The behavioral effects of KB-509 and its metabolites were investigated and compared with those of diazepam in mice and rats. The locomotor activity of mice measured by an Animex test was decreased with relatively large doses of KB-509 and diazepam. The anticonflict effect of KB-509 in rats was approximately as potent as that of diazepam. The lever pressing responses in the unpunished period were reduced by diazepam at a dose of 50 mg/kg p.o., while they were not affected by KB-509 even at a large dose such as 100 mg/kg p.o. KB-509 inhibited conditioned avoidance responses of the rat in a shuttle box at an extremely large dose such as 1000 mg/kg p.o. This effect of KB-509 was much less potent than that of diazepam. KB-509 was more potent than diazepam in inhibiting footshock-induced fighting behavior in mice. KB-509 and diazepam prevented maximal electroshock, pentetrazol, and strychnine induced convulsions in mice. KB-509 was more potent than diazepam in potentiating barbital anesthesia, in impairing rotarod performance, and in muscle relaxant activity measured by a traction test in mice. The pharmacological activity of desalkyl-KB-509 was more potent than that of KB-509 and that of desalkyl-3-OH-KB-509 was approximately as potent as that of KB-509. These results indicate that KB-509 possesses pharmacological properties similar to that of diazepam, and it is slightly greater in potency and is much longer in duration of action than diazepam.

Measurement of serum concentration of trypsin by RIA-Gnost Trypsin kit (Hoechst-Japan) was evaluated. The clinical usefulness of measuring serum trypsin level in diabetic patients was assessed. The measurement of trypsin using the radioimmunoassay (RIA) kit revealed good precision and reproducibility with intraassay error ranging from 3.6 to 5.5% in C.V. corresponding to mean trypsin concentration of 236.5-838.7 ng/ml and interassay error ranging from 8.1 to 11.1%. Tests for recovery and dilution were satisfactory for clinical use. Clinical materials included 35 normal subjects, 88 diabetics, 22 patients with liver diseases, 3 with acute pancreatitis, 7 with chronic pancreatitis and 3 with chronic renal failure. Serum trypsin concentration in normal controls was 157.6 +/- 59.9 ng/ml (m + 1 S.D.). Diabetic patients treated with diet therapy alone revealed serum trypsin level of 203.6 +/- 74.8 ng/ml (n = 50). In diabetics treated with sulfonil urea serum trypsin was 171.3 +/- 83.0 ng/ml (n = 25). In patients receiving insulin serum trypsin level was 90.5 +/- 49.0 ng/ml (n = 13). In patients with liver diseases, acute pancreatitis, chronic pancreatitis and chronic renal failure serum trypsin concentration were 236.9 +/- 88.0, 520.1 +/- 80.0, 113.0 +/- 75.6, and 2557 +/- 2771 respectively. Our results may indicate impaired pancreatic exocrine function in patients with severe diabetes mellitus. Increased serum trypsin level in diabetics treated with diet therapy may be due to stimulated excretion of trypsin resulted from restricted food intake. However, further study in larger number of patients is needed.

Rat alpha 2-macroglobulin was isolated and purified from the pooled sera of turpentine-injected rats by sequential use of dextran sulphate, DEAE-cellulose and gel filtration chromatography. The final protein product obtained by this procedure proved to be alpha 2-macroglobulin of a high degree of purity based on electrophoretic, immunologic and centrifugal analysis. The alpha 2-macroglobulin preparation also binds stoichiometrically to trypsin preventing subsequent inhibition by protein trypsin inhibitors. SDS-polyacrylamide gel electrophoresis of rat alpha 2-macroglobulin after incubation with trypsin suggested that there are at least two susceptible peptide bonds in the 170,000-dalton alpha 2-macroglobulin subunit. The concentration of alpha 2-macroglobulin in the sera of rats was measured by electroimmuno assay a monospecific antiserum against alpha 2-macroglobulin. Purified alpha 2-macroglobulin was used as a standard. Sera from normal male rats contained 32 +/- 4 micrograms of alpha 2-macroglobulin per ml. To determine the time course of response of alpha 2-macroglobulin to inflammation, rats were subjected to either laparotomy or subcutaneous injection of turpentine. After either type of injury, the concentration of alpha 2-macroglobulin increased rapidly, reaching a maximum value of 110-140 times that of the control value by 24 h. Little difference was noted in responsiveness between the two sexes.
Seven cases of phalangeal microgeodic syndrome in childhood are reported. Reviewing the literature, we have found a total of thirty Japanese cases, including the cases of our own. On the other hand, only ten cases have been reported outside Japan. The mean age of onset of the Japanese was six and a half years, which is older than that in the initial description. Of further interest is the fact all the cases in Japan occurred sporadically and exclusively in winter, and regressed spontaneously and fairly rapidly within six months. Furthermore, most of the cases reported in Europe also occurred in the winter months. These facts imply an etiological relationship between the lowered temperature and the bone lesions.
In two patients wih aneurysmal changes due to arteriosclerosis in the axillary and the subclavian artery respectively, spiral vein grafts were used in a substitute for vascular reconstruction. After resection of the aneurysm, these vein grafts resulted in an excellent adaptation to the recipient vessel. Complicated techniques were not required and the construction time was minimal. Such an approach may be used even in adjacent sites to a joint.
A method is described for the production of rabbit anti-Gm sera using normal IgG proteins and fragments thereof as the immunogens. Anti-G3m(16) and anti-G3m(21) were readily producible by immunizations with protein A-purified IgG3 proteins of G3m(13, 15, 16) and G3m(21) type, respectively. Anti-G1m(2), anti-G1m(3), and anti-G3m(5) were obtained by immunizations with type Gm(1, 2, 21) Fc fragments, type Gm(1, 3, 5, 13) IgG1-rich IgG, and type G3m(5, 13, 21) IgG3, respectively. These immune sera can distinguish the 9 Gm phenotypes in the Japanese.
Wistar female rats were injected with testosterone (T) or 5 beta-dihydrotestosterone (5 beta-DHT) for the first 5 days of life. Neonatal treatment with 1 mg of T resulted in anovulatory persistent oestrous syndrome in 100% of the animals. In the females injected with 1 mg of 5 beta-DHT, 74% of the treated rats became sterile at 120 days of age. In addition, 0.5 mg 5 beta-DHT was also effective in inducing anovulatory persistent oestrus; the incidence of sterility was 10 and 80% at 60 and 120 days of age, respectively. When daily dose of 5 beta-DHT was reduced to 0.1 mg, however, only 33% of the rats resulted in sterility. These results suggest that the free form of T and non-aromatizable androgen, 5 beta-DHT, can permanently suppress the development of female type of neuroendocrine regulation. The possible participation of the process other than the central aromatization in androgenization will be discussed.
A rapid and simple cytologic screening system for urinary tract cancer stained with toluidine blue was described. Utilizing this system, a hospital population was screened for urinary tract cancer from specimens sent for routine urinalysis. In this study of 16,062 fresh urine sediments selected at random from non-urologic patients, three cases histologically proven bladder cancer were detected, leading to the detection rate of one bladder cancer in 5,000 individuals. The results emphasizes the usefulness of this simple screening technique in combination with clinical urinalysis and cytology for early detection of urinary tract malignancy.
