[Investigations for the use of micromodification of lymphocyte transformation test in patients with lung disease (author's transl)].

In a preliminary study a micromodification of lymphocyte transformation test was examined for use in patients with specific and nonspecific lung diseases. Investigation was carried out about the influence of different conditions in the cell culture system. A micromodification was defined and introduced in the clinic simultaneously to the conventional macromodification of lymphocyte transformation test. This micro lymphocyte transformation test is of use for determination of cell mediated immunity. However, in the described form it is doubtful for the detection of present specific sensibilization.

[Allergic alveolitis. Diagnosis and therapy in practice].

The diagnostic steps for the disease are listed and are discussed. The diagnosis includes: 1. the anamnesis, 2. the clinical examination, 3. the chest X-ray, 4. serological-immunological methods of examination including detection of antibodies, skin tests, allergen inhalation test, 5. lung function tests. The therapy consists in antigen exclusion and corticosteroids.

[Frequency of lung function disturbances of industrial poultry farmers (author's transl)].

The frequency of antibodies against organic dusts of fowls were examined on 339 poultry farmers. 22% of workers had 1 antibody and 3% had 2 antibodies in the serum. The ventilation, the arterial oxygen tension, the residual volume, the Slope Index, the closing volume of each person were measured in the mobile laboratory. Signs of central obstruction and equal ventilation were found in sensitized persons (2 antibodies) about four times more than non-sensitized persons. Suitable lung function screening parameters were the FEV1/FVC and the Slope Index.

[Sensitivity and specifitivity of the counterimmunoelectrophoresis for determination of precipitating antibodies (author's transl)].

Counterimmunoelectrophoresis (C.I.E.) is suitable for routine-work in clinical medicine to give quantitative evidence of precipitating antibodies. Sensitivity of this method is high (lowest level being 200 to 400 ng albumin/ml serum) and range of evidence being very broad (14-15 titer steps). The direction of migrations of proteins with anodal migration (Ig) is changed by carbamylation. The experience gained in more than 3,000 investigations, concerning methods and evaluation of the results, is reported in detail. Counterimmunoelectrophoresis is superior to immunoelectrophoresis and double diffusion (Ouchterlony) due to its higher sensitivity and to RAST or RIST by reason of its lower costs.

[Allergy and lectins: action between IgE-mediated histamine release and glycoproteins from Viscum album L. (mistletoe)].

Low-molecular constituents from Viscum album (toxic) release, in irritatively toxic manner, histamine from human leukocytes without destroying the cells (cytotox test). This histamine release is prevented by preincubation with viscum lectins. The viscum lectin blocks the Fc-portion of allergen-specific IgE against anti-IgE [125I]. Lectin also inhibits the allergen-specific histamine release from human leukocytes occurring through IgE; this can be demonstrated on leukocytes from patients with atopic extrinsic asthma. The inhibitory effect of lectin could also be due to a primary membrane effect, in addition to blockage of IgE.

[Immunoglobulin E--formation, determination and significance].

The detection of class E of immunoglobulins 10 years ago led to a decisive progress in the pathogenesis and diagnostics of allergic diseases. In the literary survey several essential findings concerning the structure of the IgE and concerning the quantitative and qualitative recognition of the total IgE and the allergen-specific freely circulating or cellularly fixed IgE-antibodies are cited. Diagnostically usable changes of concentrations of the level of total IgE are described.

[Isolation and characterization of mistletoe extracts (Viscum album L.). II. Effect of agglutinating and cytotoxic fractions on mouse ascites tumor cells].

Lectin from mistletoe (Viscum album L.) was studied for its relations with the toxins from Viscum album, ascites tumor cells of mouse, and human immunoglobulins. Using affinity chromatography on glutaraldehyde-crosslinked IgG (human) from viscum crude extract, a fraction was isolated which exhibited full agglutination capacity and high toxicity. The supernatant showed no agglutination capacity but a strong toxic effect on mouse ascites tumor cells. This toxic effect could not be influenced by further additions of insolubilized IgG. Chromatography on DEAE cellulose also gave agglutinating fractions with toxic effects and a non-agglutinating toxic portion. Column chromatography on Sephadex G 75 allowed separation of toxic from agglutinating components. The molecular weight of the toxin remaining after lectin removal was above 10,000. Lectin was found to bind more readily to mouse ascites tumor cells than to erythrocytes.

[Report on the application of various Kveim antigens in diagnostic of sarcoidosis (author's transl)].

In the Institute for Pulmonary Diseases and Tuberculosis, Novi Sad, a total number of 1500 patients was tested with a Kveim antigen prepared in this institute. Simultaneously these patients were tested with other antigens (SILTZBACH, BEHREND, DOUGLAS). Almost identical results could be found. In patients with sarcoidosis the Kveim test was positive in 60 to over 90%, depending on the stage of the disease. Within a cooperation 230 patients of the Research Institute for Pulmonary Diseases and Tuberculosis, Berlin-Buch, were tested with several Kveim antigens since 1972. 2/3 of these patients were suffering from sarcoidosis. All patients received Kveim antigen prepared in Novi Sad; one part of them was also tested with Kveim antigens prepared in Berlin-Buch. Dependent on the quality of the material processed the percentage of positive results was different. They varied from 65 to 30%. If there were less granulomas in the original material, as for the tissue of a spleen, used in Berlin-Buch, only 30% of sarcoidosis patients showed positive results. Therefore it is profitable to analyse the difficulties and the possibilities of improving the Kveim test as a valuable and rather simple diagnostical method.
