In an attempt to examine the specific metabolic relationship between the alkane substrates and lipid products the pattern of formation of cellular lipid, especially fatty acids, has been studied in the course of incubation of cells of C. rugosa with n-alkanes in mineral medium. C18:2, C18:1, C16:1 and C16:0 fatty acids were the major products formed, irrespective of the odd or even chain length of the alkane used as substrate. These unsaturated fatty acids reached a maximum in the stationary phase. In the case of cells grown on odd-chain n-alkanes (from C11 to C19), the proportion of odd-chain cellular fatty acids were markedly high, reaching 77--88% in the n-pentadecane and heptadecane-grown cells. The resulting acids are then metabolized by beta-oxidation or inserted directly or after elongation with C2-units into the cellular fatty acids. The total lipid content of n-alkane (n-C10-C20) grown cells in the stationary phase was 16.3--19.0% of th dry cell weight, which was about three times as much as that of glucose-grown cells. The chain length of alkane substrates had a significant effect on the lipid content. Ergosterol production from n-hexadecane was twice higher than that from glucose.
Akita Prefecture has an especially high mortality rate from stroke, and its age-adjusted death rate from stroke is the highest in Japan. We have carried out an epidemiological survey of cardio- and cerebrovascular diseases (CVD) in farming villages in this prefecture since 1963, with a response rate of 84%. During our 8-year follow-up, 94 new stroke cases were observed among 1,814 subjects. Multiple logistic function analysis was carried out on nonstroke and stroke cases from subjects aged 40 to 69 years at time of initial examination, in order to clarify the risk factors for stroke. The analysis used eight variables: age, sex, systolic blood pressure, obesity index, urinary sugar, urinary protein, serum total cholesterol, and total protein. The results showed that hypertension was the most important risk factor for stroke. However, regarding cholesterol, multivariate analysis showed that among men and women aged 40 to 69 years at entry, subjects with low serum total cholesterol levels were more prone to cerebral hemorrhage, but that serum cholesterol level had no weight as a risk factor for cerebral infarction. These results correspond well with the observed fact that stroke incidence or death rate in Japan is higher in populations with high prevalence of hypertension and low concentration of cholesterol, and also with the fact that death rate from hemorrhage declines with the increment of serum total cholesterol and the westernization of diet.
X-ray examination and intraoperative fiberscopy of the small intestine were performed in 20 patients with familial adenomatosis coli. Roentgenographic study detected polypoid lesions in the upper jejunum in 2 patients (10%), and in the terminal ileum in 9 (45%). In contrast, endoscopic study revealed polyps in the jejunum in 10 patients (50%), and in the ileum in 14 (70%). Polyps in most cases were multiple and small in size (less than 3 mm in diameter). Histology revealed the lesions of the jejunum to be adenomas, and those of the ileum to be lymphoid hyperplasia with or without adenoma. The present study suggests that intraoperative endoscopy is much more valuable than roentgenography in detecting these minute polyps of the small intestine in this disease.

Three cases are reported in which multiple sessile polyps of the stomach disappeared spontaneously during the follow-up period from 9 to 34 mo. All of the patients were middle-aged females, and the distribution of the polyps was limited to the portion of the stomach with fundic glands. Histolgoic examination revealed simple hyperplasia of the fundic glands with microcysts. The macroscopic and histologic pictures were the same as the gastric lesions ("fundic gland polyposis") found in cases with familial adenomatosis coli; however, colonic polyposis was not observed in our cases. The possible cause of the spontaneous disappearance of the polyps was discussed.

[14-14C]16 alpha-Hydroxy-C-18- and C-19-steroid hormones were obtained in good yields by microbiological hydroxylation of correspondingly labelled steroids by Streptomyces roseochromogenes NRRL B-1233. Trace quantities of the labelled substrates were incubated on a rotary shaker (220 rpm) at 27 degrees C. The radioactive products were chromatographically separated, identified and the radiochemical purity was established by isotopic dilution analysis. The specific activities of 16 alpha-hydroxy-steroids obtained were assumed to be the same as those of the substrates, namely, 57.5 mCi/mmole for 16 alpha-hydroxy-4-androstene-3,17-dione, 57.5 mCi/mmole for 5-androstene-3 beta,16 alpha,17 beta-triol, 57.5 mCi/mmole for 16 alpha-hydroxy-dehydroepiandrosterone, 55.7 mCi/mmole for 16 alpha-hydroxy-estrone, and 57.5 mCi/mmole for 16 alpha-hydroxy-testosterone.
The introduction of a 16 alpha-hydroxyl function into the steroid nucleus was studied in resting cells of Streptomyces roseochromogenes NRRL B-1233. The oxidation product of dehydroepiandrosterone (DHEA) was identified as 16 alpha-hydroxy DHEA by using thin-layer and gas-liquid chromatography. A linear relation between cell concentration and 16 alpha-OH-DHEA formation was observed. 16 alpha-Hydroxylase showed good activity at pH 8.0 for 16 alpha-OH-DHEA formation. The enzyme showed good activity at 3.1 x 10(-4) M DHEA. The oxidation products of pregnenolone, 4-androstene-3,17-dione, estrone, and 5-androstene-3 beta,17 beta-diol as well as of other substrates were identified as the 16 alpha-hydroxy steroid, respectively. The rates of microbial 16 alpha-hydroxylation were as follows: 76.9% for DHEA, 50.4% for pregnenolone, 43.9% for 4-androstene-3,17-dione, 34.3% for estrone, and 19.6% for 5-androstene-3 beta,17 beta-diol. The organism tested catalyzes 16 alpha-hydroxylation of a wide variety of steroids.
