[Study on novel mutations of MEF2A gene in Chinese patients with coronary artery disease].

OBJECTIVE: To explore the mutations of MEF2A gene in Chinese patients with coronary artery disease(CAD). METHODS: With polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) and DNA direct sequencing, the mutation analysis of exon 11 of MEF2A gene was performed to 156 patients with CAD and 93 normal controls. RESULTS: By DNA sequence analyzing the samples of abnormal mobility shift of SSCP, the MEF2A gene mutations were found in three patients with CAD. One of mutations was 147130(C>A)(P431Q), and the second one was 21 bases deletion(147108-147128) which was leading to the absence of 7 amino acids (424QQQQQQQ430), and the third was 147191(G>T). Three mutations were all found in one patient, but meanwhile 21 bases deletion was found in the other two patients. CONCLUSION: Mutations in exon 11 of MEF2A gene exist in the patients with CAD, and the mutations may be pathological.

[Evaluation on environmental quality of Pb, Zn and Cu contents in vegetable plantation soils and vegetables in Hangzhou suburb].

Contents of heavy metals (Pb, Zn, Cu) in soils from 4 main vegetable plantations in Hangzhou suburb were investigated to evaluate environmental quality of heavy metals of soils and vegetables. Different evaluation methods and evaluation standards of heavy metals were used for comparison. Kriging method was applied to study the spatial variability of heavy metals in soil-vegetable system. The distribution maps of the spatial variability of heavy metals in soil-vegetable system in Jianggan District were obtained based on GIS method. The results show that the soils were polluted with Pb, Zn, and Cu using natural background values as evaluation standard. While using National Standard as evaluation standard, the soils were not polluted. Contents of heavy metals in Gongshu District were the highest of four investigated districts. The concentration distribution of heavy metals had different spatial variability. The content of Pb in vegetables was higher than those of Zn and Cu and exceeded the national vegetable sanitation standard. The order for their enrichment coefficients was as follows: Zn> Pb> Cu. The accumulation coefficient for green vegetable was higher than that of Chinese cabbage, indicating green vegetable could easily accumulate heavy metals from soils than Chinese cabbage.

[Testing of seed-borne fungi of Glycyrrihiza seed and disinfection effect of several fungicides on seed-borne fungi].

OBJECTIVE: To study the dominant seed-borne fungi of Glycyrrihiza seeds which were from different producing area and compare the disinfection effect of several fungicides on seed-borne fungi of Glycyrrihiza seed. METHOD: Petri-dish testing was used to determine the external and internal seed-borne fungi and the disinfection effect of fungicides. RESULT AND CONCLUSION: The result showed that the amount of spore on the surface of one Glycyrrihiza seed varied from 0.3% to 37.0% among samples. Penicillium spp. and Aspergillus spp. were the two major dominant fungi and there was few differences in the type of the fungi among producing areas but differences in the isolation frequency of the furgi; Penicillium spp. , Rhizopus spp. , Aspergillus spp. and Alternaria spp. were the internally dominant seed-borne fungi, including seed capsule and the internal tissue of seed and their fungi-carrying percentage was 8.0%-48.3% and 3.5%-42.0% respectively. There were differences in fungi percentage and dominant seed borne fungi among different producing areas. The disinfection effect of both mancozeb and thriam was up to 89.0%, so it is suggested to use them to disinfect Glycyrrihiza seed.

[Study on the method of using ICP-MS to determine Se in the edible fungi].

The sample was digested by the quartzose digestion pot at high-pressure under relatively low temperature, reducing the loss of Se during the digestion process. Study on the method of using ICP-MS to determine Se in the edible fungi shows that it exhibits positive correlation between the concentration of sample and integration time, and the response value. When the concentration of Se in the sample is higher than 50 microg x L(-1), the integration time can be 0.1 s; or it can be 2.0 s when the concentration of Se is lower than 5 microg L(-1). The standard curve was accomplished for 1.0-500 microg x L(-1) when determining the content of Se in the edible fungi. The recoveries are in the range of 99.96%-102.7%. This method has advantages of simplicity, speediness, delicacy, high stability, high accuracy, etc. It is suitable for the determination of trace Se in the edible fungi.

[Clinicopathologic features of gallbladder adenocarcinoma with marked stromal fibrosis--a report of 19 cases].

BACKGROUND & OBJECTIVE: Macropathologic types of gallbladder cancer are mostly polyp type, intumescent type, and cauliflower form lump. Its histological types include well or poorly differentiated adenocarcinoma, mucinous adenocarcinoma, and undifferentiated cancer. This research was to explore the clinicopathologic features of gallbladder adenocarcinoma with marked stromal fibrosis. METHODS: Pathology of 19 cases of gallbladder adenocarcinoma with marked stromal fibrosis was observed using a light microscopy and SP immunohistochemistry. Clinicopathologic features of 19 patients were analyzed. RESULTS: Most of the patients had long-term history of cholecystitis gallbladder calculus. B ultrasound showed that the gallbladder wall was irregularly thickened or presented nodosity. Observed with naked eyes, gallbladder adenocarcinoma with marked stromal fibrosis did not form cancer nodule and extrude into the gallbladder lumen, the gallbladder wall showed regional thickening, a few cases showed diffuse irregular thickening. Observed under a light microscope, the adenocarcinoma cells were mostly arranged as single layers, seldom arranged as multiple layers, and formed adenoid structures with different sizes, various shapes, and irregular arrangement; the nuclei were heterogenic with haryomitosis presented in a few cases; inflammatory cells were infiltrated in hyperplastic fibrous connective tissue of some cases. According to immune phenotyping, CK (AE1/AE3), CK (AE1), CK7 (OV-TL12/30), CK8 (C51), CK18 (Dc-10), CK19 (RCK108), and EMA (Mc-5) showed strong expression, CEA (COL-1), CK20 (Ks20. 4), and MUC-5AC (CLH2) showed moderate expression, and MUC-2 (B306. 1) showed weak expression; CK17 (E3) showed focal expression. CONCLUSIONS: The clinical manifestation, macropathologic type, histological characteristics of gallbladder adenocarcinoma with stromal fibrosis are different from other types of adenocarcinoma. Its genesis may be related to chronic cholecystitis: long-term inflammation causes regional hyperplasia and heterogeneity of the gland body, lead to focal or regional thickening of the gallbladder wall, and result in gallbladder adenocarcinoma with stromal fibrosis finally.

[An analysis of islet beta-cell function in hyperuricemia].

OBJECTIVE: To study the islet beta-cell function in patients with simple hyperuricemia (HUA) and to explore the relationship between the level of serum uric acid and insulin resistance. METHODS: The first and second phases of insulin secretion of beta-cell were observed after L-arginine stimulation test and oral glucose tolerance test (OGTT) in 56 hyperuricemic patients and 40 healthy controls. RESULTS: The peak value of insulin secretion in both groups was at the 2nd min and it began to decrease at the 4th min after L-arginine stimulation. However, the peak value in the HUA group was significantly higher than that in the control group (t = 3.71, P < 0.01). Fasting plasma insulin, 120-min plasma glucose and 120-min insulin during OGTT as well as modified beta-cell function index and Homa-insulin resistance (HOMA-IR) in the HUA group were obviously higher than those in the control group (t = 2.46-5.56, P < 0.05, P < 0.01). Multiple step regression analysis showed that serum uric acid was positively correlated with low density lipoprotein-cholesterol, HOMA-IR, male sex, mean arterial pressure and creatinine (t = 2.15-3.60, P < 0.05). CONCLUSION: The islet beta-cell function in hyperuricemic patients increases compensatively and the serum uric acid level is positively correlated with insulin resistance.

[Effect of sulfated polymannuroguluronate on Tat induced proinflammatory cytokines release in THP-1 cells and its mechanism of action].

AIM: To investigate the effects of sulfated polymannuroguluronate (SPMG), a novel candidate anti-AIDS drug in Phase II clinical trial, on Tat-induced release of proinflammatory cytokines (i.e., TNFalpha, IL-1beta and IL-6) and its related mechanism. METHODS: The effects of SPMG on Tat induced TNFalpha (4 h), IL-1beta and IL-6 (6 h) secretion in THP-1 cells were measured by ELISA. Western blotting analysis was used to study the effects of SPMG on Tat induced PKCzeta, PKCtheta and PKCsigma phosphorylation. RESULTS: SPMG (50 to 100 microg x mL(-1)) markedly suppressed TNFalpha, IL-1beta and IL-6 secretion in Tat activated THP-1 cells. In THP-1 cells the phosphorylation levels of PKCzeta, PKCtheta and PKCsigma significantly increased following Tat stimulation, and only PKCsigma phosphorylation levels was inhibited by SPMG (50 to 100 microg x mL(-1)). CONCLUSION: SPMG suppresses the secretion of proinflammatory cytokines in THP-1 cells may be by inhibiting PKCsigma activation.

[Construction of prokaryotic expression system of Salmonella paratyphi A spaO gene and immunogenicity and immunoprotection of the expressed product].

OBJECTIVE: To study the immunogenicity and immunoprotection of the recombinant expressing product (rSpaO) of S. paratyphi A spaO gene, and to demonstrate the frequencies of spaO gene carrying and expressing in S. paratyphi A isolates. METHODS: The spaO gene of a clinical S. paratyphi A strain JH01 was amplified and then cloned. After sequencing of the cloned spaO gene, a prokaryotic expression system of the gene was constructed. SDS-PAGE were applied to examine the rSpaO expression. Ni-NTA affinity chromatography was performed to collect rSpaO. Immunogenicity of rSpaO was determined by Western blot assay. A PCR assay and an ELISA were established to respectively detect the carrying and expressing frequencies of the spaO genes in 98 S. paratyphi A isolates. The immunoprotective effects of rSpaO in S. paratyphi A strain 50001 infected mice were observed. RESULTS: In comparison with the reported corresponding sequences, the nucleotide and putative amino acid sequence homologies of the cloned spaO gene were 99.45%-99.89% and 99.01%-100%, respectively. The expression output of rSpaO was approximately 75% of the total bacterial proteins. S. paratyphi A antiserum could recognize as well as combine with rSpaO. rSpaO could efficiently induce rabbits to produce specific antibody. 94.9% (93/98) of the S. paratyphi A isolates had spaO gene and 91.4% (85/93) of the spaO+ strains could express SpaO. 58.3% and 50.0% of the mice that oral-taken or subcutaneous injected with 500 microg of rSpaO for immunization were survival after challenged by lethal dose of S. paratyphi A strain 50001. When co-immunized with 5 microg rLTB, the survival rates of the mice increased to 88.3% and 75.0%, respectively. CONCLUSION: The S. paratyphi isolates had relatively high carrying and expressing frequencies of spaO gene. rSpaO showed a fine immunogenicity and a certain immunoprotective effect, which could be used as an antigen candidate for developing genetic engineering vaccine of S. paratyphi.

Protection of mice from lethal endotoxemia by chimeric human BPI-Fcgamma1 gene delivery.

To evaluate the potentiality of applying gene therapy to endotoxemia in high-risk patients, we investigated the effects of transferring an adeno-associated virus serotype 2 (AAV2)-mediated BPI-Fcgamma1 gene on protecting mice from challenge of lethal endotoxin. The chimeric BPI-Fcgamma1 gene consists of two parts, one encods functional N-terminus (1 to 199 amino acidic residues) of human BPI, which is a bactericidal/permeability-increasing protein, and the other encodes Fc segment of human immunoglobulin G1 (Fcgamma1). Our results indicated that the target protein could be expressed and secreted into the serum of the gene-transferred mice. After lethal endotoxin challenge, the levels of endotoxin and TNF-alpha in the gene-transferred mice were decreased. The survival rate of the BPI-Fcgamma1 gene-transferred mice was markedly increased. Our data suggest that AAV2-mediated chimeric BPI-Fcgamma1 gene delivery can potentially be used clinically for the protection and treatment of endotoxemia and endotoxic shock in high-risk individuals.

[Effect of rapamycin and dexamethasone on differentiation and maturation of murine bone marrow-derived dendritic cells].

AIM: To observe the effect of rapamycin (Rap) and dexamethasone (Dex) on differentiation and development of murine bone marrow-derived dendritic cells (DC) in vitro. METHODS: DC cells generated from C57BL/6 murine bone marrow cells were induced by GM-CSF and IL-4. During the course, Rap or Dex was added to the culture and the cells were then stimulated by LPS. (1)Morphology development of DCs was observed by inverted microscope and scanning electron microscope.(2)The cells were analyzed by flow cytometry (FCM) to determine the proportion of CD11c(+) cells and the change of CD86 and MHC class II molecule.(3)The influence of DCs treated by Rap or Dex on the allogeneic T cell proliferation was studied by one-way MLR. RESULTS: (1)The morphology of DCs maintained in a durable state of immaturity after Rap or Dex pretreatment.(2)The expression of CD11c and MHC-II slightly decreased but CD86 dramatically reduced on Rap-treated DC cells. There was close relationship between the expression of CD11c on Dex-treated cells and the dosage of Dex. The surface expression of CD86 and MHC-II dramatically reduced on Dex-treated DCs. Moreover, DCs treated by Rap or Dex were both able to resist the maturation triggered by LPS.(3)Bone marrow-derived DCs cultured with Dex or Rap had a lower stimulatory effect on allogeneic T cells compared with that of mature DCs. CONCLUSION: Rap and Dex could keep DCs in durable immaturity. Compared with Dex, which inhibited the expression of co-stimulatory molecules, Rap hardly influenced the differentiation of DCs and the expression of MHC class II molecules.

[Diagnosis and treatment of cauda equina syndrome secondary to lumbar disc herniation].

OBJECTIVE: To discuss the diagnosis and treatment of cauda equina syndrome. METHODS: Thirty-four cases of cauda equina syndrome from 1993 to 2005 were analysed retrospectively. Its clinical representation, pathology character and clinical outcome were observed. RESULTS: At follow-ups of 3 to 102 months after the treatment (averagely 43 months), the muscle strength and function of the constrictor began to resume to different degrees. Muscle strength resumed gradually one week after the operation, resumed faster 3 months after the operation, and resumed completely more one year later. Function of the constrictor began to resume 3 days after the operation, and resumed mostly six months or so after the operation. Sense resumed slowly. CONCLUSION: Clinical outcome of cauda equina syndrome is good. Resumption after the operation obviously correlates to the time of operation. One week after the operation, the kinetic function resumes best, function of the constrictor resumes well and the sense function of the saddle area resumes not very well.
