Experimental diabetes and platelet aggregation in the rat.

Alloxan-induced diabetes and platelet aggregation were studied in male Sprague-Dawley rats. Platelets from diabetic rats receiving insulin (one unit protamine-zinc insulin/100 g BW) were consistently more responsive to aggregating agents (ADP, thrombin) than platelets from control rats (with or without insulin administration). Also, serotonin release from platelets was consistently greater in diabetic compared to control rats following thrombin-stimulated platelet aggregation.

Health and social characteristics of long-term psychotropic drug takers.

Prolonged use of drugs for the treatment of specific chronic disorders such as hypertension. diabetes and asthma can be explained largely in clinical terms. The use of psychotropic drugs, on the other hand appears to be in response to such a broad range of indications that prolonged use requires investigation from a sociomedical perspective. In this paper we consider the contributions of illness behavior and social network theories to an understanding of long-term use of psychotropic drugs. The present sample emerged from a cohort of general practice patients a new episode of psychotropic drug treatment. Twenty-six patients from a cohort of 124 continued to receive prescriptions for at least 6 months and 19 were interviewed for this pilot study.

Self-assessment of health: an exploration of the effects of physical and psychological symptoms.

The clinician has no objective criteria of good health; he is likely to identify its presence only in the absence of disease and dysfunction. Yet the layman is able to give a global assessment of his own health as good, average or poor. The present paper explores the correlates of self-assessment of health in a large community sample and, by the use of two linear models, describes the effects of age, sex, reported symptoms, and long-standing illness on self-rated health status. It is suggested that global ratings are influenced by age and sex and by the experience of both physical and psychological symptoms, and that there are no interactive effects between symptoms and the demographic variables.

Intra-arterial chemotherapy of malignant fibrous histiocytoma of the pelvis.

In 3 cases of malignant fibrous histiocytoma of the bony pelvis, intra-arterial chemotherapy was followed by surgical resection when possible in an attempt to control local disease without unacceptable deformity and treat undetected systemic metastases. Cis-diamminedichloroplatinum (CDDP) and a combination of intra-arterial doxorubicin, 5-(dimethyltriazeno)imidazole-4-carboxamide, and intravenous cyclophosphamide (CYADIC) were used. In 2 patients who underwent surgical resection, no viable tumor cells were found. The third patient did not respond to these or other forms of treatment. The patients treated with resection have shown no evidence of local recurrence at 24 and 9 months, respectively.

Radiographic and angiographic changes in osteosarcoma after intraarterial chemotherapy.

Transcatheter arterial infusion has improved the preoperative control of osteosarcoma. The conventional radiographs and angiograms of osteosarcoma were evaluated in 53 patients before and after treatment. In the responders, two types of calcifications representing healing are observed: peripheral and central. The calcification is readily differentiated from that of tumor new bone. Angiography revealed a decrease in tumor vascularity and stain in responding tumors. Excellent angiographic and pathologic correlation of these changes is presented.

Intraarterial cis-platinum for patients with inoperable skeletal tumors.

Twenty-three patients with inoperable skeletal tumors were treated with intraarterial cis-platinum prior to attempted surgery. The antitumor effect of intraarterial cis-platinum was monitored clinically by radiologic imaging techniques, and whenever possible, evaluated histopathologically by examinatin of surgical or biopsy tumor specimens. Objective responses were noted in 12 patients (52%) and included 2 complete, 7 partial and 3 less-than-partial remissions lasting from 14 to 70 weeks. Limb-saving surgery or hemipelvectomy became subsequently feasible in four and one patients respectively. Preoperative intraarterial cis-platinum is a safe procedure which might be used effectively in combination with other, more conventioal postoperative adjuvant chemotherapy against skeletal tumors.

Factors affecting the consumption of psychotropic drugs.

This paper presents some results from a survey conducted in West London in 1977. The 2-week prevalence of psychotropic drug consumption was found to be 10.9% and almost identical figure to that obtained in an earlier survey conducted 8 years previously. The reasons for the absence of the expected rise in prevalence of consumption are discussed. Consumption was found to be strongly associated with various measures of health, and there was a marked sex difference in drug consumption (females greater than males) which was independent of health.

Tissue distribution, uptake, and requirement for alpha-tocopherol of rainbow trout (Salmo gairdneri) fed diets with a minimal content of unsaturated fatty acids.

The metabolism of and requirements for alpha-tocopherol in rainbow trout fed diets containing 1% linolenic acid as sole source of unsaturated fat and graded levels of tocopherol (0.06-10 mg/100 g) were examined. Fish grew 5-fold over a 16-week period. In liver, tocopherol was concentrated in mitochondria with little in cytosol. Orally administered [3H]-tocopherol was rapidly taken up by plasma and liver but uptake into erythrocytes and white muscle was much slower; in most tissues radioactivity reached a plateau after about 3 days but in red muscle radioactivity increased over a 10-day period. Activities of enzymes that prevent free radical initiated tissue damage did not change in tocopherol deficiency. Tocopherol-deficient trout had no gross or subcellular pathologies even though liver and muscle were severely depleted of the vitamin. Ascorbic acid-stimulated lipid peroxidation in liver organelles indicated a tocopherol requirement of 2-3 mg/100 g diet; the molar ratios of polyunsaturated fatty acids to tocopherol in livers of trout fed diets lacking or supplemented with tocopherol (100 mg/100 g) were 980 and 170, respectively.
