Reversible asymmetric septal hypertrophy in acute myocarditis. Serial findings of two-dimensional echocardiogram and thallium-201 scintigram.

Serial two-dimensional echocardiographic and thallium-201 scintigraphic findings are described in a patient with acute myocarditis diagnosed by endomyocardial biopsy. On the 4th day of illness, just before the onset of congestive heart failure, the echocardiogram showed asymmetric septal hypertrophy (IVS/PW = 16 mm/10 mm = 1.6) and thallium-201 scintigram showed the ventricular septal thickening. On the 8th day of illness, when severe congestive heart failure was seen, asymmetric septal hypertrophy disappeared (IVS/PW = 8 mm/8 mm = 1.0), the left ventricle dilated markedly (LVDd = 63 mm), and the wall motion became poor (EF = 0.24). After one month, when congestive heart failure and clinical inflammatory findings disappeared, the contractility somewhat improved (EF = 0.43), although marked left ventricular dilatation remained. Thallium-201 scintigram showed some scattered persistent perfusion defects, thinning of the ventricular septal thickening, and dilatation of the left ventricle. The right ventricular endomyocardial biopsy revealed the histologic findings of the late stage of acute myocarditis. It is concluded that transient thickening of the ventricular wall may represent early changes in acute myocarditis.

Autoradiographic evidence for dopaminergic innervation in guinea pig spinal cord.

In studies of the localization of the dopaminergic nerve terminals in the cervical cord of guinea pig, autoradiographic analysis of the spinal cord loaded with [3H]dopamine [( 3H]DA) was done under conditions that prevented the nonspecific uptake of [3H]DA. There was specific labeling in the gray matter and a high density of [3H]DA was present in the dorsal horn (DH). Moderate labeling was observed in the neuropil in the vicinity of the central canal. There were grain concentrations in close approximation to the cell bodies of numerous neurons in the DH and to the cell bodies of a few of the motoneurons in the ventral horn (VH). These dopaminergic terminals are possibly linked to sensory transmission and somatic motor function.

1H NMR spectroscopic studies of interactions between dimyristoylphosphatidylcholine (DMPC) and europium nitrate in the presence of tetracaine.

1H NMR spectroscopic studies have been carried out for multidispersed vesicles of DMPC and DOPC in the presence of Eu(NO3)3 and anesthetics (tetracaine and lidocaine). 1H NMR signals of DMPC vesicles disappeared, while those of DOPC did not, in the coexistence of Eu(NO3)3 and the anesthetics. An addition of EDTA-Na4 to this solution regained the signals of DMPC vesicles, implying cancellation of the effect of Eu(III) ions by chelating with EDTA anions. Inter-vesicular aggregation was proposed as a molecular mechanism responsible for the signal disappearance.

Effects of aspartame on diabetic rats and diabetic patients.

The effects of aspartame (L-aspartyl-L-phenylalanine methyl ester) on plasma glucose and insulin levels were investigated in diabetic rats and patients with non-insulin-dependent diabetes mellitus. The oral administration of 0.45 mg aspartame per 100g body weight, which is equivalent to 150 mg of glucose in sweetness, to streptozotocin-induced diabetic rats had no effect on the plasma glucose or insulin levels. Also, 225 mg oral aspartame loading, which is equivalent to 75 g of glucose in sweetness, to patients with non-insulin-dependent diabetes mellitus did not increase plasma glucose or insulin levels, although 75 g of oral glucose loading increased plasma glucose and insulin levels in diabetic patients as expected. Aspartame ingestion for three days at a dose of 24-48 mg per day and the intake of snacks flavored with 240 mg of aspartame also did not increase fasting plasma glucose levels. These results suggest that acute administration of aspartame has no influence on plasma glucose or insulin levels in diabetic rats and patients with non-insulin-dependent diabetes mellitus.

[Pulfrich stereoeffect during tracking eye movements].

An experiment was conducted to compare the magnitudes of the Pulfrich stereoeffect under two viewing conditions: fixating a stationary point and tracking a moving target. Three different transmittances of ND filters were used for each condition. The null method was employed to estimate the magnitude of the stereoeffect, in which the apparent depth was "nulled" by adjusting the physical depth of the target moving in an elliptical path in the eye-level horizontal plane so that no depth was seen. The results obtained from two experienced subjects showed that: the stereoeffect increased as the transmittance of the filter was decreased, and the stereoeffect was larger in the tracking condition than in the fixating condition for all three transmittances of the filters. The results were discussed within the context of depth information available for the visual system.

Chemical modification of sulfazecin. Synthesis of 4-(substituted methyl)-2-azetidinone-1-sulfonic acid derivatives.

In expectation of improving the antibacterial activity of sulfazecin by chemical modification at the 3- and 4-positions, a number of 3-[2-(2-aminothiazol-4-yl)-(Z)-2-(substituted oxyimino)-acetamido]-4-(substituted methyl)-2-azetidinone-1-sulfonic acids were synthesized. Among various 4-substituents explored, the carbamoyloxymethyl group was found to provide a good effect to the antibacterial activity of these 2-azetidinone derivatives. An extensive study of structure-activity relationships led to selecting (3S,4S)-3-[2-(2-aminothiazol-4-yl)-(Z)-2-carboxymethoxyiminoace tamido]-4- carbamoyloxymethyl-2-azetidinone-1-sulfonic acid, AMA-1080 (Ro 17-2301), which has highly potent antibacterial activity against Gram-negative bacteria including Pseudomonas aeruginosa, for further biological and subsequent clinical evaluation.

Effects of nifedipine on insulin secretion and glucose metabolism in rats and in hypertensive type 2 (non-insulin dependent) diabetics.

The effects of nifedipine (Adalat) on glucose metabolism and insulin release were studied in rats and in patients with type 2 diabetes mellitus complicated with hypertension. 1. In rats, 2.5-50 micrograms/kg of intravenous nifedipine reduced glucose tolerance and insulin release after intravenous glucose in a dose related fashion, although fasting blood sugar and insulin were not affected at 50 micrograms/kg of nifedipine. 2. Daily 20 to 60 mg of oral nifedipine for 12-75 weeks to 14 type 2 diabetics with hypertension did not affect their fasting blood glucose or hemoglobin A1. Mean glucose tolerance curve after the treatment was significantly ameliorated, although insulin response during the oral glucose loading did not show any significant change. Those results suggest firstly that there may be a difference in insulinopenic effect of nifedipine between the species, and secondly that long-term administration of nifedipine produced no adverse influence on glucose metabolism in type 2 diabetics.
