography (MEKC) were investigated. Selectivity in the separation of seven penicillin antibiotics was improved by using MEKC in comparison with capillary zone electrophoresis. Plasma proteins, which might interfere with drug analysis in conventional chromatography, were solubilized by the micelles employed in MEKC and eluted later than the drugs. This permitted the determination of the drugs in plasma by a direct sample injection method. One analysis of a plasma sample was performed within ca. 20 min without pretreatment. Good linearity and recovery were also obtained in the range of plasma levels usually encountered in clinical analysis with a correlation coefficient r = 0.999 and 94-104% recovery. The limit of detection for aspoxicillin was 1.3 micrograms ml-1 at a signal-to-noise ratio of 3.

A very small frame-shifting deletion within exon 19 of the Duchenne muscular dystrophy gene.

We report the molecular characterization of a Japanese Duchenne muscular dystrophy (DMD) patient. The analysis of genomic gene by polymerase chain reaction indicates that the individuals have a limited deletion within an amplified region, which encompasses exon 19 of DMD gene. The amplified region was sequenced. Comparison of the deletion joint sequence with the normal amplified region sequence indicates that both 5' and 3' deletion end points are present within exon 19 and the deletion removes total 52 bp out of 88 bp of exon 19. Both his mother and sister are carriers of the deletion-containing allele. The mutation introduces a termination codon at residue 791 in exon 20, and is predicted to result in the production of a severely truncated protein. This sort of deletion (designated as DMD-Kobe) might be classified as a new type of DMD gene abnormality.

Separation and determination of lipophilic corticosteroids and benzothiazepin analogues by micellar electrokinetic chromatography using bile salts.

The separation of corticosteroids and benzothiazepin analogues by micellar electrokinetic chromatography (micellar EKC) was studied in comparison with capillary zone electrophoresis. The separation of these substances was not successful under neutral and alkaline conditions because they migrated with the same velocity as that of the electroosmotic flow. Micellar EKC with sodium dodecyl sulphate (SDS) solutions was also not successful because these substances migrated with almost the same velocity as that of the SDS micelle, owing to their high lipophilicity. The use of bile salts, which have a similar skeleton to corticosteroids, as the micellar phase permitted the separation of these substances with high theoretical plate numbers (150,000-350,000) within a short time (ca. 15 min). Sodium cholate was particularly useful. The effects of bile salt concentration, pH and the addition of methanol were investigated. Micellar EKC was also applied to the determination of the drug substances in tablets and cream using the internal standard method and to purity testing of drug substances and tablets.

Isolation and characterization of three positional isomers of diglucosylcyclomaltoheptaose.

Three positional isomers of diglucosylcyclomaltoheptaose [(G)2-beta-cyclodextrin], 6(1),6(4)-di-O-(a-D-glucopyranosyl)-cyclomaltoheptaose (1), 6(1),6(3)-di-O-(a-D-glucopyranosyl)-cyclomaltoheptaose (2), and 6(1),6(2)-di-O-(a-D-glucopyranosyl)-cyclomaltoheptaose (3) were isolated by h.p.l.c. on a reversed-phase column from the mother liquors of a large-scale preparation of beta CD with Bacillus ohbensis cyclomaltodextrin glucanotransferase (EC 2.4.1.19) and were characterized by h.p.l.c. analysis of partial hydrolyzates and by 13C-n.m.r. spectroscopy. Their molecular weights were confirmed by f.a.b.-m.s. Their characteristic chromatographic behavior on four h.p.l.c. columns of different separation modes was found to be very useful for their identification. It is particularly noteworthy that the first application of a graphitized carbon column to CDs enabled a fine separation of all three positional isomers.
