Enzyme studies in GM2 gangliosidiosis, and their application in prenatal diagnosis.

Assay of hexosaminidase A and B enzymes in four cases with developmental regression and cherry red spot on fundus examination confirmed that three cases had Tay-Sachs disease, and one case had Sandhoff disease. Prenatal diagnosis was carried out by hexosaminidase enzyme assay in amniotic fluid and cells in one family, and chorionic villus sample in the second family. The fetus was diagnosed to be unaffected in one, and affected in the other family. Assay of hexosaminidase A and B is useful for specific diagnosis of GM2 gangliosidosis, and for prenatal diagnosis to reduce the burden of these disorders.

Dietary protein regimens and chronic ethanol administration effects on sodium- and proton-dependent solute uptake in rat intestine.

The effect of feeding ethanol daily for 40 days has been studied on intestinal uptake of glucose, glycine, and leucine in rats fed control, 8% protein (LP), and 30% protein (HP) diets. Na(+)-dependent uptake of glucose and glycine both at pH 7.2 and pH 5.5 was significantly depressed (p < 0.001) in ethanol or LP diet-fed animals and remained unaffected in HP-fed rats compared to the controls. But ethanol administration to protein-malnourished rats enhanced the Na(+)-linked glucose and glycine uptakes. Leucine uptake remained unaffected under these conditions. Glucose uptake remained unaltered whereas glycine uptake was reduced when ethanol was administered to rats given HP diet. In the absence of Na+, uptake of glucose, glycine, and leucine was more at acidic pH compared to that at pH 7.2 under all the experimental conditions investigated. Proton-linked uptake of solutes was unaffected by feeding ethanol, LP, or HP diet in rats. Thus, chronic ethanol feeding specifically depresses the Na(2+)-dependent uptake of glucose and glycine. Receptor-Ck-dependent regulation of genes in HL-60 cell line.

The selective and conspicuous absence of a novel receptor-Ck (having affinity for cholesterol moiety in lipoproteins and intrinsic tyrosine kinase activity) in various leukemic cell lines/patients, prompted us to explore the nature of interrelationship between receptor-Ck deficiency and over-expression of genes coding for bcl-2, cyclin 'D', chimeric bcr-abl, c-myc and LDL-receptor in the human promyelocytic leukemic cell line (HL-60). This study revealed unambiguously that deregulated expression of these genes is primarily due to the inability of these cells to express the receptor-Ck gene. Based upon the observations we propose that 'receptor-Ck' deficiency may be responsible for the initiation of chronic myeloid leukemia.

Metaplastic carcinomas of the breast (light microscopic and immunohistochemical features).

Metaplastic carcinomas of the breast are uncommon tumors that contain other cellular components besides the glandular component. These carcinomas have a poor prognosis as compared to breast carcinomas without metaplasia. It is, therefore, important to recognise the metaplastic components. The study was conducted over a three year period. All breast carcinomas received during this period in the department of pathology were analysed for areas of metaplastic carcinomas. The light microscopic features, special stains as well as the immunohistochemical markers of metaplastic carcinomas are highlighted in this paper.

The zymodemes of Entamoeba histolytica in New Delhi, India.

The prevalence of various pathogenic and non-pathogenic zymodemes of Entamoeba histolytica in New Delhi, India was studied by characterising its isoenzyme patterns. Non-pathogenic zymodeme I was found to be most common and was detected in 39 of 49 infections. The other non-pathogenic zymodemes detected were XVII and XVIII in two cases each. The pathogenic zymodemes detected were II (four cases), II alpha- (alpha) (one case) and XIV (one case). Approximately 72% of those harbouring non-pathogenic zymodemes and 33% of those harbouring pathogenic zymodemes were asymptomatic. All the symptomatic individuals harbouring non-pathogenic zymodemes were seronegative for amoebiasis. However, subjects harbouring pathogenic zymodeme but asymptomatic were seropositive for amoebiasis at a titre of greater than 1:80, indicating a past infection or subclinical tissue invasion by the parasite. The new zymodeme isolated from a case of acute dysentery was designated II alpha- on the basis of the banding pattern of the enzyme glucose phosphate isomerase.

Leprosy among circus workers.

Seven (2.31%) circus workers of two circus companies visiting Amritsar were detected to be suffering from leprosy. Whether or not they acquired the disease during their stay in circus is not so important, but it is necessary to detect and treat these itinerant persons. Our experience clearly underlines the necessity for undertaking surveys of various workers living in crowded conditions (like circus), because of their occupation.

Monkey bites: a public health problem in urban setting.

Animal bites constitute an important public health problem. Monkey bites accounted for 8.8% of total animal bites (1987-1991) treated in Postgraduate institute of Medical Sciences, Rohtak. It was noted that monkeys are becoming the second most common animal to inflict bites. The hospital data revealed that 63.5% of the total animal bites belonged to class-III type. The article also details two case studies of monkey bites.
