[Progress in leukocytospermia research].

Leukocytospermia is a most common cause of male infertility, but the distribution, origin and role of leukocytes in semen are still controversial. Some reports on leukocytospermia have indicated its negative effects on semen parameters and even in vitro fertilization (IVF). Recent literature has made it clear that the most deleterious effect of leukocytospermia is that the increased reactive oxygen species (ROS) may cause sperm damage, leading to significantly increased male infertility. The treatment and prevention of leukocytospermia have been proven of help for improving semen parameters.

[Characteristics of human adrenal microvascular endothelial cells in morphology, phenotypes, and function].

OBJECTIVE: To investigate the phenotypic and functional characteristics of human adrenal microvascular endothelial cells (AdrEC). METHODS: AdrEC were isolated and purified from a sample of human adrenal tissue by sub-cell clone method. The cells identified by flow cytometry for classical endothelial markers von Willebrand factor (vWF) and CD31, uptake of Dil-labeled acetylated low density lipoprotein (Dil-Ac-LDL), as well as phenotypes. The cell fenestrations were checked by scanning electron microscopy. The expressions of endogenous vascular endothelial growth factor (VEGF) mRNA and protein were detected by reverse transcription-polymerase chain reaction (RT-PCR) and immunocytochemistry. The glucocorticoid-induced cytotoxicities in different organs-derived microvascular endothelial cells were compared. RESULTS: Human AdrEC expressed those classical endothelial markers such as vWF, CD31, and uptake of Dil-Ac-LDL. The phenotypic analysis indicated that alpha-1 proteinase inhibitor, tumor necrosis factor receptor p55, and intercellular adhesion molecule-1 were expressed in human AdrEC. Scanning electron microscopy demonstrated that there were many microvilli and fenestrations on cellular surface. RT-PCR and immunocytochemistry showed that there was expression of endogenous VEGF in AdrEC. In response to glucocorticoid-induced cytotoxicity, microvascular endothelial cells (MVEC) derived from human brain were highly susceptible, MVEC derived from human lung and human liver sinusoidal endothelial cells were sub-sensitive, while AdrEC were highly resistant. CONCLUSION: Human AdrEC are specially differentiated and have characteristics that are different from other organ-derived MVEC in phenotypes and functions.

[Enzyme activities in Paeonia ostii rhizosphere and non-rhizosphere soil of Tongling copper mining].

The study on the calatase, polyphenol-oxidase, invertase, urease and phosphatase activities in Paeonia ostii rhizosphere and non-rhizosphere soil of Tongling copper mining showed that all test enzyme activities were higher in rhizosphere than in non-rhizosphere soil. Soil calatase, urease and phosphatase were sensitive to heavy metals pollution, and their activities could be used as the indicators of heavy metals' joint pollution. The effects of rhizosphere environment on the soil enzyme activities were in the sequence of phosphatase > urease > calatase > invertase > polyphenol-oxidase, and the affecting rate was 131.562%, 92.492%, 87.557%, 59.673% and 34.076%, respectively. The test enzyme activities were negatively correlated with soil heavy metals pollution, and the correlation coefficients were all higher than -0.898, suggesting the inhibitory effects of heavy metals' joint pollution on soil enzyme activities. P. ostii could effectively improve soil environment, and thus, enhance the activities of soil enzymes.

[Comparable effects of percutaneous transluminal septal myocardial ablation and subaortic ventriculotomy in treating patients with hypertrophic obstructive cardiomyopathy, an echocardiography follow-up study].

OBJECTIVE: To compare the effect of percutaneous transluminal septal myocardial ablation (n = 20) with subaortic ventriculotomy (n = 17) in patients with hypertrophic obstructive cardiomyopathy by echocardiography. METHODS: The pre- and post-operation (1 week post procedure) gradients of left ventricular out flow (LVOTGs) were measured by echocardiography. RESULTS: Post operation LVOTGs were significantly reduced than that of pre-operation in patients treated with either percutaneous transluminal septal myocardial ablation [(36 +/- 20) vs. (99 +/- 19) mm Hg (1 mm Hg = 0.133 kPa), P < 0.05], or subaortic ventriculotomy [(28 +/- 17) vs. (117 +/- 32) mm Hg]. Post operation LVOTGs were also similar with the two procedures [(36 +/- 20) vs. (28 +/- 17) mm Hg, P > 0.05]. CONCLUSION: Percutaneous transluminal septal myocardial ablation is as effective as subaortic ventriculotomy in treating patients with hypertrophic obstructive cardiomyopathy.

[Effects of salt stress on the root growth and leaf water use efficiency of cucumber seedlings].

By the method of water culture, and with the seedlings of two cucumber cultivars having different tolerance to salt stress as test materials, this paper studied their root growth, vitality, and membrane permeability, as well as their leaf growth, transpiration rate (Tr) , relative water content (RWC) , and water use efficiency (WUE) under effects of different NaCl levels. The results showed that under salt stress, the absorption surface area of root was declined, whereas its membrane permeability was increased significantly. The leaf number per cucumber seedling was decreased, compared with that of the control. Both Tr and RWC were markedly decreased after 2 days exposure to salt stress. During the period of salt treatment, root vitality and leaf WUE were increased initially, but decreased then. For the salt-susceptible cultivar Jinchun No. 2, the decrement of its root vitality was 18.01% , 12. 17% and 10. 95 % higher than that of the salt tolerant cultivar Changchun Mici on the 9th day exposure to 50, 75 and 100 mmol x L(-1) NaCl, and the decrement of its leaf WUE was 2. 74% , 5. 27% and 0. 23 % higher than that of Changchun Mici on the 8th day exposure to 50, 75 and 100 mmol x L(-1) NaCl, respectively. Under short-term salt stress, the absorption capacity of root was raised to compensate its reduced absorption surface area, while the decrease of Tr and the increase of WUE could reduce the leaf water loss, which alleviated the imbalance between water demand and need and improved the salt-tolerance of cucumber seedlings. After 5 days exposure to salt stress, both root vitality and leaf WUE decreased, which showed more severe water imbalance. It could be concluded that the decrease of root absorption capacity was one of the main reasons of water imbalance, and the decrease of leaf WUE was one of the responses to water imbalance. Both the absorption capacity of root and the WUE of leaf were closely related to the salt-tolerance of cucumber cultivar.

[Pedicle screws in the posterior reconstructive operation of the lower cervical vertebra].

OBJECTIVE: To explore the efficiency of the cervical pedicle screws in the posterior reconstructive operation of the lower cervical diseases. METHODS: From November 2001 to July 2003, 31 patients suffering from the lower cervical diseases were treated with the posterior pedicle screws fixation. We put the screws in the inferior articular process of the upper centrum, the lateral of the intervertebral facet center (3 - 5 mm the medial edge of lateral mass). The angles of toe-in sagittal plane varied from 30 degrees to 45 degrees, and the screws were from 18 to 26 mm (22.3 mm) in average. RESULTS: There were 193 pedicle screws in this group, and no one had spinal cord injury or vertebral artery injury. The improvement rate was 84.5%. Follow-up was from 9 to 28 months (17 months in average). The screws were fixed properly without being loose and broken, and the bone graft fusions within 9 to 12 months in the X-ray and CT scan examination were satisfactory. The Japanese orthopaedics association score (JOA) was 9.7 preoperatively, but it was raised to 16.3 postoperatively. CONCLUSION: The posterior reconstructive operation with cervical pedicle screws is a good choice for patients with lower cervical diseases.

A rice glutamate receptor-like gene is critical for the division and survival of individual cells in the root apical meristem.

Glu receptors are known to function as Glu-activated ion channels that mediate mostly excitatory neurotransmission in animals. Glu receptor-like genes have also been reported in higher plants, although their function is largely unknown. We have identified a rice (Oryza sativa) Glu receptor-like gene, designated GLR3.1, in which mutation by T-DNA insertion caused a short-root mutant phenotype. Histology and DNA synthesis analyses revealed that the mutant root meristematic activity is distorted and is accompanied by enhanced programmed cell death. Our results supply genetic evidence that a plant Glu receptor-like gene, rice GLR3.1, is essential for the maintenance of cell division and individual cell survival in the root apical meristem at the early seedling stage.

Sensing of lysophospholipids by TRPC5 calcium channel.

TRPC calcium channels are emerging as a ubiquitous feature of vertebrate cells, but understanding of them is hampered by limited knowledge of the mechanisms of activation and identity of endogenous regulators. We have revealed that one of the TRPC channels, TRPC5, is strongly activated by common endogenous lysophospholipids including lysophosphatidylcholine (LPC) but, by contrast, not arachidonic acid. Although TRPC5 was stimulated by agonists at G-protein-coupled receptors, TRPC5 activation by LPC occurred downstream and independently of G-protein signaling. The effect was not due to the generation of reactive oxygen species or because of a detergent effect of LPC. LPC activated TRPC5 when applied to excised membrane patches and thus has a relatively direct action on the channel structure, either because of a phospholipid binding site on the channel or because of sensitivity of the channel to perturbation of the bilayer by certain lipids. Activation showed dependence on side-chain length and the chemical head-group. The data revealed a previously unrecognized lysophospholipid-sensing capability of TRPC5 that confers the property of a lipid ionotropic receptor.

An association study between the transthyretin (TTR) gene and mental retardation.

It is known that in the pathogenesis of mental retardation (MR), both genetic and environmental factors (particularly iodine deficiency) appear to play a critical role. Transthyretin (TTR) transports between 20% and 30% of serum thyroxine in normal individuals and it is the main T(4)-binding protein in CSF. Variability in the TTR gene may influence risk for iodine-deficiency-based MR. The SNPs we selected from dbSNP were detected and identified using ARMS-PCR and sequencing methods, and we identified five novel sequence variants. Singular-locus association analysis indicated no association between the TTR gene and MR. In haplotype analysis, however, we found a haplotype CGTG+ (rs723744/G+6649C/T+6690C/rs2276382/del9) showed a weak positive association with MR (chi(2) = 6.699, p = 0.035). Finally, we concluded that the weak positive result is more likely to be due to sampling error and the small size of this haplotype resulting from its relative low frequency. Our negative results provide no evidence that variants of TTR gene influence susceptibility to MR in the iodine-deficient areas of China and suggest that there may be a compensatory mechanism(s) in humans and mice, which work(s) to compensate the effect of mutation in the TTR gene on MR.

Effects of surface modification of poly (3-hydroxybutyrate-co-3-hydroxyhexanoate) (PHBHHx) on physicochemical properties and on interactions with MC3T3-E1 cells.

As a new member of the polyhydroxyalkanoate (PHA) family, poly (3-hydroxybutyrate-co-3-hydroxyhexanoate) (PHBHHx) has better mechanical and processible properties than poly (3-hydroxybutyrate) (PHB). Still, it is difficult to introduce functional groups to the polyester carbon chain of PHBHHx, which restricts the modification of PHBHHx for a wide range. In this study, a procedure for the modification of the surface of PHBHHx films under strongly alkaline conditions was described. Through this kind of modification, carboxyl and hydroxyl groups were introduced to the surface and the total surface free energy was increased, which was mainly due to the increased polar components. Meanwhile, this process makes the surface rougher, resulting in larger total surface areas. After mineralization in simulated body fluids (SBFs), the apatite nucleation and growth on the surface-hydrolyzed PHBHHx films were significantly faster than on the unmodified PHBHHx films. This phenomenon should have a close relationship with the increased carboxyl and hydroxyl groups. The physicochemical properties also influenced the cell response to PHBHHx films. Compared to unmodified PHBHHx, fibronectin adsorption, and MC3T3-E1 cell attachment and proliferation were significantly greater on surface-hydrolyzed PHBHHx, which may be due to the increased surface free energy and rougher surface. Therefore, surface hydrolysis makes PHBHHx more suitable for osteoblast cell response and for application in bone-tissue engineering.

Enhanced sensitivity to chemotherapy in esophageal cancer through inhibition of NF-kappaB.

BACKGROUND: Apoptosis induced by chemotherapeutic drugs is blocked by activation of nuclear factor-kappaB (NF-kappaB) in some cancer cell lines. Therefore, inhibition of NF-kappaB by adenoviral delivery of an IkappaBalpha super-repressor (Ad.IkappaBalpha-SR) should make esophageal cancer cells more sensitive to 5- fluorouracil (5-FU) and taxotere chemotherapy. MATERIALS AND METHODS: SEG1 and BIC1 human esophageal cancer cells were studied. Chemotherapy-induced NF-kappaB activation was assessed by luciferase reporter assay and by electrophoretic mobility shift assay (EMSA). Cell growth inhibition was assessed by cell proliferation assay. Apoptosis was determined by flow cytometry and caspase 3/7 assay. RESULTS: 5-FU and taxotere significantly induced NF-kappaB activation as measured by luciferase reporter assay (P < 0.005) and EMSA. In both cell lines, Ad.IkappaBalpha-SR enhanced apoptosis by caspase 3/7 assay as compared to chemotherapy alone. In SEG1 cells, pre-treatment with Ad.IkappaBalpha-SR followed by 5-FU (0.25 mm) or taxotere (0.01 ug/ml), led to an increased apoptosis of 183% and 55%, respectively (P < 0.005). Similarly, apoptosis in BIC1 cells was significantly increased by pre-treatment with Ad.IkappaBalpha followed by 5-FU (0.1 mm) or taxotere (0.01 ug/ml) (P < 0.005). Growth inhibition in both cell lines was also significantly increased by pre-treatment with Ad.IkappaBalpha followed by 5-FU (0.01 mm) or taxotere (0.001 ug/ml) (P < 0.0005). CONCLUSION: Treatment with 5-FU or taxotere induced NF-kappaB activation in human esophageal cancer cells. Inhibition of NF-kappaB by Ad.IkappaBalpha-SR increased their sensitivity to 5-FU and taxotere chemotherapy. Therefore, these results may provide the basis for novel clinical approaches using cytoxic therapy with NF-kappaB inhibition to improve outcome in patients with esophageal cancer.

Tethered bilayer lipid membranes based on monolayers of thiolipids mixed with a complementary dilution molecule. 1. Incorporation of channel peptides.

Tethered bilayer lipid membranes (tBLMs) are described based on the self-assembly of a monolayer on template stripped gold of an archea analogue thiolipid, 2,3-di-o-phytanyl-sn-glycerol-1-tetraethylene glycol-d,l-alpha-lipoic acid ester lipid (DPTL), and a newly designed dilution molecule, tetraethylene glycol-d,l-alpha-lipoic acid ester (TEGL). The tBLM is completed by fusion of liposomes made from a mixture of diphytanoylphosphatidyl choline (DPhyPC), cholesterol, and 1,2-diphytanoyl-sn-glycero-3-phosphate (DPhyPG) in a molar ratio of 6:3:1. Melittin and gramicidin are incorporated into these tBLMs as shown by surface plasmon resonance (SPR) and electrochemical impedance spectroscopy (EIS) studies. Ionic conductivity at 0 V vs Ag|AgCl, 3 M KCl, measured by EIS measurements are comparable to the results obtained by other research groups. Admittance plots as a function of potential are discussed on a qualitative basis in terms of the kinetics of ion transport through the channels.
