Glycosaminoglycans and acidic glycoproteins in rabbit uterus under estrogenic conditions.

Uterine slices obtained from the estrogen-treated rabbits were digested with pronase. Glycosaminoglycans and acidic glycopeptides were then isolated by Dowex 1 column chromatography and preparative electrophoresis on cellulose acetate membrane (Separax), in succession. Each subfraction thus obtained was identified by the mobility on Separax electrophoresis and the digestibility with mucopolysaccharidases (Streptomyces hyaluronidase, testicular hyaluronidase, chondroitinase AC, chondroitinase ABC and heparinase). The resulting data showed that each complex saccharide (hyaluronic acid, heparan sulfate, chondroitin sulfate A, chondroitin sulfate C, dermatan sulfate, sulfated glycopeptide and sialoglycopeptide) was separated into 2-5 fractions, indicating charge and/or molecular heterogeneity of each complex saccharide.

Isolation and characterization of a sulfated glycoprotein from rabbit uterus.

Crude glycoproteins were extracted with 0.15 M NaCl from the pooled endometrial scrapings of rabbit uteri after treatment with estrogen. The crude glycoproteins were fractionated with ammonium sulfate, followed by DEAE-cellulose column chromatography, treatment with CM-Sephadex C-25 and gel filtration on Sephadex G-200. Subsequently, purification of an acidic glycoprotein was carried out by gel filtration on Sephadex G-200 and then Sepharose 4B. The results of electrophoresis and enzymatic digestion, together with analytical data and the infrared spectrum indicated that the acidic glycoprotein was a sulfated glycoprotein.

Prinzmetal's variant form of angina pectoris. Re-evaluation of mechanisms.

Thirty-five patients with typical Prinzmetal's variant angina were studied by coronary cineangiography. There was no demonstrable stenosis of the major coronary arteries in 19 patients. Nine patients with single coronary stenosis underwent aortocoronary bypass and had recurrence of the symptoms postoperatively. Administration of nifedipine effected complete cessation of the symptoms among patients formerly treated medically. Although surgical treatment did not effect permanent relief of pain, all patients initially treated surgically experienced relief of pain when nifedipine was administered. The pathophysiology of variant angina remains obscure. Our results suggest that neurohumoral factors exert more of an effect on the myocardial cell than on the coronary vessels.

On acid glycosaminoglycans (mucopolysaccharides) in pleural effusion.

The concentration of acid glycosaminoglycans (mucopolysaccharides) in pleural fluid was found to be in the range of 4.0 to 60.3 mug per ml. By chromatographic analysis and by electrophoresis of the eluates, in combination with enzymatic digestion, it was found that pleural fluid contained protein-free hyaluronic acid, protein-bound hyaluronic acid, undersulfated chondroitin sulfate A(C), chondroitin sulfate A(C), protein-bound chondroitin sulfate A(C), and dermatan sulfate. The composition of acid glycosaminoglycans in pleural fluid seems to reflect the metabolism of the pleural cavity.

Biochemical characterization of hyaluronic acid from a case of benign, localized, pleural mesothelioma.

The tissue of a benign, localized, pleural mesothelioma was digested with protease, and the crude polysaccharide was fractionated by a column of Dowex-1 (Cl-form). The eluate from the column was electrophoresed and incubated with various mucopolysaccharide lysases. Based on the results of column chromatography, electrophoresis, and enzymatic digestion, it was found that hyaluronic acid was the major constituent of the glycosaminoglycans in pleural mesothelioma. The hyaluronic acid from pleural mesothelioma seemed to be identical in structure with that from human umbilical cord; however, the hyaluronic acid from mesothelioma was eluted before that from human umbilical cord when fractionated by the column of Sepharose 4B, suggesting a differnence in molecular size between the two. Also, evidence was obtained for the presence of dermatan sulfate and heparan sulfate.

Comparative metabolic study of nimetazepam and its desmethyl derivative (nitrazepam) in rats.

1. Nimetazepam distributed more rapidly in the brain than its desmethyl derivative (nitrazepam). The brain concentration of the active metabolites of the former was about twice that of the latter at 1h after oral administration. 2. At least four kinds of reactions were involved in the biotransformation of nimetazepam and its desmethyl derivative (nitrazepam) : (i) demethylation at N-1, (ii) hydroxylation at C-3, (iii) reduction of the nitro group at C-7 to the amino group and (iv) subsequent acetylation of the amino group. 3. The 1-N-demethylation of nimetazepam was slow compared with the other three reactions. 4. Nimetazepam was rapidly hydroxylated at C-3, while the 3-hydroxylation of its desmethyl derivative (nitrazepam) was very slow. 5. The reduction of the nitro group at C-7 and subsequent acetylation were important routes for the excretion of these drugs.
