Suppression of mixed lymphocyte reaction by progesterone and estradiol-17beta.

The effects of estrone, estradiol-17beta, estriol, testosterone, progesterone, and 20alpha-dihydroprogesterone on incorporation of 3H-thymidine into cultured lymphocytes from normal men and nonpregnant women in response to allogeneic stimulation was investigated with two-way MLR. A dose-dependent reduction in the mean percent incorporation was observed in the range of progesterone concentration between 2 x 10(3) and 10(4) ng/ml with statistically significant differences from the control cultures. Estradiol-17 beta at 10(4) ng/ml produced a slight but significant inhibition of incorporation. No significant change in incorporation occurred with the other steroids tested. Since the suppressive property of progesterone in phytohemagglutinin-induced lymphocyte culture reported in our previous communication was reproduced by the present investigation in MLR, the possible role of progesterone in immunoregulation during pregnancy is emphasized as has been postulated previously.

[In-vitro motility studies on the strips of the canine gallbladder (author's transl)].

In-vitro motility of the gallbladder was studied in the longitudinally cut full-thickness strips obtained from 61 mongrel dogs. The strips, 5 mm in width and 15 mm in length, were prepared from the fundus, body and infundibulum of the gallbladder wall and the motility was examined by the changes of contractile dynamics which were transduced on a pen chart recorder. The results were summarized as follows: 1. The specimen from three parts of the gallbladder exhibited spontaneous motility which was comprised of tonic and rhythmic contractions and they seemed to be stable approximately during the period from three and half an hour to six hours in the Tyrode solution. 2. There was a characteristic pattern which was specific for each three parts in the waves of spontaneous rhythmic contraction. The specimen from infundibulum showed almost regular waves which were predominantly comprised of greater amplitude than those of other two parts. 3. The effects of gastrointestinal hormones and similar substance, caerulein, tetragastrin and secretin were investigated on the activity. Caerulein and tetragastrin were found to be potent stimulant of the dog gallbladder. The stimulated contraction waves were uniform and the pattern could be described with using a certain mathematical expression.

[In-vitro motility studies on the strips of the human gallbladder (author's transl)].

Studies in-vitro motility of the human gallbladder was investigated in the longitudinally cut strips obtained from 24 patients. Strips of the wall of the organ, containing mucosa, muscle and serosa, 5 mm in width and 15 mm in length, were cut from the fundus, body and neck. These specimens were suspended in Tyrode solution at 36 degrees C equilibrated with the 100% O2. Recordings were made on a pen chart recorder which were transduced using a isometric system by the changes of contractions. The results obtained were as follows: 1. The strips from three parts of the gallbladder showed rhythmic and tonic spontaneous contractions. 2. The tonic contractions from fundus were strongest in those three. 3. The rhythmic contractions of neck exhibited greater amplitude than those of fundus and body.

Overshoot phenomenon of phytohemagglutinin response after chemotherapy and its relationship to remission in acute non-lymphocytic leukemia.

To define the relationship between cell-mediated immunity and responses to chemotherapy or prognosis, delayed cutaneous hypersensitivity, E- and active E-rosette tests, and mitogenic responses of lymphocytes were examined in 15 patients with acute nonlymphocytic leukemia. Its results indicated that cell-mediated immunity before remission induction chemotherapy did not correlate with the outcome of treatment. In contrast, delayed cutaneous hypersensitivity and mitogenic response tested after remission induction correlated with therepeutic effect. Further a "rebound" or overshoot of phytohemagglutinin responsiveness was observed in patients achieving remission. Serial studies on cell-mediated immunity may be useful for predicting therapuetic efficacy and prognosis in patients with acute leukemia.

Pharmacological and biochemical studies on a new compound, 2-(7-indeyloxymethyl)morpholine hydrochloride (YM-08054-1), and its derivatives with potential antidepressant properties.

Antidepressant properties of a new indene derivative, YM-08054-1, and its related compounds were compared with those of tricyclic antidepressants and viloxazine. The potencies of YM-08054-1 to inhibit uptake of both 14C-norepinephrine (14C-NE) and 14C-5-hydroxytryptamine (14C-5-HT) by the rat brain synaptosomes were similar to those of amitriptyline and imipramine. Other indene derivatives with an N-alkylated morpholine ring were proved to have less effect on the uptake of either 5-HT or NE than was YM-08054-1. YM-08054-1 was the most potent among all of the tested antidepressants in the inhibition of reserpine-induced facilitation of convulsions as well as in the potentiation of reserpine-induced facilitation of convulsions as well as in the potentiation of 5-hydroxytryptophan (5-HTP)-induced syndromes in mice, though the inhibitory effect of this agent on reserpine-induced hypothermia was weaker than that of either amitriptyline or desipramine, suggesting relatively selective effects of YM-08054-1 upon 5-HT rather than NE uptake in vivo. Neither viloxazine nor iprindole potentiated the responses to 5-HTP. YM-08054-1 was devoid of peripheral anticholinergic activity and exhibited weak local anesthetic effect as well as low acute toxicity when compared with amitriptyline. The results indicate that YM-08054-1 has a novel profile as an antidepressant agent which is quite different from that of either viloxazine or tricyclic compounds.

Successive cleavage of N-terminal Arg1--Pro2 and Lys3-Pro4 from substance P but no release of Arg1-Pro2 from bradykinin, by X-Pro dipeptidyl-aminopeptidase.

X-Pro dipeptidyl-aminopeptidase (EC 3.4.14.1) purified homogeneously from the human submaxillary gland was proved to hydrolyze N-terminal dipeptide Arg1-Pro2 and subsequent dipeptide Lys3-Pro4 from substance P (Arg-Pro-Lys-Pro-Gln-Gln-Phe-Phe-gly-Leu-Met-NH2). Km and V values of hydrolysis of substance P were 2.0 mM and 3.6 mumol/min per mg protein, respectively. In contrast, the N-terminal Arg-Pro of bradykinin (Arg-Pro-Pro-Gly-Phe-Ser-Pro-Phe-Arg) was not cleaved by the enzyme.
