Curry leaf (Murraya koenigii Spreng.) reduces blood cholesterol and glucose levels in ob/ob mice.

We observed that curry leaf (Murraya koenigii) extract possesses the property to decrease blood cholesterol and blood glucose levels in diabetic ob/ob mice. Mice received daily intraperitoneal injections of 80 mg/kg curry leaf extract for 10 consecutive days. The extract significantly decreased blood cholesterol level from 277.6 +/- 16.6 mg/d (day 0) to 182.0 +/- 15.3 mg/d (day 10, p < 0.01 compared with the change in vehicle group). The extract also significantly decreased blood glucose level from 387.0 +/- 15.6 mg/dl (day 0) to 214.0 +/- 26.6 mg/dl (day 10, p < 0.01). In addition, body weight was reduced after extract treatment. Our data suggest that curry leaf may be proved to be of clinical importance in improving the management of high cholesterol level and type 2 diabetes.

Defensins HNP1 and HBD2 stimulation of wound-associated responses in human conjunctival fibroblasts.

PURPOSE: To study the responses of human conjunctival fibroblasts (HCFs) to stimulation by human neutrophil defensin 1 (HNP1) and beta defensin2 (HBD2). METHODS: Defensin-stimulated gene expression in primary cultures of HCFs was analyzed by real-time PCR after exposure to various concentrations of HNP1 or HBD2. Gene and protein expression for selected collagens, matrix metalloproteinases, and tissue inhibitors of metalloproteinases were determined by real-time PCR and ELISA analysis. Activation of p42/44 mitogen-activated protein (MAP) and Akt was analyzed by Western blot. RESULTS: HCFs did not express significant levels of the genes for HNP1 or HBD1-3. However, HNP1 and HBD2 stimulated HCF proliferation, the activation of p42/44 MAP kinase, and Akt kinase in a dose-dependent manner. HNP1 and HBD2 were not found to be chemotoxic for HCFs. It was demonstrated with the use of U0126 and wortmannin that the activation of p42/44 MAP kinase and Akt was responsible for the increased HCF proliferation observed under HNP1 and HBD2 stimulation. HNP1 stimulated the expression of the genes for collagen I, III, VI, and VIII. In addition, it reduced the secretion of collagen I protein but increased its intracellular retention. HNP1 and HBD2 upregulated the transcription and translation of MMP1. Small increases were observed in MMP14 gene expression after HNP1 stimulation and MMP2 gene expression after HBD2 stimulation. CONCLUSIONS: The results of this study suggest that HNP1 and HBD2 have a potential role in the biosynthetic and tissue remodeling responses of conjunctival fibroblasts.

Growth inhibition and differentiation induced by peroxisome proliferator activated receptor gamma ligand rosiglitazone in human melanoma cell line A375.

BACKGROUND: Ligands of peroxisome proliferator-activated receptors (PPARs) have been demonstrated to be antitumorgenic in vitro and in vivo due to their antiproliferative, prodifferential, and antiangiogenic effects. The aim of this study is to evaluate the effects and mechanisms of PPARgamma ligand rosiglitazone (ROZ) on the growth, apoptosis, and differentiation in human melanoma cancer cell line A375. METHODS: The effects of ROZ on A375 cell proliferation were measured by clonogenic assay, apoptosis and cell cycle kinetics by FACS with ROZ for 72 h, PPARgamma protein was detected by Western blot analysis and immunocytochemical staining, and PPARgamma mRNA expression by RT-PCR. The differentiation effect of ROZ was determined by measurement of melanin content and tyrosinase activity. The levels of Bcl-2, P53, p-ERK, and ERK were also detected by Western blot analysis. Inhibition of tumorigensis was observed in nude mice. RESULTS: ROZ inhibited colony formation and induced apoptosis in A375 cells, and the cells were arrested in G1 phase. This effect was associated with a decrease of the expression of Bcl-2 and increase of the expression of P53. ROZ also induced the differentiation in A375 cells. ROZ increased expression of PPARgamma and decreased the expression of ERK and p-ERK. Data in vivo showed that ROZ could inhibit tumorigensis in nude mice. CONCLUSION: These results demonstrated that ROZ inhibited growth of A375 cells via the induction of apoptosis, necrosis, and differentiation in a PPARgamma-dependent manner and might present a promising therapeutic approach in certain human maligancies.

Field demonstration of reduction of lead availability in soil and cabbage (Brassica Chinensis L.) contaminated by mining tailings using phosphorus fertilizers.

A field demonstration of reduction of lead availability in a soil and cabbage (Brassica Chinensis L.) contaminated by mining tailings, located in Shaoxing, China was carried out to evaluate the effects of applications of phosphorus fertilizers on Pb fractionation and Pb phyto-availability in the soil. It was found that the addition of all three P fertilizers including single super phosphate (SSP), phosphate rock (PR), and calcium magnesium phosphate (CMP) significantly decreased the percentage of water-soluble and exchangeable (WE) soil Pb and then reduced the uptake of Pb, Cd, and Zn by the cabbage compared to the control (CK). The results showed that the level of 300 g P/m(2) soil was the most cost-effective application rate of P fertilizers for reducing Pb availability at the first stage of remediation, and that at this P level, the effect of WE fraction of Pb in the soil decreased by three phosphorus fertilizers followed the order: CMP (79%)>SSP (41%)>PR (23%); Effectiveness on the reduction of Pb uptake by cabbage was in the order: CMP (53%)>SSP (41%)>PR (30%). Therefore our field trial demonstrated that it was effective and feasible to reduce Pb availability in soil and cabbage contaminated by mining tailings using P fertilizers in China and PR would be a most cost-effective amendment.

Highly sensitive spectrofluorometric determination of trace amounts of mercury with a new fluorescent reagent, 2-hydroxy-1-naphthaldehydene-8-aminoquinoline.

A new fluorescent reagent, 2-hydroxy-1-naphthaldehydene-8-aminoquinoline (HNAAQ), was synthesized. The fluorescent reaction of this reagent with mercury was also studied. Based on this chelation, a highly sensitive spectrofluorometric method was developed for the determination of trace amounts of mercury in a water-ethanol (5 + 1, v/v) medium at pH 8.0. Under these conditions, the Hg-HNAAQ complex has excitation and emission maxima at 406 and 445 nm, respectively. The linear range of the method is from 0 to 16 microg L(-1) and the detection limit is 0.056 microg L(-1) of mercury. The interference of other ions was studied. In order to enhance the selectivity in the determination of mercury by the present method, we also applied the separation of mercury by distillation. Thus, the selectivity of the method could be increased remarkably. The procedure can be easily performed, and affords good precision and accuracy. This method has been successfully applied to the determination of mercury in waste water and prawns.

[The wild-type alpha-synuclein over-expression to induce the protein aberrant aggregation of alpha-synuclein in HEK293 cells in vitro].

OBJECTIVE: To investigate over-expression of wild-type alpha-synuclein inducing the aberrant aggregation of alpha-synuclein in HEK293 cell in vitro. METHODS: The cDNA encoding the human alpha-synuclein without the stop code was cloned into PGEM T-easy vector. Using enzyme map and DNA sequencing analyzed and determined the recombinant plasmid, and then sub-clone the alpha-synuclein cDNA fragment into pEGFP-N1 vector. The recombinant plasmids alpha-synuclein-pEGFP were transfected into HEK293 cells by lipofectamin 2000. The aberrant aggregation of alpha-synuclein was measured by EGFP fluorescence, anti-alpha-synuclein immunocytochemistry. The inclusions in the cultured cells were identified with HE staining. RESULTS: The restriction enzyme map suggested that eukaryotic expression vector for human wild-type alpha-synuclein gene was constructed successfully. By EGFP fluorescence, anti-alpha-synuclein immunocytochemistry, it could be observed that the alpha-synuclein protein could aggregate in cytoplasm and the Lewy body-like inclusions found in cytoplasm of cultured cells. CONCLUSION: The over-expression of wild-type alpha-synuclein can induce protein aberrant aggregation and Lewy body-like inclusions formation in cytoplasm of HEK293 cell in vitro.

Human limbal progenitor cell characteristics are maintained in tissue culture.

INTRODUCTION: To determine the differentiation of human limbal epithelial cells in tissue culture. MATERIALS AND METHODS: Epithelial cells from the human limbus (n = 29) were isolated and cultured in supplemental hormonal epithelial medium (SHEM) in the presence of mitomycin C-treated 3T3 feeder layer. Confluent cells were airlifted to form multiple layers. The expression of cytokeratin 3 (K3), cytokeratin 12 (K12), involucrin, connexin 43 (Cx43), proliferation cell nuclear antigen (PCNA) and p63 was studied in normal and airlifted cells by immunohistochemistry. Expression levels of K3 and K12 mRNA were examined by real-time polymerase chain reaction (PCR). RESULTS: The colony-forming efficiency of primary cultured (P0) cells was about 19.35 +/- 6.46% (mean +/- SD, n = 7). Real-time PCR analysis showed that the transcription level of K3 and K12 in cultured cells was lower than in freshly isolated limbal cells or cells from central cornea (P <0.01). Few cells were positive for K3 in P0 or P1 cells [(1.99 +/- 1.27)% (n = 7, P0) and (3.96 +/- 1.35)% (n = 4, P1), P = 0.046]. More cells at all levels were found to stain positive for PCNA and p63 as compared to K3, K12 and involucrin. After air-lifting, cell sheets of 3 to 5 epithelial cell layers formed. Involucrin showed positive staining in suprabasal layers of the cell sheets while connexin 43 was only observed in the basal layer. Staining of K3 remained sparse. CONCLUSIONS: Human limbal cells isolated from cadaveric tissues were able to proliferate in vitro and exhibited a phenotype with characteristics similar to that of the limbal stem or progenitor cells.

[A preliminary study on the mechanism of neurotoxicity of MDMA--oxidative stress harm].

OBJECTIVE: Establishing a long-term neurotoxic model to explore the mechanism of neurotoxicity of 3,4-methylenedioxymethamphetamine (MDMA) and the putative protection conferred by Vit C against oxidative stress harm. METHODS: Male Wistar rats were randomly assigned to control group (A) and MDMA treatment groups(B, C, D, E). Rats of group B were given MDMA 20 mg/kg; groups C, D, E were given Vit C 250 mg/kg 30 min before administration of MDMA (Vit C 30 min group) and 3 h (Vit C 3 h group) and 5 h (VitC 5 h group) after administration of MDMA, respectively. Rats of control group were treated with the same volume of saline. Concentrations of ATP and ADP in brain cortex and 5-HT in hippocampus and occipital cortex were measured by high perfor-mance liquid chromatography; the expression of SERT mRNA was detected by in situ hybridization; and the expression of protein GFAP was detected by immunohisto-chemistry. RESULTS: hours after MDMA treatment, the concentration of ATP in brain cortex was lessened, compared with control (P <0.05). On the 7th day after MDMA treatment, the concentration of 5-HT in rat hippocampus and occipital cortex was decreased, compared with control (P<0.05). The expression of SERT mRNA in hippocampus was decreased, whereas the expression of GFAP in brain tissue was increased (P<0.05). The adminstration of Vit C 30 min before MDMA treatment and 3 h after MDMA treatment did not curb the decrease of ATP, 5-HT and the expression of SERT mRNA, but Vit C administrated 5 h after MDMA treatment could curb the decrease of ATP and the functional markers of 5-HT. And Vit C given at three time points did downregulate the GFAP expression. CONCLUSION: MDMA could deplete the direct energetic substance ATP. MDMA could exert neurotoxic effect on 5-HT system. Vit C given 5 h after MDMA administration could provide neuroprotection for ATP and 5-HT system.

[The influence of bonding strength of denture soft reline resin by different surface treatments].

PURPOSE: This study evaluated the influence of bonding strength of heat-polymerized denture soft reline resin by 4 different surface treatments bonded to denture base resin. METHODS: Denture base resin (8mm in diameter and 15 mm in length) were fabricated from heat-polymerized acrylic resin (Densply). And heat-polymerized denture soft reline resin disks (Physio liner) were also made (4 mm thick and 10 mm diameter). Two denture base resins were bonded to the denture reline resin disk at the centure on the two sides of the disk. All the specimen were divided into 4 groups (n=10) with 3 different surface treatments: (1) no treatment(control), (2) sandblasted, (3) surface liner application, (4) surface liner application+sandblasted. Bond strength (MPa) was measured by tension bond testing at a 12.7 mm/min speed until the soft reline resin debonded from the base resin. Data were statistically analyzed by SPSS 10.0 software package for ANOVA. RESULTS: The mean bond strengths of group 1, 2, 3, 4 were 3.518, 2.834, 4.077, 3.852MPa respectively. The bonding strength of all the three groups showed significant difference (P<0.0014). The group with surface liner application showed significantly highest bonding strength, while the group with sandblasted showed the lowest bonding strength. CONCLUSIONS: The use of surface liner can significantly improve the bonding strength of heat-polymerized denture soft reline resin to the denture base resin, while the bonding strength decreased after using sandblasting.

[Effects of Cd on rape growth and antioxidant enzyme system].

The study with pot experiment showed that the fresh weights of rape plant and leaf decreased with increasing Cd concentration, and the decrease of leaf fresh weight resulted in the decrease of whole plant fresh weight. The permeability of cell membrane and the content of MDA increased with increasing Cd concentration. When the concentration of Cd was 20 mg x kg(-1), compared with the control, cell membrane permeability and MDA content were increased by 29.68% and 15.19% respectively, and there was a significant correlation between these two indices. When the Cd concentration was 5 mg x kg(-1), the contents of chlorophyll a, b and a + b reached their peaks, and were 23.97%, 33.63% and 26.45% higher than the control, respectively. Cd stress had no significant effect on carotenoid content. The sensibility of the pigments to Cd stress was chlorophyll b> chlorophyll a> chlorophyll a + b> carotenoid, and that of protective enzymes was POD>CAT>SOD. The IC50 of test physiological indices suggested that rape could only grow well on the soil with < 5 mg x kg(-1) Cd.

[Value of CT scanning in the diagnosis of early cervical carcinoma].

OBJECTIVE: To investigate the value of CT scanning in preoperative diagnosis of early cervical carcinoma. METHODS: Abdominal and pelvic CT scans (MPR pictures for 50 patients) in 102 cervical carcinoma patients proven by biopsy pathology were retrospectively analyzed and compared with surgical pathologic findings. The pathological stages included > or = Ia in 39 patients, Ib 1 in 35, Ib 2 in 21, IIa in 6, IIb in 1. The tumor detection ability, thickness of cervical tumor invasion, T stage and lymph node stage showed by CT scan were evaluated. RESULTS: CT was not able to detect < or = Ia cervical carcinoma, however, CT was able to detect tumors in 71.4% of > or = Ib 1 stage (45/63) and 71.4% > or = Ib 2 (20/28), it also showed the depth of tumor invasion in 39.7% > or = Ib 1 (25/63). The overall accuracy of CT staging for tumor was 69.6% and the rate of metastatic lymph node detection 63.6% (7/11). With additional MPR pictures of spiral CT, the detection ability, vaginal involvement and objective shape of cervical tumor was improved. The cervical carcinoma in CT scan was found to be as homogeneous in 54.9% of patients (56/102), hypodense or heterogeneous in 42.2% (43/102), lower density nodual in 41.9% (18/43); occasionally as heterogeneous or hyperdensity noduals or irregular border of the cervix. The features of metastatic lymph nodes in CT scan were rounded or spheroid shape of soft tissue noduals with heterogeneous density caused by central necrosis and rim enhancement. CONCLUSION: CT scanning may be valuable in detection of > or = Ib 1 rather than < Ib 1 stage early cervical carcinoma. The additional MPR pictures of spiral CT is not only helpful in detecting tumor but also in determining the extension of cervical carcinoma.
