Bradykinin-induced transient accumulation of inositol trisphosphate in neuron-like cell line NG108-15 cells.

Studies were undertaken to further elucidate the mechanism(s) by which bradykinin-dependent phosphoinositide metabolism takes place in neuroblastoma X glioma hybrid NG108-15 cells [(1984) J. Biol. Chem. 259, 10201-10207] using [3H]inositol-labelled cells. Bradykinin produced net increases in the level of [3H]inositol phosphates, especially of [3H]inositol trisphosphate which is formed transiently and most rapidly. The results indicate that bradykinin activates a phosphodiesterase to break down phosphatidylinositol 4,5-bisphosphate, generating two recently recognized intracellular messengers, 1,2-diacylglycerol and inositol trisphosphate.

Characterization of the spontaneous DNA-synthesizing and/or IgG-secreting cells in peripheral blood from patients with systemic lupus erythematosus.

Characterization of the cells responsible for spontaneous DNA synthesis and/or IgG secretion in systemic lupus erythematosus (SLE) was undertaken by fractionation of the peripheral blood mononuclear cells (PBM). Each fraction was analyzed for its capacity to incorporate [3H]thymidine [( 3H]TdR) and secrete IgG without mitogen. The non-E rosette-forming cell (non-ERRC) fraction, which consisted of the surface immunoglobulin-positive [sIg(+)] cells and null cells, revealed a markedly increased spontaneous DNA synthesis (620.0 +/- 586.9 cpm) during the first hour of culture and an elevated spontaneous IgG secretion (8639 +/- 2630 ng/ml) during 9 days of culture. Of particular interest was the finding that both increased responses were conducted by the null cells; the null cell population had approximately a fourfold relative increase of [3H]TdR incorporation and a 60-fold relative increase of IgG secretion compared with the sIg(+) cell population. These results suggest that SLE patients have a small population of preactivated B-cell lineage cells, which lack sIg or have a very low density of sIg.

Dynamic changes of the chicken erythrocyte membranes induced by N-methyl-N-aryl-N-nitrosoureas and their tumorigenicity.

The dynamic changes of chicken erythrocyte membranes induced by a series of N-methyl-N'-aryl-N-nitrosoureas (I X) were investigated in comparison with those of N-methyl-N-nitrosourea (MNU) and N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) by electron spin resonance spectroscopy. Both I X and MNNG increased the intensity of the less mobile lipid component (h1) during the incubation while MNU decreased it. The more mobile lipid component (h2), however, was not much influenced by these agents. A positive correlation was observed between the degree of the spectral alterations (h2/h1) caused by I-X compounds and their tumorigenic potency on mouse skin with the exception of the chloride derivative (I-C1). The present results suggest that the reactions of ultimate carcinogens with cell membranes may play an important role in the tumor promoting process.

The relationship between diet and bone mineral content of multiple skeletal sites in elderly Japanese-American men and women living in Hawaii.

The relationship of dietary intake of nutrients assessed by 24-h recalls and supplementary intake of selected minerals and vitamins to the bone mineral content (BMC) of the radius, ulna, and os calcis measured by the photon absorption technique was cross-sectionally studied in a sample of elderly Japanese residents (1208 men and 912 women) in Hawaii. Dietary intakes of milk, calcium, and vitamin D were significantly and positively associated with BMC in both sexes after adjusting for age, weight, height, strenuous exercise (men), history of nonviolent fracture, thiazide use, and estrogen use (women). However, independent contributions of milk or nutrients to BMC were only modest as compared with those of major confounders. No consistent result was obtained concerning the effects of supplementary minerals and vitamins upon BMC.

The relationship of dietary fat and cholesterol to mortality in 10 years: the Honolulu Heart Program.

This report examines the relationship of dietary fat and dietary cholesterol to mortality during a 10-year surveillance of a cohort of men of Japanese descent residing in Hawaii. The consumption of dietary fat (measured in grams) is related inversely and significantly to total mortality. No significant relationships exist between grams of dietary fat and any of the specific causes of death examined. No significant relationships are found between dietary saturated fatty acids (SFA, measured in grams) or dietary cholesterol (measured in milligrams) and any of the specific causes of death examined. In contrast, percentage of calories as fat is related inversely not only to total mortality, but to cancer mortality and to stroke mortality; and it is related directly to coronary heart disease (CHD) mortality. Percentage of calories as SFA is related inversely to cancer mortality and to stroke mortality, and it is related directly to CHD mortality. Only the relationship to stroke mortality remains significant in multivariate analysis if calories from alcohol are excluded from the computation. Dietary cholesterol per 1000 calories is related directly to CHD mortality. While these data provide support for the diet-heart hypothesis, they also suggest that men with low fat intakes have a higher total mortality rate than men with higher fat intakes. This increased risk, due to an excess risk of death from stroke and cancer, indicates that there is no overall beneficial effect from a low fat diet in this cohort.

Qualitative and quantitative differences in hLH species in the first and second LH release induced by continuous stimulation with synthetic LHRH in normal menstrual cycle as assessed by isoelectrofocusing.

It is known that continuous stimulation with LHRH induces a biphasic release of LH. The two pools theory is generally accepted in interpreting this phenomenon. The present study was conducted in order to elicit qualitative differences in LH included in the two pools. Five hundred micrograms synthetic LHRH was infused for 2h in 10 women in various phases of the menstrual cycle and 5 h blood sampling was performed during and after the infusion. Biphasic release of LH was demonstrated in all the cases investigated. The responsiveness to LHRH was most predominant in the midcycle, followed by the luteal phase. Pools of plasma samples at 30-105 min and 150-225 min of the LHRH infusion (plasma pools I and II; corresponding to the initial and the second LH release, respectively) were subjected to isoelectrofocusing (IEF) fractionation, pH range of 3.5-10. LH in plasma was classified in terms of isoelectric point (pI); i.e. A(pI = 9.13), B(8.60), C(8.16), D(7.67), E(7.24), and F(LH species migrating in the acidic pH area). Percentage of high alkaline LH species was significantly elevated in plasma pool I of the midcycle. No phasic difference was observed in the IEF profiles in plasma pool II. Consistent and significant increase and decrease in the percentage of acidic and high alkaline LH species, respectively, were observed throughout the menstrual cycle when the IEF profiles of plasma pool II were compared with those of pool I. The ratio of biological to immunological LH activities (B/I ratio) was markedly depressed in acidic LH species. The highest B/I ratios were obtained in the LH species migrating in the mid-alkaline region. The acidic LH species might represent the young generation of LH molecules prior to acquisition of biological potency. LH species migrating in the mid-alkaline region might be mature and bio-potent forms of the hormone. High alkaline LH species might represent LH molecules over-processed and having lost a degree of biological potency during a prolonged period of storage.

Serum and urinary ferritin levels in patients with rheumatoid arthritis.

The serum and urinary ferritin levels in 52 RA patients were measured by the 2-site immunoradiometric assay method. Serum ferritin levels in RA patients correlated with C-reactive protein (CRP) and erythrocyte sedimentation rate (ESR) but not with serum iron levels and hemoglobin concentrations, although they were within the normal range. High serum ferritin levels were associated with sera with hyper gamma-globulin and rheumatoid factors. In sequential studies, serum ferritin changed in parallel with ESR, CRP and disease activity in a majority of the patients. The urinary ferritin levels and u/s ratios in some RA patients were higher than control values. Higher values were found particularly in the group of patients under gold therapy but not in groups under other treatments.

Tumor promoters potentiate the adrenomedullary secretion induced by acetylcholine and excess K possibly by stimulating phorbol ester receptors.

Catecholamine secretion from the perfused cat adrenal in response to 15 mM KC1 or acetylcholine was significantly increased by treatment with active tumor promoters. A good correlation was found between potentiation of the secretory response and the phorbol ester receptor binding activity, suggesting that promoters modulate the secretory mechanism via the receptors.

Dihydropyridine BAY K 8644 enables reduction of Ca concentration to induce catecholamine secretion from the perfused cat adrenal.

Dihydropyridine derivative BAY K 8644 increased the rate of catecholamine secretion from the perfused cat adrenal. The magnitude of the response was inversely proportional to the extracellular concentration of Ca, i.e. the lower the Ca concentration, the larger the response. The response was markedly inhibited by Mg or nifedipine.

[A case of acute promyelocytic leukemia complicated with severe esophageal stenosis caused by aclacinomycin A].

Aclacinomycin A (ACM) is different from other anthracycline antibiotics in its antitumor activity and clinical effect. We report a case of acute promyelocytic leukemia complicated with severe esophageal ulcer by BH-AC . AMP therapy. The patient was 24 years old. In 1983, he achieved complete remission with BH-AC . DMP therapy but we confirmed relapse in April, 1984. ACM caused nausea, vomiting, diarrhea, alimentary tract bleeding and sore throat, together with a complication of esophageal stenosis. In about half of 10 cases receiving BH-AC . AMP therapy in our hospital, we noticed severe bleeding in the urinary treat, genital organs and alimentary tract. Care should therefore be taken with regard to mucosal injury when ACM therapy is used.

A case of endometriosis of the lung treated with danazol.

A 25-year-old Japanese woman, complaining of catamenial hemoptysis and severe cough complicated with menorrhalgia, was diagnosed as having pulmonary and pelvic endometriosis. She was treated with danazol for 20 weeks. Significant improvement of her condition was achieved during the treatment period. Catamenial hemoptysis recurred at the first menstruation after termination of the treatment. Readministration of danazol was refused. Therefore, surgical removal of the affected lobe of the right lung was performed. Cases of this rare disorder are reviewed.

[Breast metastases from extramammary malignancies in men].

Metastases to the breast from extramammary carcinomas are rare. Carcinoma of the male breast is generally regarded as primary in origin and uncommon, accounting for less than 0.42% of all malignancies in men. Tow men who presented with breast malignancies in the course of their prostatic carcinoma are described. One was metastasis to the breast from prostatic cancer, the other from pancreatic cancer. The prostatic origin of these carcinomas, was confirmed by histological findings and immunocytochemical demonstration of prostatic acid phosphatase with the avidin-biotin-complex method.

Bradykinin-induced rapid breakdown of phosphatidylinositol 4,5-bisphosphate in neuroblastoma X glioma hybrid NG108-15 cells.

External application of bradykinin to neuroblastoma X glioma hybrid NG108-15 cells produced a sustained depolarization preceded by a transient hyperpolarization. Bradykinin also increased the frequency of miniature end-plate potentials recorded from cultured striated muscle cells which had been innervated by NG108-15 cells. Parallelism between facilitative phases of miniature end-plate potentials and depolarization indicates that bradykinin caused an enhanced synaptic transmission from NG108-15 cells due to depolarization. Effects of bradykinin on phospholipid metabolism in the hybrid cells were then examined to shed light upon the mechanism by which bradykinin-receptor interaction leads to facilitation of synaptic transmission. Bradykinin induced specific incorporation of 32Pi into phosphatidic acid and phosphatidylinositol without affecting [3H]glycerol incorporation into these phospholipids by 10 min after its addition. The addition of bradykinin to hybrid cells prelabeled with 32Pi caused a transient decrease (maximal effect seen at 10-30 s) in the radioactivity from phosphatidylinositol 4,5-bisphosphate (PI-4,5-P2) which was followed by the accumulation of radioactivity in phosphatidic acid and phosphatidylinositol. A Ca2+ ionophore, A23187, failed to induce the initial degradation of PI-4,5-P2. The data show that the magnitudes of bradykinin-induced PI-4,5-P2 degradation and membrane potential changes in NG108-15 cells are both dependent on the concentration of bradykinin and that the degradation of PI-4,5-P2 precedes the electrophysiological responses. Taken together with the finding that bradykinin induced a transient increase in Ca2+ influx (at 10-20 s), it appears that a rapid and transient degradation of PI-4,5-P2 might be related to the initiation of the NG108-15 cell activities through mobilization of extracellular Ca2+ into the cells.

[An improved method for plasma methimazole assay and its clinical application].

An improved method for plasma methimazole assay using high performance liquid chromatography is described. The plasma samples were treated with sodium bisulfite and ammonium sulfate prior to extraction with chloroform. This pretreatment of the samples raised the extraction coefficient to 90%, while simple extraction yielded only 55%. The minimal detection limit was 0.02 microgram/ml, and the coefficient of variation at the level of 0.2 and 1.0 microgram/ml was less than 5%. Pharmacokinetics of methimazole was studied after a single oral dose (20 mg/m2) in six subjects including two healthy adults and four thyrotoxic children. Plasma levels of methimazole showed a peak concentration of 1.03 +/- 0.25 microgram/ml approximately one hour after the drug administration. Plasma half-life, area under the curve and distribution volume were 4.56 +/- 0.71 hr, 7.05 +/- 0.95 microgram/ml X hr, and 630 +/- 110 ml/kg respectively.
