A unique phosphoprotein, which contains an uncommon amino acid, alpha-amino adipic acid (alpha-AAA), was isolated from unerupted bovine teeth by extensive EDTA demineralization. The alpha-AAA in the protein hydrolysates was identified based on the elution time on an amino acid analyzer using the sodium and lithium citrate buffer in a dual column system, and its identity was confirmed by comparisons of its DNS derivative with that of authentic alpha-AAA. This result suggests that the lysine residues in the phosphoprotein may be deaminated by an amine oxidase into allysine and further oxidized to alpha-AAA.
By the indirect immunofluorescence technique, IgM antibodies to the cell surface of an Epstein-Barr virus (EBV) producer cell line, P3HR-1, were detected in sera from infectious mononucleosis (IM) patients but not in sera from patients with Burkitt lymphoma or nasopharyngeal carcinoma nor in sera from healthy adult donors having antibodies to EBV-specific viral capsid antigen (VCA). Titers of the IgM antibodies were higher in the earlier stages of IM, a pattern similar to that for IgM antibodies to VCA. The IgM antibodies to the cell surface were identified as being those against the EBV-specific membrane antigen (MA) by the following criteria: (1) The antibodies were reactive to MA-positive cell preparations but to MA-negative cell preparations. (2) Titers of the IgM antibodies were not significantly affected after absorption of sera with sheep red blood cells which could completely eliminate heterophil antibodies in the same sera. Detection of the IgM antibodies to MA may have a particular diagnostic value for providing evidence of a recent EBV infection.
Forty-eight patients with a variety of primary renal diseases and systemic lupus erythematosus (SLE) were examined for the proportion of circulating T lymphocytes bearing receptors for IgM (T mu cells) or IgG (T gamma cells). Although the control group showed strikingly similar mean values for both T mu and T gamma cells, the whole group of patients with primary renal diseases and SLE showed a wide scatter of values. Sixteen patients with primary renal diseases and SLE had higher proportions of T gamma cells than the control group, whereas seven patients with chronic glomerulonephritis (CGN), membranoproliferative glomerulonephritis (MPGN), lipoid nephrosis (LN), and SLE showed very marked decrease in the proportions of T gamma cells in the peripheral blood. On the other hand, six out of the total group of patients had low proportions of T mu cells in the peripheral blood. However, no consistent relationship between the proportion of T mu and T gamma cells was found in our study. These findings indicate that there exists a heterogeneity of T-lymphocyte subpopulation distribution in some patients with primary renal diseases and SLE. The possible significance of these phenomena in the pathophysiology of renal diseases is discussed.
To determine whether initial sympatho-adrenal stimulation during stress was responsible for inducing a parasympathetic stimulation, the autonomic nervous activities were estimated in rats subjected to either restraint stress or restraint and water immersion stress. The parasympathetic and sympatho-adrenal activities were estimated functionally by measuring responses of gastric motility and heart rate, and also chemically by measuring catecholamine content in urine and tissues such as the heart and adrenals. Restraint and water immersion produced a definite parasympathetic predominance in contrast to the case of restraint alone. But both kinds of stress developed an increase in the sympatho-adrenal activity, and the extent of their increase was almost the same when evaluated on the basis of urinary catecholamine content. These findings indicate that initial sympatho-adrenal stimulation is not directly associated with parasympathetic stimulation. A marked fall in body temperature during stress was assumed to participate somewhere in the induction of parasympathetic stimulation.
In the experiment on continuous perfusion of rat stomach in vivo, genipin inhibited only the gastric acid secretion induced by carbachol, but not by tetragastrin, or histamine. In the experiment on isolated organs, genipin showed a weak competitive anti-acetylcholine action on the intestinal contraction. Based on these facts and results reported previously, it is conceivable that anti-cholinergic action at least partly contributes to the genipin-induced inhibitory effect on gastric functions. Erythritol clearance was increased with the increase in bile flow by administration of genipin. Genipin showed a significant choleretic action, and just then the concentration of biliary bile acid was decreased inversely. In the relationship between bile flow and biliary bile acid excretion rate, the slope of regression line obtained from genipin-treated group was not significantly different from that of control, and these lines were approximately parallel to each other. Genipin did not affect the concentration of sodium, potassium, chloride, or bicarbonate in the bile collected during the initial stage, in which bile flow was increased, after administration. It is concluded from these results that genipin-induced choleretic action proceeds by a mechanism wherein water is driven along osmotic gradient which originates in the transport of bile acid-independent fraction from hepatocytes into canaliculi, mainly through active Na+ transport.

In-vitro motility of the gallbladder was investigated in the circularly cut full thickness strips obtained from 19 mongrel dogs. The present study was undertaken to determine an appropriate size of the strips for observation and to know any difference, if exists, between motilities of the strips prepared from the neck, body and fundus. Recordings were made on a pen chart recorder using an isometric tension system. The results obtained were as follows. 1) The most appropriate size of the specimens to examine the changes of contractile dynamics was 5 mm in width and 15 mm in length. 2) The strips from three parts of the gallbladder showed similar rhythmic and tonic spontaneous contraction patterns.
The influence of inflammation on the motility of the human gallbladder was investigated through an in-vitro experiment. Motility of longitudinally cut strips from 50 patients were examined by the same method as described in our previous articles. We pathohistologically classified the severity of the inflammation of the specimens into four grades: grade 0 is normal, grade I, slightly inflamed; grade II, moderately inflamed; grade III, highly inflamed. Relationships between the grade of inflammation and the motility of the strips were summarized as follows. 1) In grade 0 cases, the strongest tonic contractions were observed in the strips from the fundus. This tendency was also found in cases of grade I and II but III inflammation. 2) The tonic contractions of the strips from the fundus or body decreased along with the grade of the inflammation. 3) In any strips, the frequency of the rhythmic contractions decreased progressively with the grade of the inflammation. 4) The amplitudes of rhythmic contractions of the strips with grade I and II inflammation were larger than those of grade 0 inflammation.

The target antigen for antibody-dependent-cellular cytotoxicity (ADCC) on Epstein-Barr virus-(EBV) carrying lymphoblastoid cells expressing EBV-specific membrane antigen (MA) were examined with human serum antibody and adult human peripheral lymphocytes as effector cells. These studies confirmed that anti-MA-positive but not MA-negative sera were reactive in the ADCC. The ADCC reaction was positive with cells in which the MA consisted of late (LMA) and early (EMA) components. These included 1) MA-positive cells prepared by EBV antigen-adsorption, 2) cells carrying de novo-synthesized MA without adsorbed MA, and 3) EBV-producer cells expressing MA spontaneously. In all these preparations, the target cells were lysed roughly in parallel with the frequency of MA-positive cells. Inhibition of LMA synthesis in EBV-superinfected cells by phosphonoacetate (PA) reduced ADCC sensitivity significantly and to a far greater extent than MA synthesis as measured by immunofluorescence. This suggests that a target for ADCC is the PA-sensitive LMA. No ADCC reaction occurred with the cell preparation comprised of a high percentage of MA-positive cells induced by 5-iodo-2'-deoxyuridine, which is believed to be EMA only. These results strongly suggest that the target antigen for ADCC in EBV-positive cells is a late MA but not early MA.

Microencapsulated mitomycin C (MMC) was utilized for arterial embolization of dog kidneys. Renal damage was evaluated by angiographic and pathologic examinations. The most extensive embolic necrosis was associated with the use of microencapsulated MMC compared to blank microcapsules, nonencapsulated MMC, and a combination of both. Arterial infusion of microencapsulated MMC kept the concentration of MMC low in the circulating blood and sustained the biological potential of MMC in the kidney for prolonged periods of time as compared with nonencapsulated MMC. Transcatheter infusion of microencapsulated MMC could be clinically applied to the therapeutic arterial embolization of the tumor-supplying vessels together with regional anticancer chemotherapy.
