Some properties of rabbit anti-human thymus serum.

A highly specific anti-human T lymphocyte serum (AHTS) was obtained by sequentially absorbing rabbit anti-human thymocyte serum (ATS) with human red cells, neutrophils, Burkitt's lymphoma cells (Daudi cells), liver, kidney and glutaraldehyde insolubilized human serum. The immunofluorescent staining revealed that the basement membrane of human renal glomeruli reacts with AHTS before being absorbed with kidney. Specificities of AHTS were studied by the cytotoxicity test, inhibition of PHA responses, inhibition of E-resulting and immunofluorescent staining.

Quantitative studies o the activities of anti-human tonsil sera and anti-human spleen sera against B lymphocyte.

Anti-human B lymphocyte serum (AHBS) was obtained by absorbing rabbit anti-human tonsil serum (ATOS) or rabbit anti-human spleen serum (ASPS) with human red cells, liver, neutrophils, thymocytes and glutaraldehyde insolubilized human serum. Specificities of AHBS were checked by the cytotoxicity test, inhibition test of EAC-rosetting and inhibition of PWM responses. Raji cells of Burkitt's lymphoma originating from B-cells and acknowledged as non-immunoglobulin bearing on the cell surface were stained with AHBS according to the immunofluorescent technique. ATOS showed a higher cytotoxic titer against peripheral B-cells and inhibited EAC-rosetting stronger than ASPS did. On the other hand, macrophages in peripheral blood, Kupffer cells in the liver and certain cells of the thymus-medulla were stained with ASPA, but not with ATOS, even after intensively absorbing ASPS with thymocytes.

Development of the lung in fetal rabbits with experimental diaphragmatic hernia.

Congenital diaphragmatic hernias were successfully produced in fetal rabbits. The lungs of these animals remained small and hypoplastic on gross examination, although histologically they have matured significantly from the original pseudoglandular stage. It is our impression that the so-called hypoplastic lung in diaphragmatic hernias is small in size but potentially functioning when relieved of compression.

Depolarization-induced calcium release from sarcoplasmic reticulum fragments. I. Release of calcium taken up upon using ATP.

Ca2& taken up by sarcoplasmic reticulum membrane fragments (SRF) upon using ATP could be released rapidly by changing the anion outside the vesicles from methanesulfonate to chloride. It is considered that this anion exchange caused depolarization of the sarcoplasmic reticulum membrane. Similar rapid release of Ca2& taken up by SRF was also caused by a change from high to low osmotic pressure, probably due to bursting of the membrane. On the basis of experiments in which these two types of Ca2& release were discriminated, it was concluded that Ca2& bound inside the membrane was released directly by anion exchange (depolarization). However, Ca2& release was not caused by cation exchange. Sucrose inhibited these two types of Ca2& release. Cia2& taken up in the presence of oxalate could not be released by any treatment used. Liver microsome fraction also has Ca2& uptake activity. However, Ca2& was not released upon anion exchange, but was released upon oxmotic change. These results show that Ca2& release from SRF upon anion exchange is specific to the sarcoplasmic reticulum membrane. In conclusion, SRF membrane retains the ability to respond to the depolarization caused by ion exchange and can release the accumulated Ca2&.

Depolarization-induced calcium release from sarcoplasmic reticulum fragments. II. Release of calcium incorporated with ATP.

Ca2& incorporated in vesicles of sarcoplasmic reticulum fragments (SRF) by diffusion could be released rapidly by changing the ionic environment, by dilution from methanesulfonate (MS) to chloride. This ion exchange is considered to make the membrane potential of SRF inside-negative. Much faster release of Ca2t was also observed upon osmotic change from high to low. These responses were very similar to the Ca2& release from SRF after take up using ATP, but the release rate was slow in the case of anion exchnage. The behavior of K&, Na&, sucrose, and inulin incorporated in SRF was followed upon similar treatment. These ions and nolecules were not released upon ion exchange, but were immediately released by osomtic treatment. Therefore, the Ca& release upon anion exchange was not due to the bursting of SRF, but to a direct effect such as a membrane potential change of the SRF. The behavior of anion such as C1- and propionate could not followed by the same method because of the large permability of these anions. It was also shown that Ca& release upon ion exchange was not a direct effect of pH change. Liver microsomes did not show Ca& release upon the same treatment as SRF.

Histopathological studies on intrahepatic bile ducts in the vicinity of porta hepatis in biliary atresia.

Intrahepatic bile ducts in the vicinity of the porta hepatis were histologically investigated in 9 cases of biliary atresia and the pattern of bile duct was divided into two groups. In the first group, there were a few large bile ducts which had continuity to the extrahepatic bile duct. In the second group, many small bile ducts were histologically observed, but there was no large bile duct at the porta hepatis. This type of abnormal bile ducts was seen in the cases of obliteration of bile ducts at porta hepatis.

The effect of oxisuran on differential inhibition of cell-mediated immunity.

Oxisuran, 2-[(methylsulfinyl) acetyl] pyridine suppressed allogeneic skin graft rejection in rats. In PHA responses and one way mixed lymphocyte culture, suppression of cell-mediated immunity was indicated. However, little inhibition of PFCs was observed with Oxisuran, and unlike Azathioprine or Neocarzinostatin, Oxisuran did not concomitantly suppress antibody-producing system in rats. These data suggest that selective suppression of cell-mediated immunological responses by Oxisuran is possible in rats, and Oxisuran may be a better immunosuppressant for clinical organ transplantation.

Acetylcholine-binding substance extracted by using organic solvent and acetylcholine receptor of electric organ of Narke japonica.

1. Proteolipid was extracted from the electric organ of Narke japonica by using chloroform/methanol (2:1, v/v). This extract was separated into acetylcholine-binding and non-binding substances by column chromatography. However, acetylcholine-binding substances did not show the characteristic properties of protein. 2. The membrane fragments of the electric organ were separated into three main parts by sucrose density gradient centrifugation. From the heaviest, the fractions were acetylcholine receptor rich, ATPase rich, and acetylcholinesterase rich. 3. The membrane fraction having acetylcholine receptor showed the excitability, the increase of Na+ permeability by the application of cholinergic agonists. However, the acetylcholine binding substance extracted by the organic solvent was richer in the lighter fraction. This substance differed from the true acetylcholine receptor.

Follow-up studies of long term survivors after hepatic portoenterostomy for "noncorrectible" biliary atresia.

Fourteen patients with "noncorrectable" biliary atresia are living without jaundice for more than 2 yr after hepatic portoenterostomy or its modification. Retardation of physical growth was observed in one of them, and mental retardation in another, both of which seemed irrelevant to biliary atresia. Serial tests for liver function after operation revealed early recovery of serum bilirubin, transminase, and turbidity, and delayed improvement of alkaline phosphatase. Postoperative needle biopsy of the liver disclosed that changes in hepatic parenchyma and ductular proliferation were rapidly improved after successful operation. Improvement of fibrosis of the liver was delayed, and it was not satisfactory in patients whose preoperative changes in the liver were severe or in whom ascending cholangitis had been a frequent complication. Histologic features of hepatic cirrhosis were observed in the liver in three cases, in two of which there had been frequent episodes of cholangitis. Only one of these showed clinical signs of portal hypertension. Functional and morphologic cure can be achieved in "noncorrectable" biliary atresia by hepatic portoenterostomy or its modifications, although varying degree of hepatic fibrosis may remain according to severity of preoperative changes of the liver and postoperative complication of ascending cholangitis.
