Effect of monomer structure on the mechanical properties of light-cured composite resins.

The effects of monomer structure on the mechanical properties of visible light-cured composite resins based on the seven types of aromatic dimethacrylates were investigated. The results of this study suggested that the mechanical properties of the composite resins were dependent upon the chemical structure of the dimethacrylate monomers employed. The composites based on dimethacrylates with hydroxy groups showed a relatively significant decrease in flexural strength, elastic modulus, and compressive proportional limit under wet conditions. The segmental mobility of dimethacrylate monomers considerably influenced the nature of cured composites. Bis-GMA-F-based composite showed superior mechanical properties to a conventional Bis-GMA-based material. The SEM observation of fractured surfaces revealed that failure mainly occurred through the resin matrix of the composite resins.

[Effect of filler system on the mechanical properties of light-cured composite resins. I. Effect of various types of silica fillers on the mechanical properties of the composite resins].

To investigate the effect of silica fillers on the mechanical properties of visible light-cured composite resins, Bis-GMA-based composites with four types of silica fillers were prepared. The mechanical properties of the composites with splinter-shaped silica fillers increased with increasing the filler fraction. Although the spherical silica filler could be filled more with resin monomer than the splinter-shaped silica filler, the mechanical properties of spherical silica-filled composite were relatively lower than those of the composite with splinter-shaped silica fillers. The micro particle silica-filled composite showed no obvious increase in the mechanical properties in either the dry or wet conditions. SEM observations of the fractured resin surface revealed that fracture occurred through the resin matrix as well as the resin/filler interface and the mechanical properties of each composite resin were correlated with the nature of crack propagation.

[Effect of filler system on the mechanical properties of light-cured composite resins. II. Mechanical properties of visible light-cured composite resins with binary filler system].

To improve the fracture resistance of the composite resin system under highly stressed conditions, a variable amount of microfiller was incorporated into the light-cured composite resins containing splinter-shaped silica filler. With the increasing the microfiller content (10-30 wt%) in the resin matrix, the elastic modulus, compressive proportional limit and compressive fracture strength of the composite resin were increased. The plastic deformation of the composite resins under compressive stress were decreased with increasing microfiller content. These findings suggested that the combination of a larger silica filler and microfiller would increase the compressive fracture resistance of the composite resin systems.

Growth, morphology, function, and morphogenetic properties of rat renal glomerular epithelial cells in vitro: effects of retinyl acetate.

We report here that retinyl acetate (RA) regulates growth, morphology, function and cell organization in rat renal glomerular epithelial cells (SGE1). SGE1 cells are able to grow in a serum-free medium (DHFs medium) which is supplemented with insulin, transferrin, selenium, bovine serum albumin (BSA), linoleic acid and epidermal growth factor (EGF). When 0.1 and 1 micrograms/ml RA were added to the medium, the growth rates in the sparse culture were noticeably increased, compared to those in DHFs alone, whereas more than 10 micrograms/ml RA was cytotoxic to the cells. In the confluent culture, addition of 0.1, 1.0 and 10 micrograms/ml RA prolonged the cell survival. Since 10 micrograms/ml RA is not cytotoxic to the confluent culture, the cytotoxic action of RA seems to be dependent on cell density as well as RA dose. Ultrastructural observation revealed that RA treatment caused an increase of microvilli and alteration of cell shape, from flattened to columnar. Biochemical and immunological studies revealed that RA treatment increased the activity of r-glutamyl transpeptidase (GGT) and an amount of the membrane component with molecular mass (Mr) of 108,000 which is identical to one of nephritogenic antigens, Fx1A. By using fluorescence phalloidin stain, it was found that RA treatment increased content and organization of F-actin fibers. Furthermore, in collagen-embedding culture, RA induced 3-dimensional (3D) growth of SGE1 cells leading to the formation of organoids, cystic spheres with central lumen, in a serum-free condition; the addition of DHFs to collagen gel alone was ineffective for the 3D growth.(ABSTRACT TRUNCATED AT 250 WORDS)

[Acute promyelocytic leukemia with a history of RAEB in transformation and the 15/17 translocation].

A 34-year old female was admitted to our clinic because of fever and general fatigue on March 26, 1987. On admission, peripheral blood (PB) revealed pancytopenia. Bone marrow smears revealed 9. 0% of promyelocytic cells with or without Auer rods. Diagnosis of RAEB in transformation was made. Chromosome study of the bone marrow cells showed t(15; 17) in 3 out of 20 cells analysed. After 3 months, the leukemic cells were observed in PB and increased in number. Then the patient showed bleeding tendency and fibrin degradation products (FDP) increased up to 40 micrograms/ml. And the leukemic cells were over 30% in PB at the end of July, 1987. The diagnosis of APL with DIC was made. To our knowledge, this is the first case of APL with a history of MDS with t(15; 17).

[Efficacy of home exercise program in a convalescent phase of acute myocardial infarction evaluated by Borg's indices, expiratory gas analysis and blood lactate].

The Borg's indices, the rate of perceived exertion have been widely accepted to evaluate subjective symptoms semi-quantitatively in exercise tests. We applied the indices to evaluate the efficacy of our home exercise program in patients with acute myocardial infarction. Eighty-five patients were entered in our home exercise program after their discharge from the hospital. This program consisted of everyday 2 km walk-jog exercise for 1 month keeping their heart rate at 100-110/min during the exercise. Before and 1 month after their discharge, they underwent treadmill exercise tests with expiratory gas analysis and a measurement of blood lactate. In the treadmill tests performed before and after the training, VO2 and heart rate increased linearly with Borg's indices, however VE and blood lactate had disproportionate increases after Borg's 13 which is considered to be the anaerobic threshold (AT). In fact, AT measured individually by expiratory gas analysis and blood lactate, appeared at Borg's 13 in 53% of the patients. After 1 month physical training, VO2 and heart rate significantly increased at the same Borg's indices, but there was no significant changes in blood lactate at the same Borg's score before and after the training. These results suggest that, in patients with acute myocardial infarction, exercise performance significantly increased at any score of subjective symptoms and that Borg's indices may reflect the blood lactate levels. It is concluded that Borg's indices, closely related to the dynamics of blood lactate, are not only useful in quantifying subjective symptoms but also to estimate the efficacy of physical training.

[A long-surviving case of gastric cancer with peritoneal metastasis (P3) responding to short-term high-dose chemotherapy and long-term immunotherapy].

A 37-year-old female was admitted to our hospital for further examination of epigastralgia. She was diagnosed as having multiple metastases due to advanced gastric cancer (Borrmann type 3). The operative findings showed bilateral ovarian (Krukenberg), Schnitzler and widespread peritoneal metastases involving the appendix (P3H0N2S2). She underwent total gastrectomy, splenectomy, bilateral oophorectomy, and appendectomy with CDDP (100 mg intraperitoneal administration). After operation, CDDP (50 mg/body, twice i.p. and once i.v.) and PSK (3.0 g/day) were administered. She has been followed in our outpatient department for 3 years without any recurrence. The findings suggest that combination therapy using short-term high-dose chemotherapy and long-term immunotherapy can be effective for such cases.

[Combination chemo-endocrine therapy of metastatic and stage IV breast cancer with cyclophosphamide, adriamycin, prednisolone and tamoxifen (CAPT)--with special reference to management of brain and liver metastasis].

Sixty-three evaluable patients with metastatic and stage IV breast cancer who had not previously undergone chemo-endocrine therapy were treated with a combination chemoendocrine therapy regimen consisting of cyclophosphamide 100 mg p.o. every day, adriamycin 10 mg i.v. on day 1 to 5, prednisolone 10 mg or 20 mg (20 mg was given on day 1 to 5) p.o. every day, and tamoxifen 20 mg p.o. every day. Adriamycin on day 1 to 5 was repeated three times every two weeks. After a total dose of 150 mg of adriamycin, the patients were changed to maintenance therapy consisting of cyclophosphamide 100 mg p.o., prednisolone 10 mg p.o. and tamoxifen 20 mg p.o. every day. After 72 months of the treatment there were 61 patients good for evaluation, 13 patients achieved a complete response (21.3%) with a median survival of 30.5 months and 18 patients had a partial response (29.5%) with a median survival of 21.0 months, and 30 patients failed to respond (49.2%) with a median survival of 8.5 months. There was a significant difference in survival time between responders (CR + PR) and non-responders (NC + PD) (p less than 0.001). Responses by site were seen in lung 10/18 (55.6%), liver 3/6 (50.0%), brain 2/4 (50.0%), bone 6/17 (35.3%) and soft tissue 14/24 (56.3%). A Satisfactory response for brain and liver metastasis, which are usually viewed as a sign of grim prognosis, was obtained similar to other sites of metastasis. Retreatment with CAPT, which was attempted in patients with secondary brain metastasis who responded to CAPT for initial brain metastasis, was uniformly effective. High ration of androgen to corticosteroid, positive estrogen receptors, long disease-free survival (over two years), premenopausal, high Broca' index (above 110) resulted from the chemo-endocrine therapy regimen CAPT. Toxicity was minimal and consisted of nausea, vomiting, alopecia and leucopenia.

Amino acid sequence of the calcium-binding light chain of myosin from the lower eukaryote, Physarum polycephalum.

We have established a new method for preparing Physarum myosin whose actin-activated ATPase activity is inhibited by micromolar levels of Ca2+. This Ca2+-inhibition is mediated by the Ca2+ binding to the myosin rather than by the Ca2+-dependent modification of the phosphorylated state of the myosin (Kohama, K., and Kendrick-Jones, J. (1986) J. Biochem. (Tokyo) 99, 1433-1446). Ca2+-binding light chain (CaLC) has been suggested to be primary importance in this Ca2+ inhibition (Kohama, K., Takano-Ohmuro, H., Tanaka, T., Yamaguchi, T., and Kohama, T. (1986) J. Biol. Chem. 261, 8022-8027). The amino acid sequence of CaLC was determined; it was composed of 147 amino acid residues and the N terminus was acetylated. The molecular weight was calculated to be 16,131. The homology of CaLC in the amino acid sequence with 5,5'-dithiobis-(2-nitrobenzoic acid) light chain and alkali light chain of skeletal muscle myosin were rather low, i.e., 25% and 30%, respectively. Interestingly, however, the CaLC sequence was 40% homologous with brain calmodulin. This amino acid sequence was confirmed by sequencing the cloned phage DNA accommodating cDNA coding CaLC. Northern and Southern blot analysis indicated that 0.8-kilobase pair mRNA was transcribed from a single CaLC gene. This is the first report on the amino acid sequence of myosin light chain of lower eukaryotes and nucleotide sequence of its mRNA.

Establishment and characterization of an epithelial cell line, SGE1, from isolated rat renal glomeruli.

An epithelial cell line, designated as SGE1, has been established from isolated rat renal glomeruli. SGE1 cells are able to grow continuously in a serum-free medium at similar growth rates to those in the medium containing serum. Quantitative studies of the cells in the serum-free condition demonstrated that SGE1 cells required a collagen type I or IV, fibronectin, or laminin-coated substratum for adhesion and growth, and among them, collagen type I and IV were most effective. Essential medium supplements for the adhesion and growth were epidermal growth factor and transferrin, respectively, and both effects were noticeably enhanced with linoleic acid. Morphological observation found that in a monolayer culture, SGE1 cells formed domes, and in a collagen embedding culture, they formed cystic spheres having features of a simple cuboidal epithelium, polarized formation of microvilli and tight junctions as well as a lateral cell membrane with cytoplasmic projections. In addition, SGE1 cells expressed Fx1A antigens, which are nephritogenic antigens on their microvilli.

Quantitative study on phagocytosis of human tonsillar adherent cells by the flow cytometric and immunocytochemical methods.

An in vitro phagocytosis assay of adherent cells of 12 palatine tonsils (7 children's and 5 adults' cases with persistent angina) was performed by use of flow cytometry (FCM), using fluorescent latex particles (FLP). The mean value of the percentages of phagocytic cells in the children's group (81.4%) was higher than that in the adults' group (64.0%). In addition, separate immunocytochemical stainings with each antibody--Anti-Leu-M5, OKDR, OKT6, S-100, and lysozyme--were made on smear preparations of a child's and an adult's tonsillar adherent cells after incubation with FLP. By use of the light microscope, the percentages of phagocytic cells in relation to positive cells for each antibody were calculated. The lysozyme positive cell proved to have the highest percentage of phagocytic cells. Then, the average number of phagocytized FLP in one positive cell for each antibody were calculated. With regard to the lysozyme positive cell, this cell proved to have the highest average number of FLP.

A muscarinic agonist-stimulated chloride efflux pathway is associated with fluid secretion in rat parotid acinar cells.

Cl- efflux from rat parotid acinar cells was stimulated by the muscarinic-cholinergic agonist carbachol (K0.5 approximately 1 microM). Carbachol application resulted in a rapid (T1/2 less than 10 sec.) and dramatic (approximately 50%) loss of intracellular Cl- followed by a much slower (T1/2 approximately 2 min) recovery to approximately 70% of original acinar Cl- content. Diphenylamine-2-carboxylate (1 mM), a putative Cl- channel blocker, blunted the initial Cl- loss and enhanced the subsequent recovery of Cl- content, while bumetanide (0.1 mM) blocked Cl- recovery with no effect on Cl- loss. Calculation of the rate of salivary secretion from measurements of sustained stimulated acinar Cl- release in vitro, yielded a value of 14 microliters/gland/min, in excellent agreement with previously published in vivo data. These results demonstrate that Cl- loss via this efflux pathway can account for fluid secretion from the rat parotid gland.

Brain surface invasion and metastasis of murine malignant melanoma variants.

Mouse B16 melanoma sublines were selected sequentially for their abilities to colonize brain meninges and leptomeninges of C57BL/6 mice. After 14 selections subline B16-B14b was established that formed significantly more brain tumor colonies than the parental B16 line. Examination of brains at various times after intravenous or intra-arterial injection of B16 cells by electron microscopy revealed that B14b melanoma cells lodged in small brain blood vessels, proliferated and invaded through vessel walls into brain parenchyma and also along small blood vessels at perivascular sites. Invasion into brain parenchyma was characterized by extension of melanoma cell filopodia resulting in fragmentation and sometimes enfulgment of glial and neural cells. Analysis of cell surface proteins of B16 melanoma sublines revealed increased exposure of a Mr approximately 90,000 glycoprotein on the high brain-colonizing cells. Antibodies against the Mr approximately 90,000 glycoprotein reacted with a variety of human melanoma cell lines and with some fetal and adult tissues, indicating that this melanoma-associated component is not species-, tumor- or tissue-specific. The glycoprotein could be a cell surface receptor important in the survival and growth properties of melanoma cells in brain microenvironments.
