Chemotactic activity for polymorphonuclear and mononuclear leukocytes in rheumatoid synovial fluids.

In order to why polymorphonuclear leukocytes (PMNs) are predominant and mononuclear leukocytes (MNLs) are few in rheumatoid synovial fluids, chemotactic factor(s) for PMNs and MNLs were studied in the synovial fluids of rheumatoid arthritis (RA-SF) and osteoarthritis (OA-SF) using both Boyden's and agarose methods. The RA-SF showed strong chemotactic activity for human peripheral blood PMNs compared with non-rheumatoid OA-SF. The chemotactic activity for PMNs was well correlated with the number of PMNs in RA-SF, suggesting that it was a natural mediator for PMN emigration into rheumatoid joint cavity. The major chemotactic factor for PMN in RA-SF was of apparent molecular weight of 14,000 and its activity was suppressed to less than 10 percent by anti-C5a antibody, but it failed to show any anaphylatoxin activity which was an attribute of C5a. It was, therefore, suggested to be C5a-like molecule but not C5a itself. The possibility that the factor may be a C5a des-Arg was discussed. On the contrary, the chemotactic activity for MNLs was not found neither in RA-SF nor OA-SF. These findings may explain the fact that PMNs are predominant in rheumatoid synovial fluids.

Prostate growth factor in the extracts of benign prostatic hypertrophy. Partial purification and physicochemical characterization.

The biochemical and physicochemical properties of prostate growth factor (PGF) in the extracts of benign prostatic hypertrophy (BPH) were investigated. The PGF activity stimulating the proliferations of fibroblasts (mouse 3T3 and human BUD-8 cells) was detected predominantly in BPH prostate, and also in normal human prostates and well-differentiated adenocarcinoma prostates. No significant correlation between PGF contents and BPH tissue weight or histological differences (fibromuscular or glandular type) was detected. Gel filtration and isoelectric focusing indicated that partially purified factor(s) by ion exchange column chromatography had a multimolecular form comprising three active components (80,000, 43,000 and 10,000 daltons) and acidic isoelectric points (pH 4.0, 4.3, and 6.0). The activity was susceptible to heat treatment at 80 degrees C for 10 min, and to trypsin, but the factor was devoid of esteropeptidase activity. Subcellular fractionation located the entire activity in cytosol fraction.

Structure of dictyosomes in Scenedesmus acuminatus observed by the freeze-substitution method.

The ultrastructure of dictyosomes in Scenedesmus acuminatus was studied by the freeze-substitution method. The cisternal membranes of dictyosomes clearly exhibited three-layered structures. Electron-dense particles were visible on or in the proximal cisternal membranes. The upper and lower membranes of the distal cisternae were in close contact with each other at the cisternal centre. The electron density of transfer vesicles, secretory vesicles and cisternal margins was very high. These findings differ considerably from those obtained by conventional chemical fixation, and suggest that freeze-substitution is superior to chemical fixation for preserving structures, without distortion and preventing dissolution of constituent substances.

Development of gastric somatostatin-like immunoreactivity in response to corticosterone acetate and dietary changes in young rats.

The effects of dietary changes from breast milk to solid food and corticosterone acetate administration on somatostatin-like immunoreactivity (SLI) in the gastric antrum and corpus were studied in young and adult rats. At 7 days of age, a single dose of corticosterone acetate (250 mg/kg of body weight) was injected in one group of rats, and a single dose of physiological saline was injected in another group. Each group was divided into two subgroups; one was fed only rat breast milk until 25 days of age and the other weaned (solid food alone) at 21 days of age. Gastric antrums and corpora were removed at various ages for assay of SLI concentrations by radioimmunoassay. In non-corticosterone treated rats, antral and corpus SLI increased gradually until 20 days of age, and after changing from breast milk to solid food on day 21 antral SLI increased 2-fold to the adult level, whereas corpus SLI remained constant. In corticosterone-treated rats, antral and corpus SLI on day 11 was 5-fold that on day 7, and thereafter remained constant before changing from breast milk to solid food; after weaning (solid food alone) antral SLI increased again to reach the adult level at 25 days of age whereas corpus SLI remained constant after weaning. Prolonged breast milk feeding alone did not influence antral or corpus SLI levels in either corticosterone-treated or nontreated rats as compared to levels on day 20.(ABSTRACT TRUNCATED AT 250 WORDS)

The efficacy and safety of long-term antihypertensive therapy with nifedipine.

The antihypertensive effect and safety of long-term therapy with nifedipine were evaluated in 132 hypertensive patients, including 100 aged patients who received nifedipine alone or with other antihypertensive agents for a mean duration of 28.6 months. A prompt hypotensive effect was observed 1 month after the initiation of nifedipine therapy. The hypotensive effect persisted for the entire follow-up period and was related to the pretreatment mean blood pressure level. Late drug tolerance was not observed. Dizziness, facial flushing and other unpleasant sensations were observed in 7.5% and ankle edema in 9.8% of the patients. The therapy did not significantly influence serum creatinine levels. Hypotension and side effects were the cause of discontinuation in 10.6% and 4.5% of patients, respectively. We conclude that long-term antihypertensive therapy with nifedipine is both effective and safe, not only in adult patients but also in the high-risk aged patient.

Prevalence of antibodies to Sendai virus and rotavirus in laboratory rabbits.

One hundred and sixty rabbit sera from 10 breeding colonies and 13 laboratory colonies were tested for antibodies to Sendai virus and rotavirus by enzyme-linked immunosorbent assay (ELISA). Antibodies were detected to Sendai virus in 53% and to rotavirus in 81%, indicating the prevalence of these viral infections in laboratory rabbit colonies.

Cytogenetic and clinicopathologic studies of partial moles.

Fifty-six partial moles from 55 patients were karyotyped and analyzed clinicopathologically. Forty-seven were triploid and nine diploid. Triploid partial moles were characterized histologically by focal mild-to-moderate trophoblastic hyperplasia with stromal trophoblastic inclusion. Diploid partial moles lacked marked trophoblastic hyperplasia and stromal trophoblastic inclusions. Four of the nine diploid moles were analyzed for chromosome heteromorphisms, enzyme polymorphisms, and histocompatibility leukocyte antigen specificities to determine their origin, and two were found to be normal. In the follow-up study, none of the partial moles developed into invasive mole or choriocarcinoma. These results indicate that most partial moles are triploid but a minority are normal diploid; regardless of the karyotype, however, partial moles do not have a high propensity for malignancy.

[Adult T cell leukemia accompanied by marked swelling of the left thigh].

A patient with adult T cell leukemia (ATL) whose chief complaint was marked swelling of left thigh is reported. A 72-year-old woman was diagnosed as having ATL based on a high titer of anti-ATLA antibody and abnormal lymphocytes with convoluted nuclei. Computed tomography revealed swelling of her left thigh and a large low-attenuation mass in its muscles. Aspirate from her thigh contained many abnormal cells. Though chemotherapy reduced the swelling for two weeks, she had a relapse and died. This is the first case of ATL having such an onset reported in Japan.
