Adult female rats were hypophysectomized and their pituitary glands autotransplanted beneath the left kidney capsule on day 2 (day 1 was the day of ovulation). In such rats the pituitary secretes prolactin fairly constantly and the corpora lutea secrete progesterone for several months. To induce the luteolytic effect of prolactin the rats were first injected s.c. with 2-bromo-alpha-ergocryptine (CB-154) on cycle days 12, 13 and 14 (i.e. 10, 11 and 12 days after operation) to depress prolactin secretion, and then with CB-154 vehicle (70% ethanol) daily until cycle day 21, to allow prolactin secretion to resume. One ovary was removed from each rat on day 15 and the remaining one on day 22. The mean (+/- S.E.M.) weight of the corpora lutea on day 15 was 1.46 +/- 0.06 mg and 0.98 +/- 0.07 mg on day 22 (n = 17). In contrast, rats in which the CB-154 treatment was maintained to day 21 had corpora lutea which weighed 1.31 +/- 0.09 on day 15 and 1.47 +/- 0.08 mg on day 22 (n = 15). To investigate whether indomethacin, a prostaglandin synthesis inhibitor, affected the luteolytic action of prolactin, the experiment was repeated, but on day 15 (after the removal of one ovary) the groups in which CB-154 treatment was stopped, as well as the group in which CB-154 treatment was maintained, were each divided into two groups. In one, indomethacin-containing silicone elastomer wafers and, in the other, blank silicone elastomer wafers, were placed within the bursa of the remaining ovary.(ABSTRACT TRUNCATED AT 250 WORDS)
Radioactivity of the histamine-induced paw edema was measured after intravenous injection of 67Ga-citrate in anesthetized rats. The radioactivity was fluctuated almost parallel with the edema rate following subcutaneous injection of histamine. Both the radioactivity and the edema rate were almost equally reduced by pretreatment with chlorpheniramine, an antihistaminic agent. There was a good correlation between the edema rate and the radioactivity. These results suggest that 67Ga-citrate is useful for the pharmacological study of antiinflammatory drugs, or is available for measuring the vascular permeability.
Mouse monoclonal antibody c 143 was purified and F(ab')2 fragments were generated by pepsin digestion and then radiolabeled with 125I. The 125I-labeled c 143 F(ab')2 fragments were injected into athymic mice bearing bovine lymphoid tumor cells. The fragments became preferentially localized in tumor tissues, but not in normal tissues, as determined by differential counting of tissue radioactivity. The fragments became localized specifically in those tumors that were reactive with c 143 in vitro, but did not become localized in unrelated tumors. Localization of labeled F(ab')2 fragments of a monoclonal antibody of the same isotype directed against Taka virus (a variant of Newcastle disease virus) was not observed in athymic mice bearing bovine lymphoid tumor cells. Tumors were detectable by radioimmunoscintigraphy, using radiolabeled c 143 F(ab')2 fragments, without background subtraction, and by use of silver-grain scattering in light microscopic autoradiography.
A 29-year-old male visited our hospital because of lower abdominal pain. Acute appendicitis was suspected, and surgery was performed. However, no abnormality was found in the vermiform appendix. Instead, atrophy of the greater omentum, numerous nodes of varying sizes, and a small amount of ascites were observed. On the basis of histological examination, a diagnosis of metastatic glandular cancer was made. Gastric fluoroscopy performed 2 months earlier in another hospital had revealed irregularity of the stomach wall and a large concave area on the side of the greater curvature. Photogastroscopy and CT demonstrated progressive Borrmann type III cancer at the corresponding site and metastasis to regional lymph nodes. Biopsy findings were similar to the histological findings. After 1 week, treatment with UFT (400 mg/day), MMC (10 mg/month) and LNT (1 mg/week) was initiated. After about 3 months, i.e., following administration of 40 g UFT, 30 mg MMC and 8 mg LNT, gastric fluoroscopy and photogastroscopy revealed complete disappearance of the tumor. No abnormality was found by laboratory studies. The patient has since returned to a normal life.
The dose limiting factors of cisplatinum are nephrotoxicity and emesis. Nephrotoxicity has been reduced by hydration but nausea and vomiting caused by cisplatinum have led to refusal of potentially curative therapy by a number of patients. The prevention of nausea and vomiting by a combination of antiemetic drugs administered to ovarian patients receiving chemotherapy inducing (cisplatinum 50mg/m2, adriamycin 300 mg/m2, cyclophosphamide 300 mg/m2 and 5FU 350 mg/m2) was studied. the combination antiemetic drugs were metoclopramide (1mg/kg), dexamethasone (10mg/m2), droperidol (1mg/m2) and diphenhydramine (20mg/body). These drugs without diphenhydramine were administered intravenously 30 minutes before and 2.5 hours, 5 hours and 7.5 hours after chemotherapy. Diphenhydramine was administered intramuscularly 30 minutes before and 5 hours after chemotherapy. No vomiting was noted in 82.6% (19/23) of cases, and no patient vomited more than four times. This combination regimen provided very good protection against cisplatinum induced emesis.
Tamoxifen is a nonsteroidal antiestrogen whose mode of action is, as yet, unclear. The purpose of this report is to investigate the effects of Tamoxifen on endometrial adenocarcinoma ultrastructurally. Five patients with endometrial adenocarcinoma, 3 cases well differentiated and the others undifferentiated, were treated with 20mg Tamoxifen per os a day for 7 days, and the following changes were observed: The cells became roundish and slightly swollen. Development of cytoplasmic organelles was observed. In the cytoplasm, glycogen granules did not increase. These results suggest that an anti-cancer effect is brought about by the induction of the tissue toward functional differentiation as well as inhibition of glycogen metabolism in tumor cells.

The effects of Ca2+ and calmodulin on contraction of saponin-treated (chemically skinned) uterine smooth muscle fibers of pregnant rats were examined. Ca2+ sensitivity, defined as the pCa required for half maximum activation of force production, was found to change with the progress of pregnancy; low in the early and middle stages and high in the later stages of pregnancy. The overall change of Ca2+ sensitivity was about pCa 1.5 during the period of pregnancy. The effect of calmodulin on contraction was also found to be dependent on the stages of pregnancy. Calmodulin was effective on the augmentation of the tension rather than the change in Ca2+ sensitivity, and this augmentation was large in the early and middle stages of pregnancy. The amount of calmodulin, which eluted out of uterine muscle cells during saponin treatment, was large in the early and middle stages of pregnancy. The results indicate that the contractile response of the uterine muscle cells during the period of pregnancy seems to be controlled by both the changes in Ca2+ sensitivity and in the amount of free calmodulin in uterine muscle cells.
We considered the possible relationship between the serum levels of progesterone and estradiol-17 beta, which were controlled to that of several stages in normal pregnant rats, and the Ca2+ sensitivity, defined as the pCa required for half maximum activation of force production, of chemically skinned uterine muscle fibers from ovariectomized rats. The Ca2+ sensitivity was negligibly influenced by the levels of these hormones.
Monoclonal antibody against tumor-associated antigen (TAA) expressed on bovine leukemia cells was conjugated to liposomes containing adriamycin (ADM), and the specificity and therapeutic effects of the conjugates were examined in vitro and in vivo using a TAA-positive bovine leukemia cell line as the target tumor. In vitro studies with the TAA-positive cell line clearly indicated that the antibody-conjugated liposomes containing ADM exerted selective effects on TAA-positive cells in the inhibition assay of 3H-thymidine incorporation. Three injections of liposomes containing ADM (4 mg/kg) into tumor-bearing nude mice significantly inhibited the tumor growth and the therapeutic effect of the antibody-conjugated liposomes was far greater than that of normal mouse IgG-conjugated liposomes as assessed in terms of tumor size.

While the treatment of ovarian cancer has made remarkable progress in recent years, the chemotherapy of ovarian germ cell tumor has not as yet been fully established and remains to be improved. We investigated the effectiveness of a single-agent therapy with Vincristine (VCR), Actinomycin D (ACD), and Cyclophosphamide (CPM) against a human undifferentiated dysgerminoma's cell line (JOHYL-1) with the following results. The ratio of the number of viable cells to the total cell population treated for JOHYL-1 cells of ascitic type was 70% with VCR, 58% with CPM and 25% with ACD; hence the efficacy on this malignant cell line was the greatest with ACD followed by VCR and CPM in that descending order. Notably with CPM, a striking antitumor effect was observed from 48 to 24 hours after treatment. The L.I. attained a peak at 24, 18 and 30 hours, respectively, after treatment with VCR, ACD and CPM. The M.I. was as high as a little more than 6% at 36 hours after treatment with VCR. There also was noted a shift in the cell cycle with any of the test drugs. Morphological changes observed in the cell were most obvious with ACD followed in order by VCR and CPM. LDH & isozyme pattern: In these respects there were no striking differences among the three drugs, even though the total enzyme activity was somewhat higher with VCR.(ABSTRACT TRUNCATED AT 250 WORDS)
Cyclic rats were hypophysectomized and their pituitaries transplanted beneath the kidney capsule on day 2 (day 1, ovulation); they were then either hysterectomized or sham hysterectomized (uterus intact rats) and injected sc daily with either 1.0 micrograms 17 beta-estradiol (E1) or with the sesame oil vehicle until autopsy on day 105. Blood samples were drawn by jugular venipuncture every 2 to 3 days until day 21 and once weekly thereafter. In these rats the serum progesterone level rose until about day 8 and tended to reach a plateau between days 8 and 14. The regression phase began, in general, after days 8 to 14 and continued, without change in rate, for at least 3 months. In both the uterus-intact and the hysterectomized rats, E1 seemed to induce an earlier onset of regression and a brief increase in its rate, but did not otherwise affect the rate of regression. In the hysterectomized rats regression began later than in the uterus-intact rats, but its rate was also not different from the latter. The serum progesterone pattern of rats subjected to pituitary autotransplantation on day 2 was compared with that of pseudopregnant rats which were either subjected to pituitary autotransplantation on day 2, 5, 7, or 9, or were given a pituitary homotransplant on day 2, and then hypophysectomized on day 2, 3, 5, or 7. A group of rats with sham operations served as additional controls. In these rats, the same pattern of serum progesterone already described was seen in those subjected to either pituitary autotransplantation on or before day 5, or to pituitary homotransplantation, and to hypophysectomy on or before day 5. However, when the pituitary was autotransplanted on day 7 or day 9, or when the pituitary homotransplant-bearing rat was hypophysectomized on day 7, regression began earlier and was more rapid than in rats operated on on or before day 5. In fact, in the rats operated on on day 9, regression was as rapid as it was in the intact (sham operation) controls. Another group of rats subjected to pituitary autotransplantation and hysterectomy on day 2 was divided into four subgroups on day 21. In two of these, the rats received indomethacin-containing Silastic capsules inserted into each ovarian bursa; in the other two groups blank capsules were similarly inserted.(ABSTRACT TRUNCATED AT 400 WORDS)
Adult rats were hypophysectomized and their pituitary glands autotransplanted on day 2 of an estrous cycle (day 1, ovulation); the rats were either hysterectomized or sham hysterectomized immediately before this operation. On day 21, a few days after the beginning of the 3+ month period of regression of progesterone secretion, they were injected sc with 500 micrograms prostaglandin F2 alpha (PGF2 alpha) either on day 21 alone, on days 21 and 22, on days 21, 22, and 23, or on days 21 and 23; controls were injected with saline on days 21, 22, and 23. In the latter the serum progesterone level slowly fell, as it does in untreated rats subjected to pituitary autotransplantation on day 2. PGF2 alpha treatment induced a rapid, drastic fall in the progesterone level, but in each group except the one injected on days 21 and 23, the level returned to, or close to, the control level in about 10 days. Even in the group injected on days 21 and 23, the pattern of serum progesterone resembled the others although the values were much lower. In a similar group of rats subjected to pituitary autotransplantation on day 2 (all of which were also hysterectomized on day 2), a single injection of 500 micrograms PGF2 alpha (which again induced the same changes in serum progesterone as in the first experiment) was combined with either a 6-h or 4-h withdrawal of PRL; this was achieved by injecting 2 bromo-alpha-ergocryptine (CB-154) at zero time to lower PRL secretion, and beef PRL, in a delaying vehicle, 6 or 4 h later, followed by a second dose of PRL 18 h after the first. When combined with such a brief period of PRL withdrawal, the PGF2 alpha treatment induced a very rapid, drastic and permanent fall in serum progesterone, equal to that induced by CB-154 alone, or by removing the pituitary transplant on day 21. The brief periods of PRL withdrawal, by themselves had very little (6 h) or no (4 h) effect on the progesterone level, and treatment with PRL alone was also without effect. Treatment with CB-154, PGF2 alpha, and PRL simultaneously (i.e. without a period of PRL withdrawal) had the same effect as treatment with PGF2 alpha alone.(ABSTRACT TRUNCATED AT 400 WORDS)
Penetration of aztreonam (AZT) into the uterus and the adnexal tissues and usefulness and safety of AZT for obstetric and gynecologic infections were studied with the following results. By one shot intravenous injection of AZT 1 g, the uterus and the adnexal tissues showed favorable penetration with Cmax 27.0-48.5 micrograms/g, AUC 29.4-84.9 micrograms X hr/g and Tmax 0.10-0.44 hours. MIC50, MIC80 and MIC90 of AZT for Gram-negative bacteria measured prior to administration were very low being 0.10 micrograms/ml, 0.20 micrograms/ml and 1.56 micrograms/ml, respectively. Clinical effect of AZT for 30 infection cases in obstetrics and gynecology was evaluated according to an overall efficacy criteria resulting in "good" for all the cases. With regard to microbiological effect, 90.9% of the pathogens isolated prior to the administration were eliminated by AZT. During and after the administration of AZT, side effect due seemingly to AZT was not observed in subjective and objective symptoms and laboratory values.
