Antiteratogenic effects of tumor inhibitors, caffeine, antipain, and retinoic acid in mice.

To learn the effects of tumor inhibitors on chemically induced malformations, caffeine, antipain, and 13-trans-retinoic acid were given to pregnant ICR/Jcl mice after a single dose of urethan, N-hydroxyurethan, N-methyl-N-nitrosourea, N-ethyl-N-nitrosourea, or 4-nitroquinoline 1-oxide, which induces about 50% of the malformed fetuses. When caffeine was given immediately after carcinogen treatment on Day 10, urethan- and N-ethyl-N-nitrosourea-induced malformations were significantly suppressed by caffeine posttreatment, while N-hydroxyurethan- and N-methyl-N-nitrosourea-induced malformations were not suppressed by caffeine. 4-Nitroquinoline 1-oxide-initiated teratogenesis was also suppressed, but not significantly so (p not equal to 0.07). The results were very similar to those of the effects of caffeine on tumors induced by these carcinogens. Malformations of genetic origin (cleft palates and cleft lips) in CL/Fr mice were also suppressed significantly by caffeine treatment on Days 8 to 11, although the level of inhibition was less than that in chemically induced malformations. A protease inhibitor (antipromotor), antipain, also suppressed urethan-induced malformations. The antiteratogenic effects of antipain were most effective when it was given during the period of 24 to 48 hr after urethan treatment, while those of caffeine were most effective when it was given immediately after urethan. The promoting process might be involved in chemically induced teratogenesis, as it was in carcinogenesis. A natural retinoid (13-trans-retinoic acid) also suppressed urethan-induced malformations. Thus, tumors and malformations induced by chemical carcinogens were suppressed by tumor inhibitors, suggesting the similarity of both processes in the subcellular level, in spite of their morphological differences.

[Radiotherapy for post-operative inflammatory breast cancer].

Two patients with inflammatory breast cancer treated with a combination of radical mastectomy, irradiation, and immunochemotherapy are reported. After radical mastectomy, both patients were given a dose of 4000 rad and 5000 rad to the chest wall and parasternal lymph nodes, and 5000 rad to the axillary and supraclavicular lymph nodes. However, both patients died of recurrence within the irradiated field of the chest wall and metastatic spread to the neighbouring skin. A discussion on the dose and field in radiation therapy for inflammatory breast cancer is presented.

The mammillary body is a potential site of antianxiety action of benzodiazepines.

The present study was designed to examine the anticonflict action of benzodiazepines injected into the mammillary body (MB) using the rat conflict-punishment procedure. Diazepam 20 micrograms/microliters, chlordiazepoxide 60 micrograms/microliters and midazolam 30 micrograms/microliters, bilaterally injected into the MB, produced a significant increase in the punished responses without changes in the unpunished responses. These findings have demonstrated for the first time that the MB could be a potential site of antianxiety action of benzodiazepines.

Isolation of ureaplasmas from poultry and experimental infection in chickens.

Ureaplasmas were isolated from the oropharynxes of 47 of 247 (19 per cent) Leghorn chickens (Gallus gallus domesticus) and from five Japanese bantams but none was isolated from the oropharynx or cloaca of other poultry comprising 10 Japanese game, 75 common quails (Coturnix coturnix japonica), 17 turkeys (Meleagris gallopavo) and 10 guinea fowls (Numida galeata). In apparently healthy chickens, ureaplasmas were found at various sites, including the conjunctiva, nasal cavity, oropharynx, upper and lower tracheas, but not from the air sac, lungs, yolk, oviduct, urine or cloaca. All the isolates were antigenically similar but had no serological relation to those isolated from man, monkey, cattle, goat, sheep, dog and cat. In chickens experimentally infected with an avian ureaplasma, the organisms infected the oropharynx and nasal cavity but none of the birds inoculated demonstrated any clinical signs or macroscopic lesions.

Polymorphism of the haptoglobin peptides by isoelectric focusing electrophoresis and isoelectric point determinations.

In this investigation, the authors developed two new procedures: a micromethod for haptoglobin purification and the isoelectric focusing electrophoresis on slab polyacrylamide gel for peptide subtyping. These technics are adapted to the study of large sample series for population genetic surveys. The improvements obtained enabled us to disclose in an easy and highly reproducible way the Hp alpha and alpha 2 peptide chains. Electrophoretic separation of the alpha 2 FS, SS, and FF chains were greatly improved. Their frequencies estimated in a sample already investigated by the conventional PAGE presented higher values than previously described. New Hp alpha and alpha 2 mutants were also detected. For the first time, isoelectric points of the Hp peptides were determined; the values obtained are discussed with regard to their known amino acid structure.

Mass screening for urinary tract cancer with urine cytology.

A rapid and simple cytologic screening technique for fresh urine specimens stained with toluidine blue is described. As to the validity and description of cell structure, the accuracy of urine cytology with this procedure is very similar to that of the Papanicolaou technique. Utilizing this technique, a hospital population was screened for urinary tract cancer from specimens sent for routine urinalysis. In this study of 16,062 fresh urine sediments selected at random from nonurologic patients, a total of three cases of histologically proven bladder cancer were detected, corresponding to a rate of one bladder cancer in 5,000 individuals. The results emphasize the usefulness of this simple screening technique in combination with clinical urinalysis and cytology for early detection of urinary tract malignancy.

Evaluation of limited resection of the lung for treatment of lung cancer.

The validity and indications of limited resection for lung cancer were studied based on the results of 34 patients with lung cancer who underwent surgery. This method appeared to be an effective technique for the preservation of the cardiopulmonary function after surgery, particularly in elderly patients. The prognosis of those who underwent limited resection for selected cases was not significantly different from that of those treated by lobectomy. Recurrences were seen exclusively in Stage II patients, most of whom had a poorly differentiated type of cell pathology and developed blood vessel involvement. It was concluded from this study that this technique can be feasibly applied to patients with a tumor of less than 3 cm located at the periphery of the lung and with the histologic picture of highly differentiated type.

Pacemaker implantation in a neonate with congenital complete heart block--a case report.

Reported herein is our experience with pacemaker implantation in a neonate with complete AV block and without other concomitant cardiac anomalies. A male fetus exhibited an intrauterine heart block rate of about 40 beats per minute on the electrocardiogram. An emergency pacemaker implantation was attempted at birth by means of intravenous temporary pacing to relieve signs of congestive heart failure. On the 28th day of life, a myocardial electrode was fixed on the right ventricular wall and the generator was positioned on the left side of the subcutaneous abdominal wall. A technical device was designed by means of packing pacemaker wire in a loop-forming fashion with a silastic sheet and was made in such a way as to adjust with growth of the patient. During the 2 year follow-up period, this technique proved to be effective. Our experience also supports the contention that pacemaker implantation in the newborn is feasible and beneficial in the treatment of congenital complete AV block.

The limit of application of intraaortic balloon pumping (IABP) on canine myocardial infarction.

The limit of application of intraaortic balloon pumping was evaluated in cases of canine myocardial infarction produced by either ligation of the descending coronary artery and of branches in the right coronary artery or establishment of stenosis in the left main coronary artery. The limit of application of intraaortic balloon pumping was confirmed to be as follows: 1) stenosis of seventy-five per cent or more in left main coronary artery, 2) over 45 per cent infarction of the entire left ventricular free wall, 3) endocardial viability ratio of less than 0.65, 4) blood flow of less than 18 ml/min/100 g in the left circumflex coronary artery when the left descending coronary artery was ligated, 5) and less than 0.69 in the ratio of inside blood flow to outside of the left ventricular free wall, as related to regional myocardial blood flow.

The differences in growth and activity of the tryptophan-NAD pathway between Wistar and Sprague Dawley strains of rats fed on tryptophan-limited diet.

Three-week-old weanling rats of Wistar and Sprague Dawley strains were fed on tryptophan-limited and nicotinic acid-free diets for 46 days and the following was observed. (1) Gain in body weight of the Sprague Dawley strain was significantly higher than that of the Wistar strain from the 10th day of feeding. (2) Tryptophan oxygenase [EC 1.13.11.11]activity in the Wistar strain was constant but was significantly increased in the Sprague Dawley strain during the latter period of this experiment. (3) The total amount of nicotinic acid, quinolinic acid, N1-methylnicotinamide and tryptophan in liver and kidney of the Sprague Dawley strain was significantly higher than that of the Wistar strain. (4) Total urinary nicotinic acid, quinolinic acid and N1-methylnicotinamide levels were not very different between the two strains, but it was observed that at the 38th day the levels in the Sprague Dawley strain were significantly higher than those in the Wistar strain. From the above result, it was presumed that the Sprague Dawley strain of rat was more resistant to deficiency of nicotinic acid than the Wistar strain fed on a low tryptophan and nicotinic acid-free diet. Aminocarboxymuconate-semialdehyde decarboxylase [EC 4.1.1.45] activities in livers of both strains dropped to half the original value at the end of the experiment. This change may indicate metabolic control of increase in flow from tryptophan to NAD.
