Immunohistochemical localization of collagens in the ear.

The aim of this work was to prepare specific antibodies to various types of collagen in order to study on immunohistological localization of collagen under normal and pathologic conditions. Human type I and III collagens were extracted from human dura mater and placenta using limited pepsin digestion followed by differential salt precipitation and chromatographic purification. The purity of collagens was assessed by SDS polyacrylamide gel electrophoresis. Antibodies to type I and III were prepared in rabbits, and showed a high reaction with the corresponding human collagens, and these antibodies were tested to determine activity using ELISA. However, antibody to type I also showed some cross-reaction with type III, and antibody to type III, with type I. Using these antibodies, tissue distribution of collagen was examined in the middle ear by peroxidase anti-peroxidase method. In granulation tissue, it was revealed that types I and III co-existed.

[Effects of interferon and its inducers on neutrophil chemiluminescence].

In order to evaluate effects of interferon (IFN) and its inducers on neutrophil functions, neutrophil chemiluminescence (ChL) was assayed in 12 patients treated with IFN (human lymphoblastoid interferon, 3.0 X 10(6) units/day i.m. daily) or Ge-132 (2,250 mg/day p.o. daily). The peak levels of neutrophil ChL, assayed one week after the initiation of treatment, were increased in comparison to those before treatment, but one month after treatment they were decreased to pretreatment levels in spite of the daily administration of the agent. On the basis of these results, it was concluded that IFN and Ge-132 enhanced the host defence mechanism including the activation of neutrophils, which appeared at the early phase of the host reaction.

Underlying cause of death as recorded for multiple sclerosis patients: associated factors.

The coding of multiple sclerosis (MS) as underlying cause of death (UCD) on the death certificate provides an important epidemiologic resource for both descriptive and analytic studies. However, not all deaths among MS patients will be so coded. We investigated the effect on estimated occurrence of MS and on characteristics of MS patients when only UCD codes are used to identify cases. Of 2329 MS patients living in Los Angeles County (California) or King/Pierce Counties (Washington) in 1970, 438 had died by 1980. Only 53% of the deaths were coded to MS; 47% were attributed to other causes. Based on our comparisons, the use of only MS-coded deaths to describe decedents would: underestimate the age at MS onset; overestimate the female:male ratio; underestimate age at death; and underestimate duration of MS. Also, the percentage of MS-coded deaths decreased with lengthening duration of follow-up of these prevalent cases. The effect of using only UCD codes to report characteristics of decedents with other chronic diseases may well be similar.

Variations in 5'-nucleotidase activity in established mesenchymal cell lines from syngeneic A/J mice.

5'-Nucleotidase activity was analyzed in four different mesenchymal cell lines (F, m, e and SP) established from syngeneic A/J mice. The 5'-nucleotidase activity of fibroblasts was lower in transformed cells (F and m) than in nontransformed cells (e). An increase in cell contact during confluence or during high cell density increased 5'-nucleotidase activity, and a decrease in cell contact caused a decrease in 5'-nucleotidase activity in both fibroblastic (F, m and e) and reticulum (SP) cell lines. These results are evidence that 5'-nucleotidase activity in mesenchymal cells is influenced by intercellular contact as well as transformation.

Superoxide anion (O2-) production by neutrophils in refractory anemia with excess of blasts.

The O2- production by neutrophils was examined in 4 cases of refractory anemia with excess of blasts (RAEB) in order to evaluate the possible causes of enhanced susceptibility to infection and to gain some informations on the differentiation of neutrophils in this hematological disorder. In three of the four RAEB cases there was little O2- production by neutrophils, in addition to there being morphological anomalies of the neutrophils such as a Pelger-Hüet-like anomaly, granular deficiency and binucleated cells. These results suggest that the impairment of O2- production by neutrophils in RAEB is one of the possible causes of susceptibility to infection and also suggest that the differentiation of neutrophils in this hematological disorder is faulty. The estimation of O2- production by neutrophils may be a useful diagnostic method for preleukemia.

[Prolyl hydroxylase activity in rabbit ovary during ovulatory process].

The disruption of connective tissue and dissociation of collagen fibers that occurs in the ovarian follicle during mammalian ovulation is reportedly induced by collagenolytic enzymes. To further analyse the metabolism of ovarian collagen during the ovulatory process, prolyl hydroxylase activity was assayed because the quantity of this enzyme provides some indication of the relative amount of collagen fibrillogenesis in connective tissues. Following the induction of ovulation by hCG, prolyl hydroxylase activity increased in both the ovarian follicle and stroma. The amount of activity reached a maximum of about 300% above the control level at 13hr after hCG, i.e., 1-3hr after the estimated time of ovulation. The pattern of enzyme activity during the ovulatory process was identical in both the follicle and the stroma. The prolyl hydroxylase activity remained at more than 200% above the control level throughout the secretory phase. It is concluded that the rabbit ovarian follicle and stroma contain prolyl hydroxylase activity, and that this enzyme activity increases significantly during the ovulatory process and luteinization.

[Clinical effects of human lymphoblastoid interferon in patients with hematologic neoplasms].

Sixteen patients with hematologic neoplasms were treated with Human Lymphoblastoid Interferon (HL-BI) derived from Namalwa cell line. They were 6 multiple myeloma, 8 acute leukemia and 2 malignant lymphoma patients. All patients were previously treated with anticancer agents except one case with multiple myeloma. HLBI, 3.0 X 10(6) IU/day, was daily administered by intramuscular injection at least for 4 weeks. Three of 6 multiple myeloma responded to HLBI with a decrease of more than 25% in serum myeloma protein level. A case with pleural effusion due to massive infiltration of myeloma cells treated with intrathoratic administration of HLBI, in whom complete disappearance of pleural effusion was recognized. On the other hand, no patients with acute leukemia and malignant lymphoma responded except one case with acute lymphocytic leukemia, in which bone marrow lymphoblasts decreased transiently. Fever episodes, 13 of 16 cases, were more frequently seen but were manageable. Transient leukocytopenia and thrombocytopenia were also observed in 4 and 7 of 8 cases, respectively. No anaphylactoid reaction was seen. Thus, HLBI was expected useful in the clinical management of multiple myeloma.

Collagenolytic enzyme activity in human ovary: an ovulatory enzyme system.

Collagenolytic enzyme activities presumed to play an important role in ovulation were investigated in the human follicular apex, base, and granulosa cell layer throughout the ovarian cycle. Those analyzed were human ovarian collagenase, 2,4-dinitrophenyl-Pro-Gln-Gly-Ile-Ala-Gly-Gln-D-Arg-OH peptidase, N-carbobenzoxy-Gly-Pro-Gly-Gly-Pro-Ala peptidase, and alpha-N-benzoyl-DL-arginine beta-naphthylamide hydrolase. Collagenase activity was also measured in human granulosa cell cultures. The activities of all four enzymes showed a marked preovulatory decrease in the apex. The activities in the apex were slightly higher than those in the base throughout the ovarian cycle. However, the activities in the granulosa cell layer and collagenase activity in the granulosa cell cultures increased toward preovulation and decreased after ovulation. These findings suggest that collagenase is synthesized in the granulosa cells maximally at preovulation and is consumed in the follicular apex at the same time, resulting in collagen degradation and disruption of the follicular wall in collaboration with other collagenolytic enzymes investigated here.

Studies of body movements during night sleep in infancy.

Body movements (BMs) during night sleep of 25 neurologically normal infants, 11 premature and 14 full-term, whose ages ranged from 30 conceptional weeks to 18 months post-term, were examined to evaluate the changes of their features with age. The examinations performed during sleep periods totaled 65 times, 1 to 60 times on each subject. Through visual observation and EEG recordings, the BMs were classified into 3 types: (1) Gross movements (GM), (2) localized movements (LM), both of the above two lasting more than 0.5 second, and (3) twitch movements (TM) lasting less than 0.5 second. Total GM and LM time per hour of sleep, average duration of GM and LM and number of GM, LM and TM per hour of sleep were calculated. Percentage of 20 seconds epochs without BMs (nonbody-movement-epochs) was also estimated. These BMs parameters decreased with maturation to certain low base levels. However, each parameter showed a particular time course. TM decreased initially, then LM and lastly GM reached the base level around the age of 9 to 13 months. On the other hand, nonbody movement-epochs increased progressively until 8 months of age. These three types of BMs are considered to be controlled by the CNS with different organization levels, the simplest for TM and the most complicated for GM. They are thus correlated to the maturational process of the CNS, and could be good indicators for detecting normal and abnormal CNS developments.

[DNP-peptidase activity in the rabbit ovary (author's transl)].

To examine the possible roles of collagenolytic enzymes in the follicular wall at ovulation, we measured in rabbit ovaries collagenolytic enzymes by using synthetic substrates, DNP-Pro-Gln-Gly-Ile-Ala-Gly-Gln-D-Arg OH supposed to be specific for vertebrate collagenase. Our results clearly demonstrated that DNP-peptidase activity existed, with its optimal pH7.6-pH7.8 in rabbit ovarian follicles. DNP-peptidase activity steadily increased for nine hours following the hCG injection. After nine hours of enzyme activity decreased sharply until about the tenth hour, at which time ovulation had occurred. This pattern of enzyme activities indicates that the DNP-peptidase was consumed for degradation of collagen fibers. After collagen fibers have been completely degraded, this enzyme was increased again. These changes of DNP-peptidase activities at ovulation were similar to cathepsin B1 activities with its optimal pH6.0. It is concluded that DNP-peptidase could be one of ovulatory enzymes in the rabbit.
