Evidence for the occurrence of oligosaccharides from bovine glycophorin having a branching point at C-6 of N-acetylgalactosamine.

Some large acidic oligosaccharides from bovine glycophorin were purified and their structures were determined by methylation analysis, nitrous acid deamination and selective acid hydrolysis after hydrazinolysis, Smith degradation and acetolysis. One oligosaccharide was a mixture of two decasaccharides: (formula; see text) The mixture of oligosaccharides released by NaOH-NaBH4 treatment was subjected to selective acid hydrolysis after hydrazinolysis. The occurrence of N-acetylglucosamine residue attached to C-6 of the reducing terminal N-acetylgalactosaminitol was demonstrated by isolating the disaccharide, GlcNAc(1----6)GalNAcol.

Simulation of infectious disease by Reed-Frost model with proportion of immune and inapparent infection.

A modified Reed-Frost model into which the proportions of immune, and of inapparent infections were introduced, has been examined in order to find how it might affect the predicted epidemic curve. Under the conditions of a constant community size and a constant epidemic size, the modified Reed-Frost model yielded a predicted epidemic curve which was distributed in a relatively narrower range, possessed a higher peak, and which came much earlier than that produced by the Reed-Frost model without the above modifications.

X-ray microanalysis of spontaneously formed urinary calculi in a jaundiced strain of rat.

A high incidence of spontaneously formed urinary stone was found in the females of a jaundiced strain of rat developed from a cross between Gunn's rat and Wistar-Imamichi rat. In this colony, 42.3% of the females had urinary calculi. Elemental analyses of these urinary calculi were carried out with an analytical electron microscope, a high-resolution transmission electron microscope fitted with an energy dispersive type X-ray microanalyzer and a scanning device. In the surface and middle areas of the stone, the main components were recognized as magnesium (Mg) and phosphorus (P). In the central region of the stone, calcium (Ca) and phosphorus (P) were found as the main elements with trace amounts of sodium (Na), chlorine (Cl), and potassium (K). The analyses indicated that the spontaneous urinary stone consisted of phosphate salts with calcium or magnesium. In addition, the mass ratio of the Mg/P or Ca/P in the stones was calculated from X-ray pulse intensity ratios and compared with the mass ratio of a standard sample. The results suggested that the magnesium and phosphorus in the urinary stones existed as ammonium magnesium phosphate, MgNH4PO4, and the calcium and phosphorus as tribasic calcium phosphate, Ca3(PO4)2.

Treatment of NZB/W F1 mice with NZW splenic T cells or with serum of mice experiencing the graft-versus-host reaction: suppression of ongoing anti-double-stranded (ds) DNA antibody formation and improvement of renal function.

Seven-month-old NZB/W F1 mice were inoculated with 5 X 10(7) NZW spleen cells. The mice so treated showed a clear fall of anti-double-stranded (ds) DNA antibody titers accompanied by a decrease in blood urea nitrogen (BUN) concentration. Mice so treated showed markedly increased survival times (more than 17 months) compared with untreated controls (mean +/- SD 8.38 +/- 0.75 months). A cell fractionation study suggested that T cells among NZW spleen cells play a major role. Recipients of the unfractionated or the T-cell fraction of NZW spleen cells had histologically less glomerulonephritis than untreated control NZB/W F1 mice. NZW B-cell transfer to NZB/W F1 mice had no effect. Serum obtained from NZB/W F1 mice receiving NZW spleen cells 10 days previously (graft-vs-host (GVHR) serum) was injected into 8-month-old NZB/W F1 mice. A clear fall in anti-ds DNA antibody titers and inhibition of age-associated sharp increases in BUN were observed. The mean life span of these mice (10.50 +/- 0.58 months) was significantly longer than that of untreated controls (8.69 +/- 0.38 months). GVHR serum had the ability to suppress anti-ds DNA antibody formation by NZB/W F1 spleen cells in vitro. Allogeneic GVHR serum prepared in C57B1/6 X DBA/2 (BDF1) mice by transferring C57B1/6 spleen cells was also effective in suppressing anti-ds DNA antibody formation in vitro.

Spinal evoked potentials in normal Japanese infants and children.

The spinal evoked potentials on right peroneal nerve stimulation were recorded with surface electrodes in 58 normal children of 41 weeks to 15 years old. Bipolar recordings were performed. In all subjects, tri- or quadri -phasic potentials with poorly defined initial positive phases were recorded. The latency of the evoked potentials increased progressively from the lumbar to cervical location. The peripheral conduction velocity from the stimulating cathode in the popliteal fossa to the lumbar recording location increased until 4 or 5 years, and then reached a plateau. Spinal conduction velocity between the lumbar and cervical locations increased until 7 years, and then remained constant.

A case of lissencephaly (agyria) syndrome with apnea--polygraphic study.

A 4-month-old infant with lissencephaly syndrome who was diagnosed on the basis of clinical symptoms and computed tomography experienced four apnea attacks during three polysomnographies. Apnea attacks were of two types. Type I apnea attacks were thought to be sleep apnea. The cause is unknown. Apnea was followed by tonic seizures due to hypoxia. Type II apnea attacks were thought to be epileptic apneic attacks, because they proceeded from the epileptic discharges in the right anterior temporal region (lead of electrooculogram). The origin of Type II apnea attacks was focused in the limbic system.

Free amino acid concentrations in plasma, erythrocytes, granulocytes, and lymphocytes in umbilical cord blood, children, and adults.

Concentrations of free amino acids were measured concurrently in plasma, erythrocytes, granulocytes, and lymphocytes in umbilical cord blood, neonates, children, and adults. In each age group, the patterns of free amino acids were fairly similar in plasma and erythrocytes except for aspartic acid which was more abundant in erythrocytes. Of the amino acids in granulocytes, 71-77% was taurine; in lymphocytes taurine, aspartic acid, and glutamic acid comprised 35-44%, 18-24%, and 20-28%, respectively, of the total in all age groups. Leukocytes may contribute to the interorgan transport of amino acids to about 10% of the erythrocytes' contribution. Postnatally, the levels of glutamic acid and tyrosine in plasma; threonine plus glutamine, serine plus asparagine, and tyrosine in erythrocytes; histidine in granulocytes; and glutamic acid in lymphocytes were significantly increased (p less than 0.001); while the levels of phenylalanine and lysine in plasma; taurine in erythrocytes; valine and phenylalanine in granulocytes; and threonine plus glutamine, tyrosine, and phenylalanine in lymphocytes were significantly decreased (p less than 0.001). After the neonatal period concentrations of taurine and aspartic acid in erythrocytes, taurine and valine in granulocytes, and tyrosine and phenylalanine in lymphocytes increased gradually with age; while concentrations aspartic acid in plasma, histidine in granulocytes, and glycine in lymphocytes decreased gradually with age. The levels of glycine and valine in plasma, alanine and valine in erythrocytes, serine plus asparagine, glycine, alanine, and tyrosine in granulocytes, and aspartic acid, serine plus asparagine, and alanine in lymphocytes remained constant in all age groups.(ABSTRACT TRUNCATED AT 250 WORDS)

The effect of protamine on serum complement activity in aminonucleoside nephropathy.

Combined administration of protamine sulfate (Ps) and aminonucleoside (AN) to rats causes severe nephrotic syndrome and, histologically, focal glomerular sclerosis. These changes are more distinct than those produced by AN alone. While the exact mechanism of Ps acting on AN nephropathy is not known, marked hypocomplementemia was observed regularly after the Ps + AN injection, suggesting that complement is activated and consumed in the kidney. It is suggested that complement may play an important role in AN + Ps nephropathy.

[Combined treatment with intra-arterial administration of oily anti-cancer agents and transcatheter arterial embolization--clinical and pathological study on 2 cases with resected small hepatoma].

Combined treatment with intraarterial administration of oily anti-cancer agent (SMANCS-Lipiodol; copolymer of styrene maleic acid conjugate of Neocarcinostatin disolved in Ethiodol) and transcatheter arterial embolization (TAE) was employed in 2 patients with small hepatoma. At 3-4 weeks after treatment, hepatic resection was performed. Histopathological examination of the 2 resected specimens showed total cell necrosis; CT and Softex revealed the distribution of lipiodol in the tumor and adjacent regions. This combination treatment showed combined effects, i.e. embolization by TAE, the anti-cancer effect of SMANCS and the selective delivery of lipidol to the hepatoma, especially the area of capsula invasion, the daughter nodule and tumor thrombus.

[Clinical studies on cefmenoxine concentration in prostatic tissue and bladder wall].

Clinical studies on cefmenoxime (CMX) concentration in prostatic tissue and bladder wall were made and the following results were obtained. In 40 patients undergoing operation, 1 g of CMX was administered intravenously by bolus technique and CMX levels in peripheral blood, prostatic tissue and bladder wall were examined. The maximum level of prostatic tissue was 35 micrograms/g after 0.41 hour and bladder wall was 62 micrograms/g after 0.37 hour, and T1/2 was 1.41 hours both. CMX, a new broad spectrum cephalosporin, can be considered as one of the highly useful antibiotics for the treatment of postoperative infection.

[Therapeutic results of malignant ovarian tumor].

One hundred and fifty patients with malignant ovarian tumors were treated in the Cancer Institute Hospital between 1949 and 1977. Most (103; 68.7%) of the patients had simple primary ovarian cancer. A retrospective study was performed in these 103 patients to investigate the relationship between the 5-year survival rate and the clinical stage and primary treatment. The 103 patients were classified according to FIGO criteria. The survival rates for stages I, II, III and IV were 73.7%, 50.5%, 17.4% and 0%, respectively. As the primary treatment, operations were performed on 92 of the 103 patients. To conclude, of primary importance in the treatment of ovarian cancer is the surgical, complete as possible removal of the tumors. Postoperatively, radiotherapy and chemotherapy should be used. Recently, second look operations have come into wider use. General procedures for the surgical and postoperative treatment of progressive ovarian cancer have been established. Although surgical procedures for the treatment of early ovarian cancer have also been established, there are no consistent procedures for postoperative treatment. Since the incidence of ovarian cancer is increasing, the development of appropriate methods for early diagnosis and treatment of this carcinoma is eagerly awaited.
