Purification and characterization of the neutral endopeptidase from human kidney.

The presence of an endopeptidase hydrolyzing succinyl trialanine-p-nitroanilide [Suc(Ala)3-pNA] to Suc(Ala)2 and Ala-pNA in human kidney and its partial characterization have been reported (Ishida et al. (1981) Biochem. Int. 3, 239-246). This neutral metallo-endopeptidase was separated into two fractions (A and B) on Sephacryl S-300 and fraction B was further purified to an electrophoretically pure state. The fraction B enzyme had a molecular weight of 100,000 and was inhibited by metal chelators such as EDTA, o-phenanthroline and phosphoramidon, but not by serine protease inhibitors. The enzyme was found to hydrolyze peptide bonds preferentially at the amino sides of hydrophobic amino acids such as Leu and Phe, when its specificity was studied using insulin B chain and angiotensin I. Fraction A seems to be a tetramer of fraction B, judging from its molecular weight, pI, substrate specificity and immunological properties.

Genomic analysis of antigenically related avian paramyxoviruses.

Six avian paramyxoviruses isolated from wild and domestic ducks in the United States, Hong Kong and Japan were characterized antigenically and genetically. All viruses examined were shown to be antigenically closely related. Oligonucleotide patterns of duck/Miss/334 and duck/Miss/406 were apparently distinguishable from those of duck/Miss/116 and duck/Miss/320 isolated in the same area of the United States. On the other hand, two viruses isolated from a domestic duck in Hong Kong (duck/Hong Kong/D3/75) and from a domestic duck in Japan (duck/Tokyo/41/78) were genetically very similar to that of duck/Miss/116, suggesting that these three viruses represent a genetically homogeneous group and may be of the same origin.

Effect of porcine calcitonin therapy on vitamin D metabolism and clinical response in a patient with osteogenesis imperfecta.

A 1 11/12-year-old girl with osteogenesis imperfecta was treated with porcine calcitonin. Eight bone fractures occurred in the previous 20 months before therapy, but none occurred during eight months of therapy. There was also a significant improvement in linear growth and radiographic bone density. This is the first study of the effect of calcitonin on vitamin D metabolism in a human. The high plasma levels of 1,25 dihydroxy-vitamin D (1,25-(OH)2-D) and 24,25 dihydroxy-vitamin D (24,25-(OH)2-D) before calcitonin therapy decreased after therapy. Plasma 25 hydroxy-vitamin D (25-OH-D) concentration, which normal in level before calcitonin therapy, was normal or slightly decreased during administration. It is concluded that calcitonin probably influences vitamin D metabolism in a patient with osteogenesis imperfecta.

Extracellular excitatory junctional current at the crayfish neuromuscular junction.

The nature of the falling phase of the synaptic current was studied in more detail at the crayfish neuromuscular junction and fundamental data relating to focally recorded excitatory junctional potentials (e.j.p.s) was complemented. The decay of extracellular e.j.p.s at the crayfish neuromuscular junction was almost exponential. The decay time constant of extracellular e.j.p.s varied considerably from one e.j.p. to another, the mean of the decay time constant being about 1 ms at the normal resting potential at a bath temperature of 22 degrees C. There was no detectable correlation between the decay time constant and the amplitude of extracellular e.j.p.s which were induced by a single stimulation of the excitatory axon. Prolonged application of glutamate hardly affected the decay time constant of extracellular e.j.p.s. The significance of the study on the crayfish neuromuscular system was discussed.

Characteristics of auto-antibodies to bovine TSH in the serum of two patients with Graves' disease.

In this report we describe the characteristics of auto-antibodies to bovine TSH (bTSH) detected in the serum of 2 females among 102 patients with Graves' disease. These patients had never been injected with bTSH. One patient had high LATS activity and high bTSH binding activity after isotope therapy. The other patient showed no detectable LATS activity. Interestingly, the antibody showed a specifically high binding activity for the labelled TSH preparation purified by receptor. The auto-antibody could be demonstrated by the double antibody method, polyethylene glycol method, and by gel-filtration. The antibody was polyclonal immunoglobulin G (IgG). Because the binding of [125I]bTSH with the patient's antibody was inhibited by pituitary extracts from mammalian species other than human, this antibody may cross-react with bovine, rat, dog, rabbit and whale TSH. Although the incidence of the antibody in Graves' disease is low and the pathological significance remains obscure, the existence of this antibody in the serum of patients may suggest that autoimmune mechanisms may involve not only the thyroid but also the pituitary in Graves' disease.

Elevation of cAMP levels in cerebrospinal fluid of patients with neonatal meningitis.

The cyclic 3',5'-adenosine monophosphate (cAMP) concentrations in the cerebrospinal fluid of 20 children with neonatal bacterial meningitis and aseptic meningitis were measured by radioimmunoassay method. The cAMP levels were found to be significantly elevated above control levels (P less than .01) during the acute phase in most of the patients. In the convalescent stage the cAMP concentration was decreased but levels remained significantly elevated (P less than .01) in patients with complications. During the acute phase, the cAMP levels were higher in neonatal bacterial meningitis than in aseptic meningitis (P less than .01). The results suggest that cAMP is a sensitive indicator of transient cellular metabolic disturbance in the brain and may be used to monitor the course of neonatal meningitis.

The circadian rhythms of blood ionized calcium in humans.

It is well known that several hormones and minerals develop daily variation in human blood. Recently the presence of circadian rhythms of blood ionized calcium and serum phosphate concentrations in man reported (Science 213: 672-674, 1981). To clarify the difference in race and age, we examined the serum concentration of minerals such as ionized calcium, inorganic phosphate, total calcium and magnesium, and venous blood pH during 24 hours in three 31 to 40 years old Japanese males in good health. Result [1]. There was a correlation between concentration of ionized calcium and blood pH (r = -0.467 p less than 0.001), and between concentration of ionized calcium and total calcium (r = 0.366 p less than 0.005). [2]. Apparent rhythms of ionized calciums and inorganic phosphate concentrations has been noticed.

Autologous rosette forming cells in patients with renal diseases.

We studied the distribution of autologous rosette forming cells (ARFC) in the peripheral blood of 30 healthy adult donors, 30 patients with IgA nephropathy, 20 patients with primary glomerular diseases, eight patients with systemic diseases and 25 patients with other renal diseases. The mean percentages of ARFC were markedly reduced in the IgA nephropathy patients compared with the healthy adult donors. The values for ARFC were even more significantly reduced in IgA nephropathy patients compared with patients with primary glomerular diseases, systemic diseases and other renal diseases. This means that the immunoregulatory abberation in IgA nephropathy primarily involves T cells.

Blow-out fractures.

Twenty-one cases of blow-out fractures were treated at Osaka University Hospital between June 1979 and February 1982. 52% of the patients were teenagers. The most important cause of a blow-out fracture was a fist-fight (48%). Most of the patients had diplopia caused by the disturbance of the eyeball movement, mainly in the upward or downward rotations. 19 of 21 patients were surgically treated. The prognosis was much more related to the severity of the fracture than the period between the injury and the surgical treatment.
