Carcinoma arising in fibroadenoma: case report and review of the world literature.

To date 119 cases of "carcinoma arising in a fibroadenoma" have been reported in the world literature. A total of 120 cases, including the case described here, are analyzed. The mean age of the affected females was 43 years. A few patients complained of several disorders other than breast tumor before and during the development of the tumor. The tumor was most frequently located in the upper outer quadrant of the breast. Most carcinomas in fibroadenomas were lobular carcinoma in situ followed by duct carcinoma in situ. There was only one squamous cell carcinoma. The malignant process was confined to the fibroadenoma in over half of the cases. In some cases additional foci outside the fibroadenoma were reported. Radical mastectomy was the most frequent treatment. The prognosis was usually good.

Biosynthesis of various types of collagen by human hepatoma cells in vitro.

A cloned human hepatoma cell line (HH2-1) produced and formed collagen fibers in vitro. The relative rate of collagen synthesis by the cells was increased with an enhancement of the cell density. An analysis of the components of the collagen using sodium dodecyl sulfate-polyacrylamide gel electrophoresis revealed that the cells synthesized interstitial collagen, types I and III, and other collagenous proteins. Thus, human hepatoma cells may play an important role in the formation of stromal collagen in the tumor.

Mitogenic and polyclonal B cell activation activities of synthetic lipid A analogues.

The activation of murine B cells and macrophages by synthetic lipid A analogues, solubilized with triethylamine and complexed with bovine serum albumin, were investigated in vitro. The analogues used are nonphosphorylated, C-1 or C-4' monophosphorylated and C-1,4' diphosphorylated derivatives of beta-1,6-linked D-glucosamine disaccharide possessing both ester- and amide-bound fatty acid substituents. They were divided into 4 groups, A, B, C and D in terms of the fatty acid substitution. Ester- and amine-bound fatty acids of the analogues are both tetradecanoic acids (C14) in group A, C14 and (R)-3-hydroxytetradecanoic acids (C14-OH) in group B, both C14-OH in group C and C14-OH and (R)-3-tetradecanoyloxytetradecanoic acids in group D. Mitogenic activity was exhibited in spleen cells from C3H/HeN mice by the C-1 monophosphorylated analogues in groups A and B, and by the C-4' monophosphorylated analogues in groups B, C and D, but not in cells from C3H/HeJ mice. Nonphosphorylated analogues in groups A, B and C, and C-4' monophosphorylated analogue in group A showed negative mitogenic activity. Only the nonphosphorylated analogue in group D exhibited mitogenic activity in spleen cells from C3H/HeJ mice as well as those from C3H/HeN mice. None of the other analogues exhibited the activity in C3H/HeJ spleen cells. Polyclonal B cell activation activity was exhibited by the C-1 monophosphorylated analogues in groups A and B, and by the C-4' monophosphorylated analogues in groups C and D. Nonphosphorylated analogues in all groups and C-4' monophosphorylated analogues in groups A and B showed negative PBA activity. None of the analogues tested could induce any cytostatic macrophages from thioglycollate-elicited peritoneal macrophages.

Activation of peritoneal macrophages by berberine-type alkaloids in terms of induction of cytostatic activity.

The antitumour alkaloids, berberine and coralyne were investigated in terms of their ability to induce chemiluminescent responses and cytostatic activity in macrophages against tumour cells in vitro. Berberine hydrochloride and sulfate, and coralyne chloride, which had no direct tumoristatic activities at a concentration of less than 0.15 micrograms/ml, activated macrophages to inhibit the incorporation of tritiated thymidine into EL4 leukemic cells. The activation of cytostatic macrophages by berberine was not augmented by the addition of a suboptimal dose of macrophage activating factor (MAF). Furthermore, a marked chemiluminescent response of macrophages was induced by treatment with berberine as well as with MAF or lipopolysaccharide. These results show that berberine and coralyne are potent macrophage activators for inducing cytostatic activity against tumour cells in vitro.

Effects of prolyl hydroxylase inhibition on arterial collagen synthesis and blood pressure in hypertensive rats.

Changes in arterial collagen synthesis and the effects of P-1894B, a potent inhibitor of prolyl hydroxylase, were investigated on rat hypertension, induced by deoxycorticosterone acetate (DOCA) and salt. In DOCA-salt hypertensive rats, prolyl hydroxylase activity increased significantly in the abdominal aorta and in the mesenteric artery. Incorporation of 14C-proline into the thoracic aorta, abdominal aorta and mesenteric artery was higher and the hydroxyproline content of the abdominal aorta and mesenteric artery was higher than in the control rats. Treatment with P-1894B significantly inhibited prolyl hydroxylase activity, reduced arterial collagen synthesis, but did not prevent or reverse hypertension.

The apical structure in Perophora annectens (tunicate) spermatozoa: fine structure, differentiation and possible role in fertilization.

The apical structure in Perophora annectens spermatozoa is approximately 4 micron in length and it is helically coiled. Its major component is a striated structure, which may be analogous to a perforatorium. The plasmalemma enclosing the anterior quarter of the apical structure is covered by extracellular materials, the anterior ornaments. During spermiogenesis, the apical structure is first recognized as a small blister of the plasmalemma at the apex of the young spermatid. It develops into a conical protrusion and then into a finger-like process (approximately 1 micron in length). This process is transformed into an elongated process (approximately 4 micron in length) with electron-dense material in its core. Finally, the elongated process is helically coiled to form an apical structure in which electron-dense material forms dense striations. Vesicles (50-70 nm in diameter), presumably derived from the Golgi apparatus, have been recognized in the blisters of younger spermatids, and can be followed through to the finger-like process. In the finger-like process these vesicles are transformed into smaller vesicles (20-30 nm in diameter), which probably fuse with the anterior plasmalemma of the finger-like process. This suggests that chorion lysin(s) is associated with the anterior membrane enclosing the apical structure in these spermatozoa.

Facial nerve neurinoma: two cases located in the horizontal portion.

Two small facial neurinomas located in the horizontal portion were presented to demonstrate the clinical features, diagnostic evaluation, and surgical treatment. These two cases show the early state of facial neurinoma and suggest the mechanism of facial paralysis by facial neurinoma. Facial nerve decompression is recommended for Bell's palsy unresponsive to the conservative therapy and recurrent facial palsy.

Interaction between human lymphoblastoid interferon and chemotherapeutic agents in vitro.

The combined effect of human lymphoblastoid interferon (HLBI) and anticancer agents on the growth of MOLT-4 was studied in vitro. The interferon showed a strikingly synergistic interaction in combination with aclarubicin, cytosine arabinoside or prednisolone. It was moderately synergistic in combination with adriamycin or 5-fluorouracil and tended to show additive effects with daunorubicin or vincristine. In vitro studies of combination chemotherapy with interferon and anticancer agents should yield valuable information as to the best combination for man.

[Collagen synthetic activity in rabbit ovary during ovulation and its blockage by indomethacin].

In order to investigate the possibility of the participation of prostaglandins (PGs) in the collagen synthetic activity during the ovulatory process, we measured prolyl hydroxylase (PH) activity and the hydroxyproline (Hyp) content of ovarian follicles in rabbits after the administration of hCG and indomethacin (IM). The minimum dose of IM (4mg/kg) effective in blocking ovulation was given simultaneously with hCG to mature female Japanese white rabbits. PH activity in the ovary reached its peak level at 13hr after the hCG injection, immediately after ovulation. This increase was not observed in other tissues such as skin, aorta or ear obtained from ovulating rabbits. In the IM-treated rabbits in which ovulation was blocked, PH activity was rather suppressed and did not show any prominent peaks, although IM did not directly suppress PH activity in the in vitro assay system. The amount of Hyp did not show any significant changes during the ovulatory process or during ovulation blockage by IM. It is suggested that PGs play an important role in controlling collagen synthetic activity of the ovary through the ovulatory process.

[Effect of indomethacin on collagenolytic enzyme activities in rabbit ovary].

To investigate the possible participation of prostaglandins in the activation of collagenolytic enzymes of the follicular wall at ovulation, we measured the activities of neutral and acid collagenase in the rabbit ovaries at various stages of follicular development by using synthetic substrate DNP-Pro-Gln-Ile-Ala-Gly-Gln-D-Arg OH (DNP peptide) with its optimal pH 7.6 and alpha-N-benzoyl-DL-arginine-2-naphthylamide HCl (BANA) with pH 6.0, and the effect of ovulation-blocking doses of indomethacin (4 mg/kg) on DNP peptidase and BANA hydrolase activities were investigated. DNP peptidase and BANA hydrolase activities were increased toward ovulation with the highest level 7 to 9hrs after the hCG injection and then decreased significantly at 10hr. At 11hr, around the time of ovulation, the activity stayed at its low level, then rose by 13hr. Following the concomitant administration of IM with hCG, ovulation was blocked and the preovulatory increases in DNP peptidase and BANA hydrolase activities were not observed and their activities stayed at the low level until 20hr. It is suggested that collagenolytic activity for the ovulatory process started to intensiby 6hrs and ended 3hrs prior to ovulation and PGs are necessary for the enzymatic activation of DNP peptidase and BANA hydrolase.
