Morphology and histochemistry of the spicules and gubernaculum of Haemonchus contortus (Nematoda: Trichostrongylidae).

A study has been made of the morphological, histological and histochemical characteristics of spicules and gubernaculum in Haemonchus contortus (Rud., 1803). The correlation coefficients of body length and left and right spicule lengths, left and right spicule lengths, and the distance of barb from the distal end of left and right spicule ends have been found to be statistically significant while the correlation coefficient of body length and gubernaculum length has been found to be non-significant. The sclerotized part of each spicule is composed of three layers differing in density. The shaft of the spicule possesses dorsal and ventral wings which curve towards the corresponding wings of the other spicule. Gubernaculum is a wedge-shaped structure. The spicules and gubernaculum are mainly proteinaceous. The sclerotized part consists of mainly keratin and collagen. The presence of traces of acid phosphatase in the spicules may represent some metabolic activity. The protoplasmic part is composed of proteins, carbohydrates and lipids.

BCG induced immunity to Mycobacterium leprae in mice.

One to three BCG vaccinations were given to various groups of mice--25 to 2 days preceding challenge with Myco. leprae. Harvesting of Myco. leprae was done every month upto a period of 7 months. Lymph node enlargement was taken as an indicator of successful vaccination. An increasing degree of protection was noted in BCG immunised mice as observed by a reduced number of bacilli harvested. Protection afforded was proportional to the number of doses of BCG given.

Myco. leprae infection in normal, thymectomised irradiated and thymus transplanted mice.

Normal mice and thymectomised, X-irradiated and thymic transplanted groups of mice were challenged with 10(3) Myco. leprae in the foot pad. Course of infection was studied for a period of seven months by sacrificing animals every month and counting the bacilli from the pooled tissues. In the thymectomised irradiated group the counts showed a plateau from 5th month onwards, whereas in control and thymus implant group the counts rose in 6th and 7th months.

Sphingomyelinase enzyme assay in Niemann-Pick disease.

Niemann-Pick group of diseases are rare autosomal recessive disorders of lysosomal enzymes. These are divisible into six types depending on clinical and biochemical features. On the basis of sphingomyelinase assay in five cases of Niemann-Pick disease, three cases were classified as type IA, one as type IS and one as type IIS. Their clinicopathological profiles are compared with 17 cases reported previously from India.

Serum alpha feto-protein screening in high risk pregnancies.

We assayed alpha fetoprotein (AFP) in serum samples from 2,735 women during 14 to 20 weeks of gestation. Serum AFP levels were elevated in the presence of neural tube defect (NTD) and gut atresia in fetus, twin pregnancies, preterm delivery and neonatal complications. In two of the 23 cases of fetal NTD the diagnosis was suggested by AFP assays, with apparently normal ultrasound findings. Low maternal serum AFP levels were associated with chromosomal abnormalities and hydatidiform molar pregnancy. Calculation of risk for Down syndrome based on maternal serum AFP and maternal age helped to reduce the number of women requiring amniocentesis. Maternal serum AFP assay was helpful in the management of threatened abortion, suspected intrauterine death, and maternal toxoplasma infection. In seven cases where maternal serum AFP was high but ultrasound studies were normal, male babies were delivered. Thus maternal serum AFP assay proved useful in narrowing down the group of women requiring more detailed surveillance and diagnostic studies.
