Involvement of the telencephalic hemispheres and the preoptic area in sexual behavior of the male goldfish, Carassius auratus: a brain-lesion study.

To locate the brain areas involved in sexual behavior, the effects on male sexual behavior of localized electrolytic lesions in various parts of the telencephalic hemispheres and the preoptic area and of gross ablations of parts of the telencephalic hemispheres were examined in goldfish. All three patterns of male sexual behavior examined (following, butting , and spawning) were severely impaired after bilateral lesions confined to the area ventralis telencephali pars supracommissuralis and/or posterior parts of the area ventralis telencephali pars ventralis (Vs- pVv ), the nucleus preopticus periventricularis (NPP), the lateral forebrain bundle ( LFB ), or the medial forebrain bundle (MFB). On the other hand, sexual behavior was almost unaffected after bilateral ablations of the anterior part or lateral and dorsal parts of the telencephalic hemispheres (AT or LT-DT). Bilateral lesions confined to the area ventralis telencephali pars dorsalis (Vd), the area ventralis telencephali pars postcommissuralis (Vp), the area dorsalis telencephali pars centralis (Dc), the nucleus preopticus (NPO), or the lateral preoptic area (LPA) also had almost no effect on sexual behavior. These results indicate that a particular site in the ventral part of the telencephalic hemispheres (Vs- pVv ) and the anterior part of the preoptic area (NPP) play important roles in the sexual behavior of male goldfish. It was suggested that facilitatory influences from these areas are transmitted through the LFB and/or the MFB.

Induction of murine teratocarcinoma cell differentiation by suppression of poly(ADP-ribose) synthesis.

Poly(ADP-ribose) synthesizing activity in mouse teratocarcinoma EC-A1 cells decreased markedly during differentiation induced by retinoic acid; the activities assayed in permeabilized cells decreased to 25% and 10% of the activity of control (uninduced cells) 2 and 3 days, respectively, after the addition of 0.1 microM retinoic acid to the culture medium. This change preceded changes in morphology and DNA synthesis, which became prominent after 4 days. The decrease in poly(ADP-ribose) synthesizing activity appeared to be caused by a diminution of the synthetase protein and not by a decrease in its catalytic activity, because the full activity disclosed by DNase I treatment decreased in parallel, albeit at about 20 times higher levels. When 8 mM 3-aminobenzamide or 10 mM nicotinamide, specific inhibitors of poly(ADP-ribose) synthetase, was added to the culture medium, the cells underwent differentiation after 7-9 days. An analogue, 3-aminobenzoic acid, which is not inhibitory to the synthetase, induced differentiation much less efficiently than did 3-aminobenzamide, and the effect of 3-aminobenzoic acid appeared to be ascribable to its potent cytotoxicity. Immunohistochemical analysis using anti-poly(ADP-ribose) antibody confirmed the marked reduction in poly(ADP-ribose) synthesizing activity in nuclei of the cells treated with retinoic acid or 3-aminobenzamide but not with 3-aminobenzoic acid. These results suggest that a decrease in poly(ADP-ribose) synthesis triggers differentiation of teratocarcinoma cells.

Immunoglobulin classes of anti-Sendai virus antibody detected by ELISA in infected nude mouse serum.

Sendai virus-infected nude mouse sera obtained on the seventh day after infection or later, in which anti-Sendai virus antibodies were undetectable by hemagglutination-inhibition and neutralization tests, were found to be reactive with the virus antigen by ELISA using horseradish peroxidase-conjugated anti-mouse IgG rabbit IgG. The reactivity was blocked by rabbit anti-Sendai virus antiserum and was not observed against influenza virus which served as a control antigen. Anti-Sendai virus antibody activity of fractions from Sephadex G-200 gel filtration was detected in the IgM fraction when anti-mouse mu chain-specific antiserum was used and in both IgG and IgM fractions when heavy and light chain-specific anti-mouse IgG serum was employed in ELISA. ELISA of the fractions from protein A-Sepharose affinity chromatography of Sendai virus-infected nude mouse sera showed that the eluates at pH 6.0 and pH 3.5 contained IgG1 and IgG2b anti-Sendai virus anti-bodies, respectively, and that the eluate at pH 4.5 contained both IgG2a and IgG3 antibodies.

Small nuclear RNA-protein complex anchors on the actin filaments in bovine lymphocyte nuclear matrix.

When the nuclear matrix from bovine lymphocytes was digested by RNase-depleted trypsin, the bulk of the matrix proteins, except actin, were hydrolyzed. The digestion left rapidly sedimented spherical structures (trypsin-treated nuclear matrix), which mainly were composed of actin (Nakayasu, H. and K. Ueda. Exp. Cell Res. 143, 55-62, 1983). Almost all the small nuclear RNAs of the original nuclear matrix remained associated with these actin spheres after trypsin digestion. By sonication, the small nuclear RNPs (snRNPs) in both untreated and trypsin-treated nuclear matrices were solubilized in association with proteinous filaments of various size. The sedimentation pattern of these snRNP complexes was not changed by the digestion of the bulk of the proteins. The snRNP complex was adsorbed on rabbit muscle myosin-Sepharose then eluted by the addition of 5 mM ATP. We concluded that snRNPs are associated with actin filaments in the nuclear matrix of bovine lymphocytes.

A network data base system for children with cardiac disease--echocardiographic data of MCLS.

We developed a computer-assisted network data base system for children with cardiac diseases. It aims at storage and exchange of medical information from many hospitals to prevent school children from sudden death caused by heart diseases. The computer system used was PANAFACOM U-1500 introduced at Shimane Medical University Hospital in 1980. The format of the questionnaire is designed to standardize the computer-assisted data-entry method among different hospitals. The entries of questionnaires are composed of basic information about the patient and laboratory information. The former is identification data of the patient, and the latter principally the values of M-mode echocardiographic measurement. Two types of output-tables were made from the data base. One is a table of each disease, and the other each hospital.

The development of real-time two-dimensional Doppler echocardiography and its clinical significance in acquired valvular diseases. With special reference to the evaluation of valvular regurgitation.

A noninvasive method for real time blood-flow imaging using ultrasound has long been required in the fields of cardiology and cardiovascular surgery. Recently, we developed a method of two-dimensional Doppler echocardiography (hereafter abbreviated as "2-D Doppler") for clinical use which allows us to obtain noninvasively, intracardiac blood-flow images in real time. The main purpose of this paper is to describe the newly developed blood-flow imaging system "2-D Doppler" and to demonstrate its clinical usefulness in acquired valvular diseases, particularly in the evaluation of valvular regurgitation. The device, in principle, combines a conventional pulsed-Doppler system and a newly developed auto-correlator, in which blood-flow images within a given cross section of a beating heart are noninvasively displayed in real time, simultaneously with conventional two-dimensional echocardiograms. The system can provide three kinds of information, direction, velocity and turbulence of blood flow. The 2-D Doppler examinations were carried out on 72 patients with acquired valvular diseases whose diagnoses were confirmed by angiography and/or surgery. Studies were performed comparing the findings of the 2-D Doppler and those of angiography (for aortic and mitral lesions) or with operative findings (for tricuspid lesions) in the quantitative evaluation of valvular regurgitation. Aortic, mitral and tricuspid valvular regurgitation have been quantitatively demonstrated with 2-D Doppler, and, for each value, the severity of the regurgitation showed a high correlation between the findings of 2-D Doppler and of angiography or surgery. In conclusion, we have found that (1) 2-D Doppler is very useful in detecting and estimating the severity of valvular regurgitation, and that (2) 2-D Doppler may replace conventional angiography in some situations.

A quantitative relationship between conceptus number and ovarian steroid dehydrogenase activity in pregnant rats.

The effects of a conceptus-derived substance on the activity of 3 beta-hydroxysteroid dehydrogenase (3 beta-HSD) and 20 alpha-HSD in the ovary were studied in the rat. On day 7 of pregnancy (day 1 = insemination), rats were laparotomized and the desired number of conceptuses was aspirated from the uterus; thus, rats carrying one, two, three, four, five to seven or eight to ten conceptuses were prepared. They were autopsied on day 15 and 3 beta-HSD and 20 alpha-HSD activity in the corpus luteum (CL) or non-luteal ovarian tissue ( NLO ) was determined. Conceptus number was directly related to 3 beta-HSD and inversely related to 20 alpha-HSD activity in the CL. The serum progesterone level and CL weight were also directly related to conceptus number. Neither 3 beta-HSD nor 20 alpha-HSD activity in the NLO was affected by conceptus number. These results indicated that 3 beta-HSD and 20 alpha-HSD in the CL are probably regulated by placental hormone secreted in proportion to the number of conceptuses; in the NLO these enzymes may be controlled by a different mechanism.

Detection of reaction products of thiamin degradation by catechol derivatives.

To clarify the mechanism of thiamin degradation by catechol derivatives, thiamin and its analogues were incubated with caffeic acid or catechol in 1/15 M phosphate buffer pH 7.4 at 48 degrees C or 50 degrees C. Using thin-layer chromatography and high-performance liquid chromatography, several unknown intermediates of the reaction product, which may have both pyrimidine and catechol moieties, were detected.

Isolation and characterization of a coupled compound of thiamin with catechol.

Equimolecular amounts of thiamin and catechol incubated under suitable conditions produced an unknown reaction product isolated as a light yellow powder from the reaction mixture by reverse-phase ODS column chromatography. Elemental analysis of the purified powder showed the molecular formula to be C18H22N4O4S X HCl X H2O in agreement with a molecular ion peak found at m/e 390 in the mass spectrum. These results together with the data of IR and NMR spectrometry possibly show that this material consists of an isomeric mixture of S-3,4- and S-2,3-dihydroxyphenylthiamin (I and II) which probably arise as a result of oxidative addition of the thiol group of thiamin to catechol.

A five-year immunological follow-up study of the institutionalized handicapped children vaccinated with live varicella vaccine or infected with natural varicella.

Twenty-two institutionalized handicapped children who were susceptible to varicella were vaccinated with live varicella vaccine of the Oka strain and their immune status was followed for 5 years under conditions without exposure to natural varicella. Simultaneously, 7 children infected with natural varicella were followed. Of the 22 vaccinees, 16 showed sero-positive conversion by the fluorescent antibody to membrane antigen (FAMA) test, the other 6 remaining seronegative during 5 years of observation period. All the 16 cases showing seroconversion had detectable antibody for 5 years after vaccination, and 14 of them gave a positive reaction in the varicella skin test. All the 7 cases after natural varicella gave positive reactions in both the FAMA and skin test. These results suggest that immunity conferred by the vaccination would persist long even in the absence of exposure to natural varicella, though further follow-up studies are needed.

[Clinical study on effect of local administration of amikacin for complicated cystitis accompanied by residual urine].

Amikacin 600 mg was administered into the bladder to treat complicated cystitis with benign prostatic hyperplasia or neurogenic bladder after removing residual urine in the bladder in 18 patients. The overall effective rate was 88.9% (excellent in 15 cases, moderate in 1 case, poor in 1 case). The period of effectiveness was from 1 to 8 weeks after administration of Amikacin. No side effects were seen.

Effect of T-cell transfer on Pneumocystis carinii infection in nude mice.

The effect of transfer of spleen lymphocytes from euthymic heterozygote littermates (nu/+) or rabbit antiserum on Pneumocystis carinii (P. carinii) infection in nude (nu/nu) mice was investigated. After intravenous administration of nu/+ spleen T-cells, the number of cysts in the lung of previously infected mice was markedly decreased at day 14 and further at day 30. Intense cellular reactions were seen at day 14 and subsided lesions at day 30. Increased IgG antibody titers were observed at days 14 and 30. Multiple treatments with immune rabbit serum after infection were not effective for reducing the cyst number in the lung, neither enhanced cellular reaction, while the immune serum administered prior to infection prolonged the survival time. The results indicated an important role of T-cell mediated immunity for the resistance to P. carinii infection in mice.
