Amenorrhea-galactorrhea syndrome with craniopharyngioma.

Two cases of craniopharyngioma presenting with amenorrhea-galactorrhea syndrome due to hyperprolactinemia are reported. After operation and irradiation, the tumor reduced markedly in size. Coincidental decrease in plasma prolactin level and restoration of menstruation seem to support the view that the hypothalamic prolactin inhibiting factor (PIF) had played an important role in hyperprolactinemia in these two patients.

Selective toxicity of 5-S-cysteinyldopa, a melanin precursor, to tumor cells in vitro and in vivo.

The effect of 5-S-cysteinyl-L-3,4-dihydroxyphenylalanine (cys-dopa), an intermediate in the pathway from L-3,4-dihydroxyphenylalanine (L-dopa) to pheomelanin, on the growth of eight human tumor cell lines in culture was compared to that of L-dopa. The tumor cell lines tested comprise two neuroblastomas (NB-1 and YT-nu), two amelanotic melanomas (HMV and SEKI), a gastric carcinoma (MKN-28), and three squamous cell carcinomas (HeLa-S3, KB, and a salivary gland carcinoma). Cys-dopa at a concentration of 1 mM inhibited growth of NB-1 (66%), YT-nu (67%), HMV (44%), SEKI (60%), MKN-28 (47%), HeLa-S3 (24%), KB (64%), and salivary gland carcinoma (33%), while L-dopa exhibited similar or even lower degree of inhibition at a concentration of 6 mM. On the other hand, both catechols had little effect on the growth of two fibroblasts derived originally from normal tissues (mouse fibroblast L929 and Chinese hamster fibroblast Don-6). Cys-dopa and L-dopa inhibited DNA and protein synthesis in YT-nu cells, but RNA synthesis was less affected. Treatment with cys-dopa at a dose of 1000 mg/kg i.p. for 7 days prolonged by 50% the life span of mice inoculated with L1210 leukemia. Normal mice given cys-dopa at a dose of 1000 mg/kg for 12 days showed no signs of toxicity. These results suggest the potential of cys-dopa as an antitumor agent.

In vitro secretion of follicle-stimulating hormone by pituitary chromophobe adenomas.

Of 20 female and 14 male patients with chromophobe adenomas, 4 male patients, aged 38 to 47 years, and one female patient, aged 54, showed above normal basal plasma levels of follicle-stimulating hormone (FSH). An in vitro tissue culture study that was performed using chromophobe cells obtained from 3 of the 4 male patients and the 1 female patient showed moderate FSH secretion into the culture media. The secretion of FSH by pituitary chromophobe adenomas might be more frequent than hitherto suspected.

The effect of heparin-free dialysis on abnormal lipid metabolism in patients on regular dialysis treatment.

To elucidate the contributory role of heparin to abnormal lipid metabolism in patients on regular haemodialysis (RDT), six week haemodialysis without heparin was performed using gabexate mesilate in combination with low dose aspirin. Total cholesterol and beta-lipoprotein were significantly increased at the fourth week after the start of the study. No significant changes were observed in high density lipoprotein cholesterol, phospholipid, triglyceride, lipoprotein phenotype and post heparin lipolytic activity. Routine dose heparin in RDT might have a favourable effect on abnormal lipid metabolism by inducing post heparin lipolytic activity in circulating blood, not by reducing the lipid removing activity in peripheral tissues.

Deterioration of renal function in non-dialytic uraemics caused by 1 alpha (OH)D3 which was not attributable to hypercalcaemia.

Ten patients with renal failure not on dialysis were subjected to our study to evaluate the effect of 1 alpha (OH)D3 on calcium metabolism and its influence on renal function. The results suggest the possibility that 1 alpha (OH)D3 might be effective in bone disease due to secondary hyperparathyroidism. On the other hand, 1 alpha (OH)D3 depressed creatinine clearance without any increase of Ca, Ca x P product or PRA. 1 alpha (OH)D3 is to be prescribed to patients not on dialysis only with careful attention to the dosage and the duration, and frequent observation of renal function is necessary.

Effect of an intravenous infusion of ethanol on serum enzymes and lipids in patients with alcoholic liver disease.

Administration of ethanol to healthy subjects as well as those with alcoholic fatty livers has been noted to cause mild elevation of serum enzyme activities and alcoholic hyperlipemia. To see whether this effect is also manifested in alcoholics with advanced liver damage, the serum enzyme activities and lipid content after administration of i.v. ethanol (1.2 g/kg of body wt) over 90 min were compared in 5 alcoholics with hepatic fibrosis and/or alcoholic fatty liver and in 9 patients with alcoholic hepatitis with or without cirrhosis. Disappearance rate of ethanol from the serum was nearly the same in both groups. Serum activities of asparate aminotransferase, mitochondrial isoenzyme of asparate aminotransferase, and ornithine carbamyl transferase, measured at 9 and 12 hr after termination of the i.v. ethanol, were significantly elevated in alcoholics with alcoholic hepatitis (P less than 0.05). They were not elevated in alcoholics with fatty liver. By contrast, hyperlipemic responses, measured as the serum content of triacylglycerol 3 hr after ethanol were significantly greater in alcoholics with fatty liver than in alcoholics with alcoholic hepatitis (P less than 0.05). The observed difference in responses of serum enzyme activities and lipid content after ethanol represents an enhanced susceptibility of patients with alcoholic hepatitis and cirrhosis towards alcohol-induced injury.

[Intracranial malignant lymphoma (author's transl)].

We experienced 19 histologically verified intracranial malignant lymphomas during the period of 15 years from April 1, 1965 to August 31, 1979. From the analysis of cases of our own and from the literature, the following are considered to be the clinical characteristics of intracranial malignant lymphomas. 1) Males were more affected than females. 2) The highest frequency was observed in the 5th to 7th decades. 3) Increased intracranial pressure, motor disturbance, and psychic or consciouness disturbance were 3 cardinal symptoms. 4) Tumors were localized frequently in the paraventricular region. 5) Angiography revealed hypovascular lesion in many cases, while CT scan showed homogenous, slightly high density lesion which was remarkably enhanced by contrast material. 6) Long survival was rare irrespective of all kinds of treatments. 7) Immunological classification was thought to be useful in prospecting survival time.

Chondroitinase-resistant sulfated glycosaminoglycans synthesized by cartilages of chick embryos and of newborn chickens and rats.

Biosynthesis of chondroitinase-resistant glycosaminoglycans as minor components was studied in the cartilages of chick embryos and of newborn chickens and rats. Sternal and knee cartilages were labeled in vitro with 35SO42-, and then 35S-labeled glycosaminoglycans were analyzed. In rats up to 2 weeks old, only one glycosaminoglycan could be detected as heparan sulfate. In the chick embryos and the newborn chickens, however, keratan sulfate as well as heparan sulfate could be detected. As chondroitinase-sensitive glycosaminoglycans, large amounts of both chrondroitin 4- and 6-sulfates were synthesized in the chick cartilage, but the synthesis of chondroitin 6-sulfate could scarcely be seen in the rat cartilage. The results seem to indicate that the biosynthesis of keratan sulfate has some relation to that of chondroitin 6-sulfate.

Neuroradiological aspects of suprasella germinoma.

The plain skull roentgenography, angiography, air encephalography or positive contrast medium ventriculography and computed tomography (CT) of twelve cases of suprasella germinoma were reviewed. In over half of the cases, plain skull roentgenography and angiography showed some abnormalities suggesting a suprasella mass. CT was found to be the most appropriate for routine examination. And air encephalography or positive contrast medium ventriculography offered decisive information.
