Uptake of estrone and estrone-3-sulfate by lung of macaca fuscata.

An equimolar mixture of 3H-E1-S2 and 14C-E1 was injected in one shot into the inferior vena cava near the heart of female Japanese monkey. Following the injection, blood was collected from the aortic arch at intervals of 15 s over a period of 10 min. The concentration of radioactivities in the whole blood and serum was measured. The metabolites were analyzed by DEAE-Sephadex A-25 column chromatography, enzyme hydrolysis, thin layer chromatography and paper chromatography. Both radioactivities of 3H-E1-S and 14C-E1 rapidly decreased in the first 90-s serum sample. The 3H/14C ratio in the 0-15-s serum sample was 5 times higher than the initial ratio of injected compounds. The radioactivities in the serum gradually decreased after 90 s of injection. The 3H/14C ratio in the pulmonary tissue was very low after collecting the final blood sample. This result shows that the most of 3H-E1-S passed through the lung and the larger part of 14C-E1 remained in the lung following injection of these materials. So, it is probable that E1-S is in a form to be carried in the general circulation.

Some properties of acid lipase in the defatted liver from the laying hen (Gallus domesticus).

Acid lipase activity was found in the defatted liver from the laying hen, but little neutral or alkaline lipase activity was observed in the liver. Most of acid lipase was in the insoluble fraction of the defatted liver, and the enzyme was solubilized by sonication at 9 kHz for 50 min in a slightly alkaline solution. The lipase showed its maximum activity at pH 5.0, 38 degrees C. It was stable below 40 degrees C and over the pH range from 4.0 to 9.0. Detergents, serum of the laying hen and the soluble fraction from the defatted liver homogenate from the laying hen markedly inhibited the lipase activity. The lipase solubilized by sonication was large in molecular mass, suggesting that the preparation formed colloidal particles.

Retrovirus-induced expression of interleukin 2 receptors on cells of human B-cell lineage.

We provide direct evidence for interleukin 2 receptor (IL2R) induction by human T-cell leukemia/lymphoma virus (HTLV) in human B-cell lines. A lymphoblastoid B-cell line (LCL-Ter) was established by Epstein-Barr virus-induced transformation of peripheral blood lymphocytes derived from a healthy HTLV carrier and cloned in vitro. HTLV gp21 and/or p19 antigens were detected in eight LCL-Ter clones, all of which also expressed the IL2R antigens Tac and Hiei (defined by monoclonal antibodies). However, five other LCL-Ter clones, which were negative for the HTLV antigens, did not express the IL2R antigens. Furthermore, when the IL2R-negative B-cell line LCL-Kan, derived form a normal donor, was cocultured with HTLV-producer cells, three HTLV-carrying clones were obtained and found to constitutively express IL2R. IL2R induced by HTLV on these B-cell lines bound recombinant interleukin 2 and were similar in apparent molecular mass (approximately equal to 60 kDa) to the IL2R of peripheral blood lymphocytes stimulated with phytohemagglutinin.

Two replication determinants of an antibiotic-resistance plasmid, pTB19, from a thermophilic bacillus.

Two different replication determinants were found on an antibiotic resistance plasmid, pTB19, from a thermophilic bacillus. One replication determinant (designated RepA) was functional only in Bacillus subtilis, whereas the other (designated RepB) functioned in both B. subtilis and Bacillus stearothermophilus. A deletion plasmid, pTB90, carrying the RepB derived from pTB19 coincidentally contained the specific 1.0 MDal EcoRI fragment of a cryptic plasmid pBSO2 from B. stearothermophilus. The presence of this 1.0 MDal EcoRI fragment in various deletion plasmids from pTB90 increased transformation frequencies for B. stearothermophilus 10(3) to 10(4) times and lowered plasmid copy numbers in the host strain to about one-tenth of those found for plasmids lacking this fragment.

Thanatophoric dysplasia.

Two autopsy cases of thanatophoric dysplasia in male newborn infant are described. They have markedly short-limbs and narrow thoraxes. Histologically, generalized disruption of endochondral ossification with normal resting cartilage was seen at the epiphyseal region of femur. Differential diagnosis from other 3 types of lethal chondrodysplasia is described and the histogenesis of thanatophoric dysplasia is considered with a survey of the literature.

Effect of taxol on the interaction of tubulin with myosin filaments.

The interaction between polymerized tubulin from porcine brain and myosin from rabbit skeletal muscle was examined. The addition of myosin to the solution of tubulin polymerized by taxol resulted in a remarkable increase in turbidity within a few minutes at 37 degrees C, and a dense and stable precipitate was formed. The maximal molar ratio of tubulin bound to myosin was calculated to be about 4, while the value was about 2 when 6S tubulin was used. Both podophyllotoxin and colchicine suppressed the taxol-dependent increase of the binding of tubulin to myosin, but only when they were preincubated with tubulin prior to addition of taxol. 6S tubulin inhibited with actin-activated Mg2+-ATPase activity of myosin, and polymerized tubulin inhibited the Mg-ATPase more than 6S tubulin. Dense precipitates of tubulin and myosin were observed by thin-section electron microscopy. Microtubules were observed to be entangled in myosin filaments and single microtubules were occasionally surrounded by five myosin filaments in a cross section, similar to actin-myosin arrays in muscle. After incubation of tubulin with myosin, taxol was able to induce tubulin polymerization in the same way as it polymerized microtubules in the absence of myosin.

New acute peritoneal dialysis technique: wire-guide insertion and long-term indwelling of peritoneal catheter.

This paper presents the application of wire-guide insertion of peritoneal dialysis catheter. 30 patients who required acute peritoneal dialysis were treated using a total of 37 peritoneal catheters with the wire-guide method. The catheters were left in place as long as possible (periods totalling 644 days in all). The complication of insertion was minor bleeding with blood-tinged dialysate that tended to clear spontaneously in a few days. Leakage occurred in 17 indwelling catheters but did not occur on the days of insertion. There were 3 episodes of clinical peritonitis, so the infection rate was 1 episode every 7 patient-months. This technique is safer and simpler, i.e. the incidence of peritonitis is lower in long-term indwelling catheters, and so it will contribute to the wider and earlier utilization of peritoneal dialysis.

Phosphorus-free antihypertensive lipid from dog peritoneal dialysate, Peritoneal Dialysate Depressor-II.

A phosphorus-free lipid with a transient antihypertensive effect was found in a dog peritoneal dialysate by a modified procedure. On silicic acid chromatography of the crude extract, a new hypotensive factor and a new hypertensive factor were separated besides the known antihypertensive phospholipid resembling lysolecithin (Peritoneal dialysate depressor-I). The new antihypertensive factor was purified almost completely by gel filtration on a Sephadex LH-20 column, and found to contain no phosphorus. It was tentatively named Peritoneal Dialysate Depressor-II. The main spot detected on preparative thin-layer chromatography by charring with ethanolic sulfuric acid reagent was found to be responsible for the antihypertensive action. This factor caused a sharp decrease in systemic arterial blood pressure of rats, cats and guinea pigs when injected into the femoral vein. There was no significant change in the heart rate during this hypotension. The hypotensive activity was not affected by previous treatment of the rats or guinea pigs with antimuscarinic, antihistaminic, beta-adrenergic-blocking or ganglionic-blocking agents. The antihypertensive effect of the factor was unaffected on rats pretreated with yohimbine, indomethacin or theophylline. In spinal or reserpinized rats, no detectable reduction of the depressor activity was observed. The factor did not affect isolated guinea pig ileum. Its depressor activity was not changed significantly by its treatment with proteases. No prostaglandin E or F series compounds were detected in the preparation by fluorometric assay with 15-hydroxyprostanoate oxido-reductase and resazurin. The hypotensive factor caused no aggregation of cat or rabbit platelets.(ABSTRACT TRUNCATED AT 250 WORDS)

Estrogen biosynthesis in human liver--a comparison of aromatase activity for C-19 steroids in fetal liver, adult liver and hepatoma tissues of human subjects.

After incubation of various tritiated C-19 steroids (androstenedione, testosterone, dehydroepiandrosterone, dehydroepiandrosterone sulfate) with human fetal liver, adult liver and hepatoma tissue homogenates, estrone, estradiol and estriol were analysed after a series of purification steps involving column chromatography, thin layer chromatography and co-crystallization. The findings indicated that the human fetal liver extensively aromatized various C-19 steroids to estrogens, whereas human adult liver and hepatoma tissues exhibited little or no aromatase activities. The formation of estradiol from androstenedione in human fetal liver indicated the presence of 17 beta-hydroxysteroid dehydrogenase in this tissue. It was therefore concluded that although the liver participated in the aromatization process during the fetal stage, extensive aromatization did not take place in the adult liver.

Lack of promotive effect of quercetin on methylazoxymethanol acetate carcinogenesis in rats.

Promoting effect of quercetin in the intestinal tract was examined in Sprague-Dawley strain rats. The rats were injected with methylazoxymethanol acetate (MAM) in saline solution at 25 mg/kg body weight once a week for 3 weeks and fed a diet containing 1% quercetin for the following 459 days. Most tumors found in this experimental group were also found in the group injected with MAM alone and there was no significant difference in tumor incidence between these two groups. No tumors were found in the group fed the 1% quercetin diet alone. Thus, quercetin was shown to lack of promoting activity on MAM-induced intestinal carcinogenesis in Sprague-Dawley rats.

Gated cardiac imaging using a continuously rotating CT scanner: clinical evaluation of 91 patients.

To produce electrocardiographically (ECG)-gated computed tomographic (CT) images of the heart, a post-data-acquisition ECG correlation technique was used in which data for missing angular projections are derived from the original scan data to complete 360 angular projections. Improved image quality and clinical usefulness were demonstrated compared with routine nongated CT and two-dimensional echocardiography. Gated CT was better than nongated CT in 26 of 41 positive and three of five negative cases of suspected myocardial infarction, four of 10 positive and one of 12 negative cases of suspected left atrial mass, three of 10 cases with pericardial fluid collection, and three other cases. Compared with echocardiography, CT was of additional value in eight of 10 cases of myocardial infarction, five of nine positive and one of 10 negative cases of suspected left atrial mass, four of 10 positive and one of three negative cases of suspected pericardial fluid collection, and two other cases. The equipment required for CT gating is of low cost, but the examination time is lengthy and less conveniently performed than echocardiography. However, when echocardiography is indecisive or suspected to be falsely negative, gated CT imaging of the heart is recommended.

A monoclonal antibody that defines p24, a core protein of adult T-cell leukemia virus, and its precursor.

A mouse hybridoma cell line, H15, produced monoclonal antibody reacting with all the adult T-cell leukemia (ATL) virus (ATLV)-bearing cell lines but none of the ATLV-negative cell lines tested. Binding of H15 antibody to ATLV-bearing cell surfaces was specifically blocked by anti-ATLV positive human sera. Radioimmunoprecipitation analyses revealed that the antigen detected by H15 antibody was p24, a core protein of ATLV. Pulse-chase experiments using H15 antibody led to the identification of a protein, p53, which could be a precursor of p24.

Human retrovirus-induced IL-2 receptors and their possible role in transduction of continuous cell growth signal.

The first half of this report describes direct evidence for the interleukin 2 receptor (IL-2R)-inducing ability of human T-cell leukemia/lymphoma virus (HTLV). When an Epstein-Barr virus (EBV)-transformed human B-cell line, LCL-Kan was infected with HTLV or transfected with plasmid DNA containing defective HTLV genome (long terminal repeat (LTR)-gag-pX-LTR), LCL-Kan cells were altered to express IL-2R which was indistinguishable from that of normal activated T cells. The second half of this paper reports that IL-2 could inhibit proliferation of HTLV-carrying T-cell lines which were spontaneously immortalized from IL-2-dependent cells or peripheral blood cells containing ATL leukemia cells. These results suggest that expression of IL-2R may be induced by HTLV in HTLV-transformed cells, where the IL-2R plays an important role in transduction of continuous cell growth signal.

[Combination chemotherapy consisting of cis-platinum and adriamycin in non-small cell carcinoma of the lung].

Twelve patients with unresectable and recurrent non-small cell carcinoma of the lung were treated with a combination chemotherapy consisting of cis-platinum and adriamycin. The overall response rate was 42% with two complete responses and three partial responses. The durations of response were 9+ and 8.5+ months in two complete responses, and 7.5+, 5.5+ and 2.5 months in three partial responses. The overall median survival was 8 months ranging 3.5+ to 16+ months. Three patients had leukopenia less than 2,000/mm3, two patients had thrombocytopenia less than 100 X 10(3)/mm3, and four patients had hemoglobin decrease of 3 g/dl or more. Severe nausea occurred in nine patients, whereas vomiting did in seven patients. Nearly total alopecia was observed in all patients. Renal toxicity in four patients was also observed, but was reversible. We believe that the combination chemotherapy with cis-platinum and adriamycin is promising in the management of patients with non-small cell carcinoma of the lung.
