Toxicological studies on bestatin. III. Chronic toxicity test and recovery study in beagle dogs.

Chronic toxicity and its recovery of bestatin (NK421) was studied in both sexes of 28 Beagle dogs. At dose levels of 96, 38.4 and 15.4 mg/kg, NK421 was administered orally to dogs for 540 successive days. Control dogs were treated orally with 2 g/dog of corn starch. Each group consisted of 3 males and 3 females, and 2 males and 2 females were added to the 38.4 mg/kg group for a recovery test of 35 days. As general signs, anorexia, abnormal feces (loose stool, diarrhea, mucous stool), loss of activity, loss of lustre in fur, decoloration of the visible mucosa and emaciation were transiently observed in a early stage in 1 male and 1 female of the 96 mg/kg group. In correlation with these signs, slight anemia appeared hematologically, and the increased alkaline phosphatase activity and the decreased albumin ratio in serum protein fractions were observed biochemically. Except for the slight abnormal findings observed in the liver of the above 2 dogs, no significant changes were histopathologically noticed in any organ of all the dogs examined. The maximum non-toxic dose of NK421 in this study is estimated to be 38.4 mg/kg in dogs.

Determination of prednisone and prednisolone in human serum by high-performance liquid chromatography--especially on impaired conversion of corticosteroids in patients with chronic liver disease.

A reliable and rapid method is described for the determination of prednisone and prednisolone in human serum by high-performance liquid chromatography, using a Zorbax-SIL column with dichloromethane-ethanol (92.5:7.5) as eluent, with UV detection at 254 nm. Metabolites and endogenous hydrocortisone did not interfere with the determination of prednisone and prednisolone. The alteration of corticosteroid concentrations in serum from patients with chronic liver diseases was studied following a single oral administration of prednisone or prednisolone (30 mg). The proposed method showed good separation of several corticosteroids and was time-saving, suitable and reliable for the routine analysis of corticosteroids in human serum.

Multiple regression and response surface analyses of the effects of dietary protein, fat and carbohydrate on the body protein and fat gains in growing chicks.

Multiple regression analysis of biological response as a function of three factors having a common denominator, on a triangular graph, was investigated for qualitative and quantitative evaluation of nutritional phenomena. As an example of its application, 33 groups of White Leghorn male chicks were isocalorically force-fed purified diets consisting of 5-47% protein, 6-86% fat and 2-89% carbohydrate on a metabolizable energy basis for 10 days, and prediction equations for the body weight gain, protein gain and fat gain were formulated by multiple regression techniques. Perspective views, contour maps and cross-sectional views of the response surfaces constructed from the equations revealed 1) that both body weight gain and protein gain increased with an increase in the metabolizable energy of dietary protein, and were not much affected by the calorie ratio of dietary carbohydrate to fat, showing a convex configuration of the response surfaces; 2) that fat gain decreased and appeared to become more dependent on the calorie ratio of dietary protein, fat and carbohydrate were replaceable with one another in the range of 40-50% protein metabolizable energy with respect to body weight gain and protein gain, while replaceability of fat and carbohydrate with each other seemed to decrease with an increase in protein metabolizable energy with respect to fat gain. These results demonstrated that the proposed method is a valuable tool in nutrition studies.

The recurrent branch of the lateral cutaneous nerve of the forearm.

Cutaneous nerves of the upper arm were observed in 31 Japanese cadavers dissected in a student course of gross anatomy dissection and in cadaver selected randomly for intensive dissection. Contrary to the descriptions in current textbooks of human anatomy, the lateral cutaneous nerve of the forearm supplies also the skin over the distal region of the anterolateral surface of the upper arm. A branch to the upper arm (the recurrent branch) arises from the proper trunk of the lateral cutaneous nerve of the forearm, recurs along the cephalic vein as far as the middle third of the upper arm and distributes filaments to the skin close to the vein.
