[Silver impregnation of the myelin sheath. II].

A modified method is described based on the following steps: 1. simultaneous mordanting, using an alcoholic solution of phosphotungstic acid and tannic acid, 2. treatment with pH = 10.4 silver nitrate solution, 3. physical development (one or two phases).

[A new argyrophil reaction].

A method is described, which is based on mordantiation with a concentrated silver nitrate solution at 60 degrees C followed by two-step physical development at room temperature, with formalin as the reducing agent.

[The staining of amyloid].

All types of amyloid can be quickly and selectively demonstrated by staining with an alcohol-containing Sirius red solution.

[The histochemical use of alcian green].

With the hydrochloric acid-alcoholic solution of alcian green, without any pretreatment both the sulfated and the carboxylated proteoglycans can be demonstrated. After an indirect sulfation (periodic acid-bisulfite pretreatment), the glycogen and the glycoproteins, and after an oxidation with acidified potassium permanganate, the high cystine-containing proteins can also be stained. As a counterstain, a picric acid-Sirius red solution has proved good.

[Demonstration of polysaccharides by silver impregnation (2)].

To demonstrate carbohydrate-containing tissue elements becoming argyrophil by treatment with chromic acid and periodic acid respectively, the author proposes instead of the methenamine-silver nitrate solution the successive application of a silver nitrate solution, adjustable to the optimal pH, and of a physical developer.

[Silver impregnation of connective tissue. 4].

A method for silver impregnation of connective tissue is proposed which utilizes a physical developer instead of the traditional approach using successive treatments with ammoniacal silver solution and formalin.
