The concentrations of cefotiam (CTM) in the exudate of pericardium were investigated, and determined in 8 patients accompanied open heart surgery after a single intravenous injection or intravenous drip infusion of 1 or 2 grams. The peak concentrations of CTM have been appeared after 30 minutes or 1 hour, and prolonged for 3 hours, these concentrations of CTM showed about 8--30 times of MIC values which inhibit the growth of E. coli, K. pneumoniae. P. mirabilis and S. aureus. Therefore, it is recommended that CTM will be administered by intravenous infusion just prior to the operation.

Attempting to establish a topical chemotherapy for stomach cancer, injectable 5-FU emulsion (O/W type) has been developed. The distribution of 5-FU was analyzed by the procedures of quantification and X-rays. The combination of lipiodol having a contrast ability in the oil phase was studied. During the endoscopic examination before operation, 1.0 ml of 5-FU was injected at a site, a total of 50 ml at 2 to 4 sites in the submucosal tissue of the lesion. From the result of examination of 5-FU distribution in the extracted stomach and lymph node, it was confirmed that the drug was retained at the site of injection. In addition, 0.017-0.056 microgram/g on the average of 5-FU concentration was transferred to the secondary lymph node. At the same time, its distribution was monitored by X-rays. In those cases which received intermittent administration for twice or more before operation, the 5-FU concentration was detected at a higher rate than in cases receiving a single dose. This suggests that the therapy is markedly effective for stomach cancer and its metastasis to the lymph node as topical therapy.

Many suggestions have been made regarding protective methods against lightning, without any quantitative tests of safety. As a fundamental study on lightning protection, the lethal threshold energy for the artificial lightning impulse was investigated, using 14 rabbits. The voltage and current impulse had a 1 microsecond wave front and 40 microsecond wave tail. The imposed energy was measured by simultaneous photographic recording of both voltage and current waves on separate oscilloscopes. The impulse voltage was applied to head, neck, upper chest, and lower chest while the right hind limb was grounded. Artificial lightning current impulses were fed through the skin of the animals by making the surface flash over so that the surface flash was monitored carefully to delete the data of this case which gives an erroneous energy value from the statistical analysis. The lethal threshold energy was found to be 157.6 +/- 54.1 Joule (J), and the percentage of survival was 25% when 160 J impulse energy was given. Impulses of 160 J were then applied to the heads of 21 other rabbits maintained on an artificial respirator. Under these conditions the percentage of survival increased significantly to 47.6%.
An analysis of the alkaline phosphatase (ALP) isoenzyme in 75 gastric carcinoma tissues was performed. The properties and the originating cells of ALP isoenzymes in them were clarified by using both enzyme histochemical and biochemical methods which had respective superiorities. Also the relationship between intestinal metaplasia and gastric carcinoma was discussed. Liver, placental and small intestinal type ALPs were found in the carcinoma tissues. No ALP activity was found in the carcinoma cells except in the stromal cells in 46 out of 75 cases. In all of these cases, only liver type ALP was found. While, in 29 cases, the activity was also found in carcinoma cells. In these cases, placental and small intestinal type ALPs were found in addition to liver type ALP in 26 and 3 cases, respectively These results indicated that liver type ALP came from the stromal cells and placental and small intestinal type ALPs came from carcinoma cells. The ALP found in carcinoma cells was placental (Nagao isoenzyme) type in most cases. While, the ALP in intestinal metaplastic epithelial cells was small intestinal type in all cases. No sufficient evidence indicating the relationship between intestinal metaplasia and gastric carcinoma was obtained in respect of ALP isoenzyme.
A simple method for the preparation of oligosaccharide-linked aminohexyl-Sepharose 4B (AH-Sepharose 4B) and its application to the purification of anti-glycosphingolipid antibody which is specific for the oligosaccharide moiety are described. The oligosaccharide, which was obtained from galactosyl(beta 1 leads to 3) N-acetylgalactosaminyl(beta 1 leads to 4)galactosyl(beta 1 leads to 4)glucosylceramide (asialo-GM1) by ozonolysis and subsequent alkali treatment, was covalently linked to the AH-Sepharose 4B by reductamination in the presence of NaBCNH3. Anti-asialo-GM1 antibody was purified by means of an affinity technique with the oligosaccharide-linked AH-Sepharose 4B. The antibody bound to the affinity adsorbent was eluted with 0.5 M NaSCN and 3.0 M NaSCN. Antibody with higher specific activity was recovered in the 3.0 M NaSCN fraction with 50% recovery of the activity of the starting material. The purified antibody was found to be quite specific for asialo-GM1. The presence of asialo-GM1 on the cell surface of free-type rat ascites hepatomas was confirmed by the immunofluorescence technique. The cell aggregates induced by the purified antibody were observed under a scanning electron microscope. The cell connection was found to occur at the tips of microvilli of the surface membrane. The localization of asialo-GM1 on the tips of the surface membrane was confirmed by means of the ferritin-conjugated antibody technique.
The presence of asialogangliosides on membrane surfaces of rat bone marrow cells and macrophages was studied by an immunological method using anti-glycolipid antibodies with high specificity. The antibodies against N-acetylgalactosaminyl(beta 1-4)galactosyl(beta 1-4)glucosylceramide (GA2), galactosyl(beta 1-3)N-acetylgalactosaminyl(beta 1-4)galactosyl(beta 1-4)glucosylceramide (GA1) and fucosyl(alpha 1-2)galactosyl(beta 1-3)N-acetylgalactosaminyl(beta 1-4)galactosyl(beta 1-4)glucosylceramide (Fuco-GA1) were prepared, and used for cytotoxicity testing and indirect staining by the immunofluorescence technique. In the cytotoxicity test, approximately 40-50% of rat intraperitoneal macrophages were killed by each of the anti-glycolipid antisera. This finding was confirmed by the indirect immunofluorescence staining method with the purified anti-GA2, anti-GA1, and also anti-Fuco-GA1 IgG. When a rat was preimmunized with bovine serum albumin, all macrophages induced in ascites fluid were found to possess GA1 by the staining method. Immature granulocytes were stained specifically with the anti-GA2 antibody. Granulocytes, myelocytes, and megakaryocytes were stained with the anti-GA1 antibody. Blastic myelocytes were stained with the anti-Fuco-GA1 IgG, but lymphocytes were not stained with any of these anti-asialoganglioside antibodies. From these observations, free-type cells such as macrophages and bone marrow cells were found to possess asialogangliosides as membrane components.
In order to measure Ca2+ efflux from the smooth muscle into a Ca2+-free physiological solution, the method using a Radiometer Ca2+-sensitive electrode was applied and compared with the method using an atomic absorption spectrophotometer to examine its usefulness. The results obtained show that the Ca2+ electrode used here can accurately and continuously monitor free calcium ions effluxed from the smooth muscle into a Ca2+-free solution. The Ca2+-sensitive electrode used here is a useful tool to investigate Ca2+ efflux from the smooth muscle into a Ca2+-free physiological solution.
The effects of various smooth muscle relaxing drugs on contractile responses to acetylcholine (ACh), Ba2+ and Ca2+, and on the tissue cyclic AMP levels were examined in the guinea pig ileum. Papaverine and theophylline caused a decrease both in the maximum height and the slope of dose-response curves induced by the three stimulants, and an increase in the cyclic AMP levels. Diltiazem and D-600 produced a decrease in the maximum and the slope of ACh and Ba2+ dose-response curves, shifted the Ca2+ dose-response curves to higher concentrations, in a parallel manner, but failed to change the cyclic AMP levels. Etomidoline and benactyzine shifted the curves for the three stimulants in parallel to the right, but at higher concentrations depressed the maximum of ACh and Ba2+ responses with a further parallel shift. These drugs exerted little influence on the basal level of tissue cyclic AMP, but etomidoline significantly depressed the Ba2+ -induced increase in cyclic AMP level. The smooth muscle relaxing drugs used could be classified in three types, thereby suggesting that there are at least three different mechanisms involved in smooth muscle relaxing action.
