[Studies on the behavioral pharmacology of TRH-T in cytosine arabinoside-induced ataxic mice: a comparison with genetically ataxic mice].

The effects of thyrotropin releasing hormone tartrate (TRH-T) on the behavior of cytosine arabinoside (Ara-C) induced ataxic mice were studied. The ataxic mice were prepared by injecting 50 mg/kg/day subcutaneously on the 2nd, 3rd and 4th postnatal days. Spontaneous motor activities were measured with a movement spectrum analyzer (ANIMEX-2) and the degree of ataxic gait determined by an open-field study. Cerebella from these animals were subjected to histopathologic examination at 4, 8 and 12 weeks of age. Ataxic gait became improved moderately and spontaneous motor activities increased slightly following administration of TRH-T. The effects of TRH-T on ataxia and spontaneous motility in the Ara-C injected mice varied with the age, and resembled those seen in weaver mice but not those in the Rolling mouse Nagoya. The results also suggest that the effects of TRH-T on the behavior of ataxic mice correlate with the extent of pathologic changes evoked in their cerebella.

Increased urinary kallikrein excretion in young borderline hypertensive patients.

Urinary kallikrein excretion was measured in 46 young patients with borderline hypertension and 28 age-matched normotensive subjects. Hypertensives excreted greater amounts of kallikrein than normotensives (2.31 +/- 0.20 units/day vs. 1.56 +/- 0.17 units/day, p less than 0.01). Plasma renin activity (PRA) was also increased in hypertensives. Moreover, urinary kallikrein was increased in hypertensive patients with high PRA (PRA greater than or equal to mean + 1SEM in normotensives; n = 25) as compared to patients with normal PRA (PRA less than mean + 1SEM; n = 21). In hypertensives with normal PRA, urinary aldosterone correlated to urinary kallikrein (r = 0.478, p less than 0.05), as in normotensives (r = 0.451, p less than 0.02). But, no correlation was found in patients with high PRA. Therefore, the results of the present study do not confirm the hypothesis that the deficiency of the kallikrein-kinin system is the primary cause of hypertension. In hypertensives with high PRA, there may be abnormality of the interaction between the renin-angiotensin-aldosterone system and the kallikrein-kinin system, whereas it may be normal in hypertensives with normal PRA as well as normotensives.

Effects of an antitumor agent, ascofuranone, on the macromolecular syntheses of intact cells.

Ascofuranone (AF) has antitumor protective property on experimental tumors. We examined the action of AF on lymphoma L5178Y to explore the mechanism of the antitumor activity. AF completely prevented the growth of L5178Y at 25 micrograms/ml cytostatically. The compound exhibited general inhibitory effects on the macromolecular syntheses. Among them, protein synthesis was most severely inhibited by AF and to the same extent as by cycloheximide. AF, however, did not affect protein synthesis by cell-free system even at 2 mg/ml. Although AF inhibited the incorporation of [14C]acetate into total acid precipitable products only slightly, the synthetic pattern of simple lipids from [14C]acetate was significantly changed. Especially, the incorporation of [14C]acetate into squalene was almost completely blocked at 25 micrograms/ml. The incorporation of [14C]acetate into triglyceride was inhibited and that into cholesterol was enhanced. Concerning the diglycerides, the incorporation of [14C]acetate was enhanced and that of [3H]glycerol was inhibited. The incorporation of [3H]glycerol and [3H]mevalonate into the intact cell was significantly inhibited as compared with [14C]acetate. As those effects were not observed with cycloheximide, they were suggested to be characteristic of AF. AF inhibited hypotonic hemolysis. In contrast, hemolysis by deoxycholate was stimulated. Possible mechanism of the antitumor activity of AF is discussed.

[Tumor marker in urology].

Tumor marker for tumors in urology has been widely used to testicular and prostatic tumors. A part of testicular tumor produces alpha-fetoprotein (AFP) and HCG, thus these markers can not be used for early detection of disease. However, they are very useful in typing testicular tumor, and in monitoring a course of disease which produces them. beta-HCG seems to be more specific than HCG. In case of prostatic cancer, prostatic acid phosphatase (PAP) assayed immunochemically is sensitive and specific marker. Prostate antigen seems to be another excellent marker for this tumor, and this is well correlated with PAP. In reactivated case, tissue polypeptide antigen was elevated, suggesting use of this marker.

[Biochemistry of occupational bladder cancer].

We herein discuss the biochemistry of occupational urinary tract cancer, by reviewing the history of occupational aromatic amine cancer, experimental production of aromatic amine cancers and aromatic amine double cancer.
