Metabolic rate of acidic complex saccharides in rabbit uterus under estrogenic condition.

Uterine slices obtained from estrogen-treated rabbits were incubated in vitro with N-acetyl-D-[1-3H]glucosamine together with D-[U-14C]glucose. The isotope-labelled acidic complex saccharides were then isolated by pronase digestion, Dowex 1 column chromatography and preparative electrophoresis on cellulose acetate membrane, in succession. In this way, individual acidic complex saccharides (hyaluronic acid, heparan sulfate, chondroitin sulfate A, chondroitin sulfate C, dermatan sulfate, sulfated glycopeptide, and sialoglycopeptide) were separated into 2-5 subfractions. The specific radioactivity of hexosamine in the subfractions indicated that the metabolic rate of the uterine complex saccharides as follows: hyaluronic acid greater than sulfated glycopeptide greater than heparan sulfate greater than chondroitin sulfate C greater than dermatan sulfate. In addition, metabolic heterogeneity of heparan sulfate, chondroitin sulfate A, chondroitin sulfate C, and dermatan sulfate was suggested.

Proteochondroitin sulfate A in the euglobulin fraction of human plasma.

Euglobulin fraction of human plasma was extracted with 0.01 M phosphate buffer (pH 7.3) (PB) containing 1 M NaCl. The substances in the extract were separated by DEAE-cellulose column chromatography into two uronic acid-containing fractions (0.5 Fr and 0.75 Fr). The major fraction (0.75 Fr) was eluted at the void volume in gel filtration on Sephadex G-200, yielding 0.75-G Fr. 0.5 Fr and 0.75 Fr and their pronase digestion products (0.5-P Fr and 0.75-P Fr) together with 0.75-G Fr were examined by electrophoresis on cellulose acetate membrane. The results of electrophoresis and the digestibility with mucopolysaccharidases of these fractions indicated that 0.5 Fr and 0.75 Fr contained proteochondroitin sulfate A. It is suggested that chondroitin sulfate A in 0.5 Fr might be less sulfated than that in 0.75 Fr. Furthermore, analytical data and the infrared spectrum of 0.75-G Fr supported the above view.

Biosynthesis of sulfated glycoprotein in the endometrium of rabbit uterus.

The endometrial scrapings obtained from the uteri of estrogen-treated rabbits were incubated with N-acetyl-d[1-3H]glucosamine and [35S]sulfate, and then the incubation medium (M-Fr) was separated from the tissue. The tissue was subsequently homogenized exhaustively in 0.25m sucrose, and the insoluble residue (R-Fr) was separate. The supernatant at 8,500Xg for 10 min of the homogenate was subjected to subcelular fractionation by discontinuous sucrose gradient ultracentrifugation, and a thiamine pyrophosphatase-rich fraction (g-fr) was obtained. Complex carbohydrates were then separated from M-Fr, R-Fr, and G-Fr. The radioactivities incorporated into these complex carbohydrates suggested that sulfated glycoprotein synthesized in G-Fr was secreted into M-Fr. In order to confirm the above observation, labelled sulfated glycoprotein was isolated from the incubation medium. Subsequently, N-ACETYL-D[1-3H]glucosamine was incorporated into N-acetylglucosamine residues and [35S]sulfate into sulfates located most probably at the 6-position of N-acetylglucosamine residues of sulfated glycoprotein.

A 4-year survival rate after cardiac pacing in A-V block in the heart institute of Japan.

The long-term prognosis after cardiac pacing in A-V block was assessed by a follow-up study of 59 patients in whom pacing had been introduced 4 years or more before analysis. The 1 year survival rate was 85% and the 4 year survival rate was 71%. These figures are substantially better than the survival rate reported in patients not paced. The mortality has been higher in patients older than 70 years old. Even though patients were reviewed monthly or every 3 months in a pacemaker clinic, there were 3 sudden deaths in 4 years.

Comparative metabolic study of nimetazepam and its desmethyl derivative (nitrazepam) in dogs.

1. Blood levels of nimetazepam after oral administration to dogs were relatively low at early periods after dosage and reached peak levels (7-7-7-9 mug equiv./ml) after 8 h. The time course of blood levels was similar after oral administration of its desmethyl derivative (nitrazepam) to dogs. Blood levels of the latter, however, were low compared with nimetazepam and reached a peak (5-2-6-3 mug equiv./ml) after 4 h. 2. Recoveries of nimetazepam in urine and faeces were 46 and 52% of the dose for 0-24 h, 27 and 34% for 24-48 h and 4 and 6% for 48-72 h, while those of its desmethyl derivative (nitrazepam) were 63 and 71% for 0-24 h, 12 and 21% for 24-48 h and 2 and 3% for 48-72 h. 3. At least four kinds of reaction were involved in the biotransformation of nimetazepam and its desmethyl derivative (nitrazepam): (i) demethylation at N-1 (ii) hydroxylation at C-3, (iii) subsequent glucuronic acid conjugation of 3-hydroxy derivatives and (iv) reduction of the nitro group at C-7 to an amino group. Reaction (i) proceeded very rapidly in dogs, so that the blood metabolites of nimetazepam were closely similar to those of nitrazepam. For both drugs, the major blood metabolite was nitrazepam. Reaction (ii) was rapidly followed by reaction (iii), and glucuronides were predominantly excreted in urine. Reaction (iv) as well as reaction (iii) are important in the excretion of both drugs. The subsequent acetylation of 7-amino group, however, did not occur in dogs as it did in mice and rats.

Clinical and histopathological studies on spotted grouped pigmented nevi with special reference to eccrine-centered nevus.

Clinical and histopathological studies on 16 cases of spotted grouped pigmented nevus resulted in this disease being classified into at least three types. Characteristic findings of each type are described in this paper, in particular that nevus cell proliferation, a finding common to each type, is closely related pathogenetically to skin appendages, especially with eccrine sweat ducts. The eccrine-centered nevus characterized by Mishima corresponds to the third type, according to the present author's classification. The similar characteristic histopathological finding was also observed in a specimen taken from a black papule in a case of giant pigmented nevus with disseminated pigment cell nevi. We would like to consider that the cells of eccrine-centered nevus are derived from nevoblasts present in the walls of sweat ducts.

Mechanism of recurrence of pigmented nevi following dermabrasion.

A study was made of pigment freckles which recur after skin abrasion of spotted grouped pigmented nevi, according to the lapse of time, chiefly by means of the fluorescence method of Falck and Hillarp. The mechanism of recurrence is summarized as follows. Nearly simultaneously with the epidermal regeneration, dendritic melanin-producing cells derived from hair follicles and eccrine sweat ducts appear in the basal and prickle-cell layers of epidermis. These cells then create junction activity in the basal layer of epidermis, in the hair follicles and in the eccrine sweat duct walls. Finally, these nevus cells drop off into the underlying layer of scar tissue along the epidermal appendages. The dendritic melanin-producing cells seen in the early regenerative pigment freckles were thought to be incompletely differentiated nevus cells in the pre-stage of junction nevus formation. Appearance of dendritic melanin-producing cells and formation of junction activity in the eccrine sweat duct walls suggest the following possibilities. (1) In the cases of spotted grouped pigmented nevus which we studied, nevus cell proliferation also occurred eccrine-centrically on pathogenesis. (2) An important role is played by the eccrine sweat ducts in the recurrence of ordinary pigmented nevi after incomplete removal. (3) Eccrine-centered nevus cells are derived from nevoblasts in the eccrine sweat duct walls.

[Some evaluations of the Widmark method (author's transl)].

Some evaluations were made on the Widmark method is designed for determination of blood ethanol concentration. A reagent of potassium dichromate-sulfuric acid mixture was found potent within 3 weeks after preparation when kept air-tight in hard glass ware without exposure to light, and 0.01 N sodium thiosulphate reagent was potent within 2 weeks under similar conditions. No change was observed in the potassium dichromate-sulfuric acid mixture reagent even after heating at 50-60 degrees C for 2 hours. Heating for distillation and oxidation of ethanol was satisfactory with air bath in an electric thermostat.

[Forensic significance of the fate of ingested ethanol (author's transl)].

Distribution and disappearance of ethylalcohol (ethanol) were studied using either human blood in vitro or blood sampling or corpse blood after premortal ethanol ingestion. 1) Ethanol is distributed among plasma and blood cells according to a partition law. The partition coefficient was found to vary depending upon ethanol concentration and corpse phenomenon post mortum. 2) Ratio of plasma ethanol concentration to blood cells increases below 0.5 mg/ml and decreases above 0.5 mg/ml of ethanol concentration in the whole blood. The ratio was found to be more smaller in the corpse blood. The concentration ratio in blood cells to whole blood varies in a similar fashion. 3) Accuracy of determination of ethanol levels in the corpse whole blood was not always satisfactory, partly because distribution of ethanol in the blood varies postmortum and partly because ratio of plasma to blood cells also varies. Some attempts were made using statistical manipulations of hematocrit values or ratios of plasma to whole blood, to correct the errors without recommendable success. 4) After deliberabe evaluation of errors, possibly incurred when the value post mortum is used instead of the premortal values, it is concluded that the postmortal value can be used as an indicator of premortal ebriety within the reasonable limit of errors. 5) No changes in the distribution of ethanol were observed within 24 hours when the blood is kept in ice box immediately after sampling, and also in the blood ethanol concentration for at least 20 days under similar conditions. In contrast, blood samples, if left at room temperature, deteriorate rapidly within 5 days.
